Abstract

Pectins having a combination of unexpectedly high molecular weights and low degrees of methylation have been isolated from Aloe vera plants, are superior gel pectins for forming calcium cross-lined gels. Such pectins can be obtained by extracting homogenized Aloe Vera plants or portions thereof. These pectins can be used to prepare pharmaceutical compositions comprising pharmacological agents for application to animals and humans, so as to provide controlled release of the pharmacological agent.

Claims (35)

1. A process for obtaining a purified Aloe pectin comprising:

a) homogenizing the green rind of an aloe vera plant leaf to give a homogenized material;

b) separating cell wall fibers from the homogenized Aloe material; and

c) extracting the pectin from the separated cell wall fibers.

2. The process of claim 1 wherein the separation of cell wall fibers comprises acidifying the homogenized material, and extracting with ethanol to produce a solid precipitate comprising the cell wall fibers.

3. The process of claim 2 , wherein the solid precipitate is treated with water, and the water insoluble portion comprising the cell wall fibers is separated from the water, and the cell wall fiber are then extracted in step c.

4. The process of claim 1 wherein the extraction employs a chelating agent.

5. The process of claim 4 wherein the chelating agent is EDTA.

6. The process of claim 1 wherein the extraction occurs at a pH from about 7 to about 9.

7. The process of claim 1 wherein the extraction occurs at about room temperature.

8. The process of claim 1 wherein the purified Aloe pectin has an average molecular weight greater than about 1.0×10 6 Daltons.

9. The process of claim 8 wherein the purified Aloe pectin has a degree of methylation of less than about 10%.

10. A process for obtaining a purified aloe pectin comprising:

a) homogenizing the green rind of an aloe vera plant leaf to give a homogenized material;

b) separating cell wall fibers from the homogenized Aloe material; and

c) extracting the pectin from the separated cell wall fibers; wherein the purified aloe pectin has an average molecular weight of greater than about 1.0×10 6 Daltons and a degree of methylation of less than about 10%.

11. The process of claim 10 wherein the separation of cell wall fibers comprises acidifying the homogenized material, and extracting with ethanol to produce a solid precipitate comprising the cell wall fibers.

12. The process of claim 11 , wherein the solid precipitate is treated with water, and the water insoluble portion comprising the cell wall fibers is separated from the water, and the cell wall fibers are then extracted in step c).

13. The process of claim 12 wherein the extraction employs a chelating agent.

14. The process of claim 13 wherein the chelating agent is EDTA.

15. The process of claim 12 wherein the extraction occurs at a pH from about 7 to about 9.

16. The process of claim 12 wherein the extraction occurs at about room temperature.

17. A process for obtaining a purified aloe pectin comprising:

a) homogenizing the green rind of an aloe vera plant leaf to give a homogenized material;

b) separating cell wall fibers from the homogenized Aloe material; and

c) extracting the pectin from the separated cell wall fibers;

wherein the pectin comprises 3-OMe Rhamnose at about 0.1% to about 5% by mole and wherein the pectin is capable of calcium gel formation when present at a concentration of 0.2% (w/v) in an aqueous solution comprising 2 mM calcium chloride.

18. The process of claim 17 wherein the separation of cell wall fibers comprises acidifying the homogenized material, and extracting with ethanol to produce a solid precipitate comprising the cell wall fibers.

19. The process of claim 18 , wherein the solid precipitate is treated with water, and the water insoluble portion comprising the cell wall fibers is separated from the water, and the cell wall fiber are then extracted in step c).

20. The process of claim 17 wherein the extraction employs a chelating agent.

21. The process of claim 20 wherein the chelating agent is EDTA.

22. The process of claim 19 wherein the extraction occurs at a pH from about 7 to about 9.

23. The process of claim 17 wherein the extraction occurs at about room temperature.

24. The process of claim 17 wherein the purified Aloe pectin has an average molecular weight greater than about 1.0×10 6 Daltons.

25. The process of claim 17 wherein the purified Aloe pectin has a degree of methylation of less than about 10%.
