Abstract

The present invention provides an improved, expression system for the regulated expression of an encoded protein or nucleic acid therapeutic molecule in the cells of a subject, for use in the treatment of disease. In particular, the present invention provides an improved, regulated gene expression system, and pharmaceutical compositions and uses thereof for treatment of disease.

Claims (117)

1 . A regulated expression system for use in the treatment of disease comprising at least a vector, wherein said vector comprises:

a first expression cassette comprising: i) a cloning site for insertion of a first nucleic acid sequence encoding a therapeutic molecule (TM) having a therapeutic activity, and ii) a first promoter and a first poly(A) site operably linked to said first nucleic acid sequence when inserted at said first cloning site,

wherein said TM is expressed in cells of a subject, and said TM expression or activity is orientation-dependent and is regulated in the presence of a regulator molecule (RM).

The regulated expression system of claim 1 , wherein said vector further comprises a second expression cassette comprising: i) a second cloning site for insertion of a second nucleic acid sequence encoding said regulator molecule (RM), and ii) a second promoter and a second poly(A) site operably linked to said second nucleic acid sequence when inserted at said second cloning site,

wherein said RM is expressed in said cells.

3 . The regulated expression system of claim 1 further comprising a first nucleic acid sequence encoding said therapeutic molecule (TM) inserted at said first cloning site.

4 . The regulated expression system of claim 3 further comprising a first nucleic acid sequence encoding said therapeutic molecule (TM) inserted at said first cloning site, and a second nucleic acid sequence encoding a regulator molecule (RM) inserted at said second cloning site.

5 . A regulated expression system for use in the treatment of disease comprising at least a vector, said vector comprising:

A. a first expression cassette comprising: i) first nucleic acid sequence encoding a therapeutic molecule (TM) having a therapeutic activity, and ii) a first promoter and a first poly(A) site operably linked to said first nucleic acid sequence,

wherein said TM is expressed in cells of a subject, and said TM expression or activity is dose-dependent and regulated in the presence of a regulator molecule (RM); and

B. a second expression cassette comprising: i) a second nucleic acid sequence encoding said regulator molecule (RM), and ii) a second promoter and a second poly(A) site operably linked to said second nucleic acid sequence.

6 . A regulated expression system for use in the treatment of disease comprising at least a vector, said vector comprising:

A. a first expression cassette comprising: i) first nucleic acid sequence encoding a therapeutic molecule (TM) having a therapeutic activty, and ii) a first promoter and a first poly(A) site operably linked to said first nucleic acid sequence,

wherein said TM is expressed in cells of a subject, and said TM expression or activity is dose-dependent and regulated in the presence of a regulator molecule (RM); and

B. a second expression cassette comprising: i) a second nucleic acid sequence encoding said regulator molecule (RM), and ii) a second promoter and a second poly(A) site operably linked to said second nucleic acid sequence,

wherein said RM is expressed in said cells and activated in the presence of an activator molecule (AM), thereby regulating said TM expression or activity.

7 . A regulated expression system for use in the treatment of disease comprising at least a vector, said vector comprising:

A. a first expression cassette comprising: i) first nucleic acid sequence encoding a therapeutic molecule (TM) having a therapeutic activity, and ii) a first promoter and a first poly(A) site operably linked to said first nucleic acid sequence,

wherein said TM is expressed in cells of a subject, and said TM expression or activity is dose-dependent and induced in the presence of an activated regulator molecule (RM); and

B. a second expression cassette comprising: i) a second nucleic acid sequence encoding said regulator molecule (RM), and ii) a second promoter and a second poly(A) site operably linked to said second nucleic acid sequence,

wherein said RM is expressed in said cells and is activated in the presence of an activator molecule (AM), thereby inducing said TM expression or activity.

8 . The regulated expression system of claim 7 , wherein said regulated expression system further comprises an activator molecule (AM) that is separately administered from said vector.

9 . The regulated expression system of claim 5 , wherein said TM expression or activity is induced in the presence of said regulator molecule (RM).

10 . The regulated expression system of claim 5 , wherein said TM expression or activity is repressed in the presence of said regulator molecule (RM).

11 . The regulated expression system of claim 5 , wherein said TM expression or activity is increased in the presence of said regulator molecule (RM).

12 . The regulated expression system of claim 5 , wherein said regulator molecule (RM) is activated and thereby regulates said TM expression or activity.

13 . The regulated expression system of claim 12 , wherein said regulator molecule (RM) is activated in the presence of an activator molecule (AM).

14 . The regulated expression system of claim 12 , wherein said regulator molecule (RM) is activated by a conformational change in said RM.

15 . The regulated expression system of claim 12 , wherein said regulator molecule (RM) is activated by a modification of said RM.

16 . The regulated expression system of claim 13 further comprising an activator molecule (AM).

17 . The regulated expression system of claim 16 , wherein said activator molecule (AM) is a naturally-occurring molecule or variant thereof.

18 . The regulated expression system of claim 16 , wherein said activator molecule (AM) is a modified molecule.

19 . The regulated expression system of claim 16 , wherein said activator molecule (AM) is a synthetic molecule.

20 . The regulated expression system of claim 16 , wherein said activator molecule (AM) is a chemical compound.

21 . The regulated expression system of claim 20 , wherein said chemical compound is an antiprogestin.

22 . The regulated expression system of claim 21 , wherein said antiprogestin is mifepristone.

23 . The regulated expression system of claim 5 , wherein the expression of said regulator molecule (RM) is constitutive or transient.

24 . The regulated expression system of claim 23 , wherein the expression of said regulator molecule (RM) is constitutive.

25 . The regulated expression system of claim 5 , wherein the expression of said regulator molecule (RM) is regulated.

26 . The regulated expression system of claim 25 , wherein said second promoter is a regulated promoter.

27 . The regulated expression system of claim 5 , wherein said second promoter is a tissue-specific promoter.

28 . The regulated expression system of claim 27 , wherein said second promoter is a muscle-specific promoter.

29 . The regulated expression system of claim 28 , wherein said second promoter is an actin promoter.

30 . The regulated expression system of claim 9 , wherein said regulator molecule (RM) binds to said first promoter, thereby inducing said TM expression or activity.

31 . The regulated expression system of claim 9 , wherein said regulator molecule (RM) is activated and thereby binds to said first promoter and induces said TM expression or activity.

32 . The regulated expression system of claim 5 , wherein said regulator molecule (RM) comprises a transactivation domain.

33 . The regulated expression system of claim 32 , wherein said transactivation domain is a VP16 or p65 transactivation domain.

34 . The regulated expression system of claim 5 , wherein said regulator molecule (RM) comprises a ligand-binding domain (LBD).

35 . The regulated expression system of claim 34 , wherein said regulator molecule (RM) comprises a ligand-binding domain (LBD) and said activator molecule (AM) binds to said LBD, thereby activating said RM.

36 . The regulated expression system of claim 5 , wherein said regulator molecule comprises a DNA-binding domain (DBD).

37 . The regulated expression system of claim 36 , wherein said DNA-binding domain (DBD) comprises a GAL-4 DBD.

38 . The regulated expression system of claim 5 , wherein said second expression cassette further comprises a second functional sequence operably linked to said second nucleic acid sequence.

39 . The regulated expression system of claim 5 , wherein said second poly(A) site is a simian virus 40 (SV40) poly(A) site.

40 . The regulated expression system of claim 5 , wherein said regulator molecule (RM) is a naturally-occurring molecule or variant thereof.

41 . The regulated expression system of claim 5 , wherein said regulator molecule (RM) is a modified molecule.

42 . The regulated expression system of claim 41 , wherein said regulator molecule (RM) is a protein.

43 . The regulated expression system of claim 42 , wherein said protein is a humanized protein.

44 . The regulated expression system of claim 42 , wherein said protein is a human protein or variant thereof.

45 . The regulated expression system of claim 44 , wherein said protein is a transcriptional activator.

46 . The regulated expression system of claim 45 , wherein said transcriptional activator is a nuclear steroid receptor.

47 . The regulated expression system of claim 45 , wherein said transcriptional activator is a modified nuclear steroid receptor.

48 . The regulated expression system of claim 47 , wherein said modified nuclear steroid receptor is a modified progesterone receptor.

49 . The regulated expression system of claim 5 , wherein the expression of said therapeutic molecule (TM) is constitutive or transient.

50 . The regulated expression system of claim 5 , wherein said first promoter is a regulated promoter.

51 . The regulated expression system of claim 5 , wherein said first promoter is an inducible promoter.

52 . The regulated expression system of claim 5 , wherein said first promoter is a tissue-specific promoter.

The regulated expression system of claim 5 , wherein said first promoter is selected from a group of promoters consisting of a muscle creatine kinase (MCK) promoter, hypoxia responsive element (HRE) promoter, endothelial leukocyte adhesion molecule (ELAM) promoter, CMV/actin chimeric promoter, cyclin A promoter, and cdc6 promoter.

54 . The regulated expression system of claim 5 , wherein said first promoter is a Pol II promoter.

55 . The regulated expression system of claim 54 , wherein said Pol II promoter is the E1B promoter.

56 . The regulated expression system of claim 36 , wherein said first promoter comprises a binding site for said DNA-binding domain of said regulator molecule (RM).

57 . The regulated expression system of claim 56 , wherein said binding site comprises at least one GAL-4 binding site.

58 . The regulated expression system of claim 57 , wherein said binding site comprises multimers of said GAL-4 binding site.

59 . The regulated expression system of claim 58 , wherein said binding site comprises 3-18 GAL-4 binding sites.

60 . The regulated expression system of claim 5 , wherein said first poly(A) site is human Growth Hormone (hGH) poly(A) site.

61 . The regulated expression system of claim 5 , wherein said first expression cassette further comprises a first functional sequence operably linked to said first nucleic acid sequence.

62 . The regulated expression system of claim 61 , wherein said first functional sequence encodes a 5′ untranslated region and/or intron.

63 . The regulated expression system of claim 62 , wherein said first functional sequence encodes a 5′ untranslated region comprising UT12.

64 . The regulated expression system of claim 62 , wherein said functional sequence encodes an intron comprising IVS8.

65 . The regulated expression system of claim 5 , wherein said therapeutic molecule (TM) is a variant of a naturally-occurring molecule.

66 . The regulated expression system of claim 5 , wherein said therapeutic molecule (TM) is a modified molecule.

67 . The regulated expression system of claim 5 , wherein said therapeutic molecule (TM) is a synthetic or recombinant molecule.

68 . The regulated expression system of claim 5 , wherein said therapeutic molecule (TM) is a protein.

69 . The regulated expression system of claim 68 , wherein said therapeutic molecule (TM) is a human protein or variant thereof.

70 . The regulated expression system of claim 69 , wherein said therapeutic molecule (TM) is selected from a group of proteins consisting of GMCSF, Leukine®, IFN-α, and IFN-β, or a variant of said proteins.

71 . The regulated expression system of claim 70 , wherein said therapeutic molecule (TM) is GMCSF.

72 . The regulated expression system of claim 68 , wherein said therapeutic protein (TM) is a monoclonal antibody.

73 . The regulated expression system of claim 72 , wherein said monoclonal antibody is CAMPATH®.

74 . The regulated expression system of claim 3 , wherein said vector is a plasmid vector.

75 . The regulated expression system of claim 74 , wherein said plasmid vector is selected from a group consisting of pGT1, pGT2, pGT3, pGT4, pGT11, pGT12, pGT13, and pGT14.

76 . The regulated expression system of claim 5 , wherein said vector is a plasmid vector.

77 . The regulated expression system of claim 76 , wherein said plasmid vector is selected from a group consisting of pGT23, pGT24, pGT25, pGT26, pGT27, pGT28, pGT29, and pGT30.

78 . The regulated expression system of claim 3 , wherein said vector is a shuttle plasmid.

79 . The regulated expression system of claim 5 , wherein said vector is a shuttle plasmid.

80 . The regulated expression system of claim 79 , wherein said shuttle plasmid is an adeno-associated viral (MV) shuttle plasmid.

81 . The regulated expression system of claim 80 , wherein said AAV is AAV serotype 1 (AAV-1).

82 . The regulated expression system of claim 81 , wherein said AAV-1 shuttle plasmid is selected from a group of AAV-1 vectors consisting of pGT53, pGT54, pGT57, pGT58, pGT715, and pGT716.

83 . The regulated expression system of claim 13 , wherein said activator molecule (AM) is orally administered, thereby activating said regulator molecule (RM).

84 . The regulated expression system of claim 5 , wherein said vector is administered by injection.

85 . The regulated expression system of claim 5 , wherein said vector is administered by contacting said cells with said vector in vivo or ex vivo.

86 . The regulated expression system of claim 85 , wherein said contact is in vivo and by injection.

87 . The regulated expression system of claim 84 , wherein said injection is by intramuscular injection.

88 . The regulated expression system of claim 87 , wherein said injection is by intramuscular injection of skeletal muscle cells.

89 . The regulated expression system of claim 85 , wherein said contact is ex vivo and by electroporation.

90 . A vector for use in a regulated expression system, wherein said vector is pGT1, pGT2, pGT3, pGT4, pGT11, pGT12, pGT13, or pGT14.

91 . A vector for use in a regulated expression system, wherein said vector is pGT23, pGT 24, pGT25, or pGT26.

92 . A vector for use in a regulated expression system, wherein said vector is pGT27, pGT 28, pGT29, or pGT30.

93 . A vector for use in a regulated expression system, wherein said vector is pGT53, pGT54, pGT57, pGT58, pGT715, and pGT716.

94 . A vector for use in a regulated expression system, wherein said vector is pGT53.

95 . A method for regulating the expression of a therapeutic molecule (TM) in cells of a subject using the regulated expression system according to any one of claims 5 to 7 , said method comprising contacting cells of said subject with said expression system, such that said TM is expressed in said cells, and said TM expression or activity is regulated in the presence of said regulator molecule (RM) in said cells.

96 . A method for administering a therapeutic molecule (TM) to cells of a subject using the regulated expression system according to any one of claims 5 to 7 , said method comprising contacting cells of said subject with said expression system, such that said TM is expressed in said cells, and said TM expression or activity is regulated in the presence of said regulator molecule (RM) in said cells.

97 . A method for delivering a therapeutic molecule (TM) to cells of a subject using the regulated expression system according to any one of claims 5 to 7 , said method comprising contacting cells of said subject with said expression system, such that said TM is expressed or activity in said cells, and said TM expression is regulated in the presence of said regulator molecule (RM) in said cells.

98 . A method of expressing a therapeutic molecule (TM) in cells of a subject in vivo or ex vivo using the regulated expression system according to any one of claims 5 to 7 , said method comprising contacting cells of said subject with said expression system, such that said TM is expressed in said cells, and said TM expression or activity is regulated in the presence of said regulator molecule (RM) in said cells.

99 . A pharmaceutical composition comprising at least one expression system according to any one of claims 5 to 7 .

100 . A pharmaceutical composition comprising the vector of any one of claims 91 to 94 .

101 . A kit for delivering a therapeutic molecule (TM) to cells of a subject, wherein said kit comprises at least one regulated expression system according to any one of claims 1 to 7 .

102 . A kit for delivering a therapeutic molecule (TM) to cells of a subject, wherein said kit comprises at least one vector according to claim 90 .

103 . A kit for delivering a therapeutic molecule (TM) to cells of a subject, wherein said kit comprises at least one vector according to any one of claims 91 to 94 .
