Abstract

The invention provides isolated nucleic acid and amino acid sequences of novel receptors, antibodies to such receptors, methods of detecting such nucleic acids and receptors, and methods of screening for modulators of the receptors.

Claims (27)

1 . An isolated nucleic acid encoding a polypeptide that:

(i) comprises 80% or greater amino acid sequence identity to the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ NO:14; or

(ii) comprises at least 100 contiguous amino acids of an amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14; or

(iii) comprises at least 90% identity to the amino acid sequence of SEQ ID NO:20, SEQ ID NO:22, or SEQ ID NO:24.

2 . The isolated nucleic acid of claim 1 , wherein the polypeptide comprises 90% or greater amino acid sequence identity to the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14.

3 . The isolated nucleic acid of claim 1 , wherein the nucleic acid encodes a polypeptide comprising the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10; SEQ ID NO:12, SEQ ID NO:14, SEQ ID NO:20, SEQ ID NO:22, or SEQ ID NO:24.

4 . The isolated nucleic acid of claim 1 , wherein the nucleic acid comprises the nucleotide sequence of SEQ ID NO:1, SEQ ID NO:5, SEQ ID NO:7, SEQ ID NO:9, SEQ ID NO:11, SEQ ID NO:13, SEQ ID NO:19, SEQ ID NO:21, or SEQ ID NO:23.

5 . The isolated nucleic acid of claim 1 , wherein the nucleic acid is amplified by primers that specifically hybridize under stringent hybridization conditions to a nucleic acid comprising the nucleotide sequence of SEQ ID NO:1, SEQ ID NO:5, SEQ ID NO:7, SEQ ID NO:9, SEQ ID NO:1, or SEQ ID NO:13.

6 . The isolated nucleic acid of claim 1 , wherein the nucleic acid specifically hybridizes under stringent hybridization conditions to a nucleic acid comprising the nucleotide sequence of SEQ ID NO:1, SEQ ID NO:5, SEQ ID NO:7, SEQ ID NO:9, SEQ ID NO:1, or SEQ ID NO:13.

7 . An isolated polypeptide comprising:

(i) 80% or greater amino acid sequence identity to the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14; or

(ii) at least 100 contiguous amino acids of the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14; or

(iii) at least 90% identity to an amino acid sequence of SEQ ID NO:20, SEQ ID NO:22, or SEQ ID NO:24.

8 . The isolated polypeptide of claim 7 , wherein the polypeptide comprises 90% or greater identity to the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14.

9 . The isolated polypeptide of claim 7 , wherein the polypeptide comprises the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, SEQ ID NO:14, SEQ ID NO:20, SEQ ID NO:22, or SEQ ID NO:24.

10 . An antibody that selectively binds to the polypeptide of claim 7 .

11 . An expression vector comprising the nucleic acid of claim 1 .

12 . A host cell transfected with the vector of claim 11 .

13 . A method for identifying a compound that modulates signal transduction, the method comprising:

(i) contacting the compound with a polypeptide of claim 7 .

14 . The method of claim 13 , wherein the polypeptide comprises 90% or greater amino acid sequence identity to SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14.

15 . The method of claim 13 , wherein the functional effect is determined by measuring changes in intracellular cAMP, IP 3 , or Ca 2+ .

16 . The method of claim 13 , wherein the functional effect is determined by measuring binding of the compound to the polypeptide.

17 . The method of claim 13 , wherein the polypeptide comprises the amino acid sequence of SEQ ID NO:2, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:10, SEQ ID NO:12, SEQ ID NO:14, SEQ ID NO:20, SEQ ID NO:22, or SEQ ID NO:24.

18 . A method of detecting cancer cells in a biological sample from a mammal, the method comprising steps of:

(i) providing the biological sample from the mammal; and

(ii) detecting a nucleic acid molecule encoding a polypeptide comprising the amino acid sequence of SEQ ID NO:14 in a sample from the mammal, wherein an increase in level of the nucleic acid molecule in the sample compared to normal indicates the presence of cancer cells.
