Abstract

The present disclosure relates to methods of DNA amplification with a first primer that has a random sequence of nucleotides at its 3′ end and a generic sequence 5′ of the random nucleotides, as well as a second primer with the generic sequence of the first primer. The disclosure further relates to a method of precipitating DNA on a solid medium to improve DNA amplification. In a preferred embodiment, the presently disclosed methods are used for high-throughput genotyping of DNA samples, such as bloodstains or trace amounts of DNA.

Claims (10)

1 - 70 . (canceled)

71 - 96 . (canceled)

97 . A method for preparing a DNA sample stored on a solid medium for amplification, comprising fixing said stored DNA sample to said solid medium with a solution comprising salt and alcohol, wherein said fixed DNA is suitable for amplification.

98 . A method for amplifying a DNA sample stored on a solid medium, comprising fixing said stored DNA sample to said solid medium with a solution comprising salt and alcohol, and amplifying said fixed DNA.

99 . The method of claim 97 or 98 , wherein said solid medium is filter paper.

100 . The method of claim 97 or 98 , wherein said filter paper is chemically treated.

101 . The method of claim 97 or 98 , further comprising dehydrating said fixed DNA sample.

102 . The method of claim 97 or 98 , wherein said DNA sample comprises tissue or blood.

103 . The method of claim 97 or 98 , wherein said salt is an acetate salt or a chloride salt.

104 . The method of claim 97 or 98 , wherein said alcohol is ethanol or isopropanol.
