Abstract

Methods and compositions for diagnosing, detecting and treating a colon disease associated with differential expression of MABA1 in comparison to healthy cells. Also provided are antagonists or agonists of MABA1, and methods for screening agents that modulate the MABA1 level or activity in vivo or in vitro.

Claims (31)

1 . A method for diagnosing or detecting in a subject a colon cancer, the method comprising:

determining a test level or test activity of MABA1 protein in a colon cell from the subject, and

determining a control level or control activity in a colon cell from a healthy subject,

wherein the colon cancer is related to abnormal expression or function of MABA1 protein, and

wherein the test level or test activity in the cell from the subject is different from the control level or control activity in a colon cell from a healthy subject is indicative of the presence of the colon cancer.

2 . The method of claim 1 , wherein the level of the MABA1 protein is determined using an antibody that specifically binds to an antigenic region of MABA1.

3 . The method according to claim 1 , wherein the MABA1 protein comprises the amino acid sequence of SEQ ID NO:1.

4 . The method according to claim 1 , wherein the MABA1 protein is encoded by a polynucleotide sequence comprising the polynucleotide sequence selected from the group consisting of SEQ ID NO:2.

5 . The method of claim 1 , wherein the level of a nucleic acid molecule encoding MABA1 is determined.

6 . The method of claim 5 , wherein the level of the nucleic acid molecule is determined by contacting one or more probes that specifically hybridize to the nucleic acid molecule.

7 . A method for monitoring treatment of a colon cancer in a subject, wherein the colon cancer is related to abnormal expression or function of MABA1 protein, the method comprising:

determining a first test level or a first test activity of MABA1 protein in a colon cell from the subject prior to the treatment,

determining a second test level or a second test activity of MABA1 protein in a colon cell from the subject subsequent to the treatment, and

determining a control level or control activity in a colon cell from a healthy subject,

wherein the second test level or second test activity in the cell from the subject approaches the control level or control activity when compared to the first test level or first test activity is indicative of successful treatment.

8 . A method according to claim 1 , wherein the method determines recurrence of the colon cancer.

9 . A pharmaceutical composition comprising an antagonist to MABA1 and a pharmaceutically acceptable excipient.

10 . A pharmaceutical composition according to claim 9 , wherein the antagonist is an anti-MABA1 antibody.

11 . A pharmaceutical composition according to claim 9 , wherein the antagonist is an anti-sense nucleic acid molecule or an RNAi molecule that inhibits the translation or transcription of a gene that codes for the MABA1 protein.

12 . A pharmaceutical composition according to claim 9 , wherein the MABA1 protein comprises the amino acid sequence of SEQ ID NO:1.

13 . A pharmaceutical composition according to claim 9 , wherein the MABA1 protein is encoded by a polynucleotide sequence comprising the polynucleotide sequence selected from the group consisting of SEQ ID NO:2.

14 . A method for treating colon cancer, wherein the colon cancer is related to abnormal expression or function of MABA1 protein in a colon cell, the method comprising administering to a patient in need thereof an effective amount of the pharmaceutical composition according to claim 9 .

15 . A method of inhibiting cell growth or proliferation comprising contacting cells with MABA1 antibody.

16 . A method of inhibiting cell growth or proliferation comprising contacting cells with MABA1 RNAi.

17 . A method for screening for an agent that modulates MABA1 protein activity, the method comprising:

(i) contacting a candidate agent with a preparation of MABA1 protein, and

(ii) assaying for a MABA1 protein activity, wherein a change in said MABA1 protein activity in the presence of said agent relative to a MABA1 protein activity in the absence of said agent indicates said agent modulates MABA1 protein activity.

18 . A method for screening for an agent that modulates the level of expression of a nucleic acid that codes for a MABA1 protein in a cell the naturally expresses the MABA1 protein, the method comprises:

(i) contacting a candidate agent with the cell or a cell-free preparation from the cell wherein MABA1 protein is expressed, and

(ii) assaying for the level of expression of the MABA1 protein activity, wherein a change in said level in the presence of said agent relative to a level in the absence of said agent indicates said agent modulates the expression of MABA1 protein.

19 . A method according to claim 18 , wherein the cell is a colon cell.
