Kallikrein-inhibitor therapies
View Patent ↗Methods are described for preventing or reducing ischemia, e.g., cerebral ischemia, and/or reperfusion injury, e.g., reperfusion injury associated with cerebral ischemia, in a patient.
1. A method for reducing reperfusion injury in a patient, the method comprising:
administering to the patient a composition comprising an inhibitor of plasma kallikrein, in an amount effective to reduce neurocognitive defects due to reperfusion injury, wherein the inhibitor of plasma kallikrein comprises a polypeptide that comprises the amino acid sequence of SEQ ID NO: 57.
2. The method of claim 1 , wherein the reperfusion injury is reperfusion injury associated with cerebral ischemia.
3. The method of claim 2 , wherein the cerebral ischemia is a stroke.
4. The method of claim 3 , wherein the stroke is selected from the group consisting of an embolism associated stroke, a thrombus-associated stroke, a thromboembolitic stroke and a hemorrhage-associated stroke.
5. The method of claim 3 , wherein the stroke is an embolism-associated stroke.
6. The method of claim 3 , wherein the stroke is a thrombus-associated stroke.
7. The method of claim 6 , wherein the thrombus-associated stroke is a perioperative stroke.
8. The method of claim 1 , wherein the polypeptide comprises the amino acid sequence of SEQ ID NO:2.
9. The method of claim 1 , wherein the inhibitor of plasma kallikrein is administered parenterally.
10. The method of claim 9 , wherein the inhibitor of plasma kallikrein is administered intravenously.
11. The method of claim 9 , wherein the inhibitor of plasma kallikrein is administered intraperitoneally.
12. The method of claim 1 , wherein the inhibitor of plasma kallikrein is conjugated to a water soluble polymer.
13. The method of claim 12 , wherein the water soluble polymer is a hydrophilic polyvinyl polymer or polymers.
14. The method of claim 13 , wherein the hydrophilic polyvinyl polymer is selected from the group consisting of polyvinylalcohol and polyvinylpyrrolidone.
15. The method of claim 13 , wherein the hydrophilic polyvinyl polymer is selected from the group consisting of polyalkylene oxide homopolymers, polypropylene glycols, polyoxyethylenated polyols, and copolymers thereof.
16. The method of claim 13 , wherein the water soluble polymer is polyethylene glycol (PEG).
