Abstract

Described are methods for producing libraries of cells expressing at least two separate single polypeptide chain binding proteins, in which the binding proteins have different target epitopes. Such libraries are made by integration of the nucleic acid sequences encoding the polypeptide chains into the genome of the host cell, and selecting for cells that have successfully integrated these nucleic acids. The selected cells are preferably subjected to a cloning step. Mixtures of binding proteins are produced without having to individually produce each of the components of the mixture. A library of cells wherein essentially each cell encodes at least two single polypeptide chain binding proteins having different target epitopes is also herewith provided, as well as methods for producing a composition comprising at least two separate single polypeptide chain binding proteins having different target epitopes.

Claims (24)

1. A method for producing a single cell expressing at least two separate a single polypeptide chain binding protein (SPCBP) recognizing a first target epitope and a SPCBP recognizing a second target epitope, wherein binding of each SPCBP to its target epitope is mediated by a single protein domain, wherein the first target epitope and the second target epitope are different from one another, wherein the SPCBP recognizing a first target epitope and a SPCBP recognizing a second target epitope comprise a scaffold domain selected from lipocalin and DigA16, and wherein at least one of the SPCBPs is able to antagonize a function of a target molecule comprising at least one of the first target epitope or the second target epitope, the method comprising:

providing a first library of nucleic acid molecules encoding SPCBPs that recognize the first target epitope, wherein the nucleic acid molecules encoding the SPCBPs are known;

providing a second library of nucleic acid molecules encoding SPCBPs that recognize the second target epitope, wherein the nucleic acid molecules encoding the SPCBPs are known the SPCBP recognizing a first target epitope and a SPCBP recognizing a second target epitope comprise a scaffold domain selected from lipocalin and DigA16;

transfecting cells with the first and second libraries of nucleic acids;

selecting cells from the transfected cells having integrated therein a nucleic acid molecule encoding a SPCBP from the first library and a nucleic acid molecule encoding a SPCBP from the second library into their genomes, thereby producing a first library of cells, wherein each cell in said first library comprises genomic polynucleotides encoding at least a SPCBP recognizing the first target epitope and a SPCBP recognizing the second target epitope;

contacting the first library of cells, or supernatants thereof, with the first target epitope and with the second target epitope;

screening for and selecting cells from the contacted first library of cells, or supernatants thereof, expressing SPCBP able to bind the first target epitope and expressing SPCBP able to bind the second target epitope; thereby producing a second library of cells, wherein each cell in the second library of cells expresses an SPCBP able to bind the first target epitope and expresses a SPCBP able to bind the second target epitope

performing a bioassay on the second library of cells, or supernatants thereof, so as to measure antagonistic activity of the SPCBPs against a molecule comprising at least one of the first target epitope or the second target epitope; and

selecting based upon the results of the bioassay a cell from the second library that expresses a SPCBP able to antagonize a function of a target molecule comprising at least one of the first target epitope or the second target epitope.

A method for producing a single cell expressing at least two separate a single polypeptide chain binding protein (SPCBP) recognizing a first target epitope and a SPCBP recognizing a second target epitope, wherein binding of each SPCBP to its target epitope is mediated by a single protein domain, wherein the first target epitope and the second target epitope are different from one another, wherein the SPCBP recognizing a first target epitope and a SPCBP recognizing a second target epitope comprise a scaffold domain selected from lipocalin and DigA16, and wherein at least one of the SPCBPs is able to activate a function of a target molecule comprising at least one of the first target epitope or the second target epitope, the method comprising:

providing a first library of nucleic acid molecules encoding SPCBPs that recognize the first target epitope, wherein the nucleic acid molecules encoding the SPCBPs are known;

providing a second library of nucleic acid molecules encoding SPCBPs that recognize the second target epitope, wherein the nucleic acid molecules encoding the SPCBPs are known the SPCBP recognizing a first target epitope and a SPCBP recognizing a second target epitope comprise a scaffold domain selected from lipocalin and DigA16;

transfecting cells with the first and second libraries of nucleic acids;

selecting cells from the transfected cells having integrated therein a nucleic acid molecule encoding a SPCBP from the first library and a nucleic acid molecule encoding a SPCBP from the second library into their genomes, thereby producing a first library of cells, wherein each cell in said first library comprises genomic polynucleotides encoding at least a SPCBP recognizing the first target epitope and a SPCBP recognizing the second target epitope;

contacting the first library of cells, or supernatants thereof, with the first target epitope and with the second target epitope;

screening for and selecting cells from the contacted first library of cells, or supernatants thereof, expressing a SPCBP able to bind the first target epitope and expressing a SPCBP able to bind the second target epitope; thereby producing a second library of cells, wherein each cell in the second library of cells expressing a SPCBP able to bind the first target epitope and expresses a SPCBP able to bind the second target epitope;

performing a bioassay on the second library of cells, or supernatants thereof, so as to measure activating activity of the SPCBPs against a molecule comprising at least one of the first target epitope or the second target epitope; and

based upon the results of the bioassay, selecting based upon the results of the bioassay a cell from the second library that expresses a SPCBP able to activate a function of a target molecule comprising at least one of the first target epitope or the second target epitope.

3. The method according to claim 1 , wherein each cell in the first library of cells expresses the SPCBP recognizing a first target epitope and the SPCBP recognizing a second target epitope at a different ratio.

4. The method according to claim 2 , wherein each cell in the first library of cells expresses the SPCBP recognizing a first target epitope and the SPCBP recognizing a second target epitope at a different ratio.

5. The method according to claim 1 , wherein the first target epitope and the second target epitope are present on the same target molecule.

6. The method according to claim 2 , wherein the first target epitope and the second target epitope are present on the same target molecule.

7. The method according to claim 1 , wherein the first target epitope and the second target epitope are present on different target molecules.

8. The method according to claim 2 , wherein the first target epitope and the second target epitope are present on different target molecules.
