Abstract

The invention relates to a colorimetric method for detecting bacterial or fungal pathogens by detecting peptidoglycan or (1-3)-β-D-glucan in a sample.

Claims (33)

1. A method for detecting bacteria or bacterial fragments in a platelet containing sample, comprising the steps of:

(a) incubating the sample with hemolymph from a Lepidopteran insect, a phenoloxidase substrate that generates a reaction product, and 3-methyl-2-benzothiazolinone hydrazone or other hydrazone derivative; and,

(b) detecting the formation of color, wherein formation of color indicates the presence of bacteria or bacterial fragments in the sample and the absence of color indicates the absence of bacteria or bacterial fragments in the sample.

2. The method of claim 1 , wherein the sample is a platelet unit.

3. The method of claim 1 , wherein the sample is processed by centrifugation and bacteria present in the sample are pelleted during centrifugation.

4. The method of claim 1 , wherein the sample is further incubated with a peptidoglycan binding protein.

5. The method of claim 1 , wherein the Lepidopteran insect is hornworm.

6. The method of claim 1 , wherein the phenoloxidase substrate is L-3,4-dihydroxyphenylalanine, dopamine, 3,4-dihydroxyphenyl propionic acid, 3,4-dihydroxyphenyl acetic acid, a dihydroxyphenol, a monophenol or catechol.

7. The method of claim 1 , further comprising the step of exposing the sample to an extraction solution at an elevated temperature, prior to incubating the sample with the hemolymph.

8. The method of claim 7 , further comprising the step of exposing the sample to a neutralization buffer prior to incubating the sample with the hemolymph.

9. The method of claim 8 , wherein the neutralization buffer comprises 3-methyl-2-benzothiazolinone hydrazone or other hydrazone derivative.

10. The method of claim 7 , wherein the extraction solution is an alkaline extraction solution.

11. The method of claim 1 , wherein the hemolymph is lyophilized.

12. The method of claim 1 , further comprising the step of exposing the sample to a stop reagent.

13. The method of claim 12 , wherein the stop reagent is an acid reagent, a phenoloxidase inhibitor, or a strong anionic detergent alone or in combination with a phenoloxidase inhibitor.

14. The method of claim 1 , wherein the bacteria in the sample are detected at a level as low as 100 CFU/mL.

15. The method of claim 1 , wherein the bacteria in the sample are detected at a level between 600-1,000 CFU/mL.

16. The method of claim 1 , wherein the bacteria in the sample are detected at a level between 1,000-2,500 CFU/mL.

17. The method of claim 1 , wherein the bacteria in the sample are detected at a level between 5,000-10,000 CFU/mL.

18. A method for detecting bacteria or bacterial fragments in a platelet containing sample, comprising the steps of:

(a) extracting the sample in an extraction solution,

(b) incubating the sample with hemolymph from a Lepidopteran insect, dopamine and 3-methyl-2-benzothiazolinone hydrazone or other hydrazone derivative dissolved in neutralization buffer, and

(c) detecting the formation of color, wherein the formation of color indicates the presence of bacteria or bacterial fragments in the sample and the absence of color indicates the absence of bacteria or bacterial fragments in the sample.

19. The method of claim 18 , further comprising the step of exposing the sample to a neutralization buffer prior to incubating the sample with hemolymph, dopamine and 3-methyl-2-benzothiazolinone or other hydrazone derivative.

20. The method of claim 19 , wherein the hemolymph dopamine and 3-methyl-2-benzothiazolinone hydrazone or other hydrazone derivative are lyophilized.

21. The method of claim 18 , wherein the hemolymph is tobacco hornworm hemolymph.

22. The method of claim 18 , wherein the bacteria in the sample are detected at a level as low as 100 CFU/mL.

23. The method of claim 18 , wherein the bacteria in the sample are detected at a level between 600-1,000 CFU/mL.

24. The method of claim 18 , wherein the bacteria in the sample are detected at a level between 1,000-2,500 CFU/mL.

25. The method of claim 18 , wherein the bacteria in the sample are detected at a level between 5,000-10,000 CFU/mL.

26. The method of claim 18 , wherein the sample is a platelet unit.

27. The method of claim 18 wherein the extraction solution is an alkaline extraction solution.

28. The method of claim 18 , wherein after the incubating step (b) and before the detecting step (c), the reaction is stopped with phenyl-thiourea.
