Abstract

The present invention relates to substituted triazole compounds and compositions comprising substituted triazole compounds. The invention further relates to methods of inhibiting the activity of Hsp90 in a subject in need thereof and methods for preventing or treating hyperproliferative disorders, such as cancer, in a subject in need thereof comprising administering to the subject a substituted triazole compound of the invention, or a composition comprising such a compound.

Claims (86)

1. A compound represented by the following structural formula:

or a tautomer or a pharmaceutitcally acceptable salt thereof, wherein:

ring A is an aryl, wherein the aryl is optionally timber substituted with one or more substituents in addition to R 3 ;

R a and R b , for each occurrence, is independently —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl or heteroaryl or an optionally substituted aralkyl; or R a and R b , taken together with the nitrogen to which they are attached, form an optionally substituted heteroaryl or heterocyclyl;

R 7 and R 8 , for each occurrence, is independently, —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteroaralkyl;

R 10 and R 11 , for each occurrence, is independently —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteroaralkyl; or R 10 and R 11 , taken together with the nitrogen to which they are attached, form an optionally substituted heterocyclyl or an optionally substituted heteroaryl;

R 26 is a C1-C6 alkyl;

p, for each occurrence, is independently, 0, 1 or 2; and

m, for each occurrence, is independently, 1, 2, 3 or 4.

2. The compound of claim 1 wherein the compound is represented by the following structural formula:

The compound of claim 8 , wherein R a and R b are each independently a hydrogen, a C1-C6 straight or branched alkyl, optionally substituted by —OH, —CN, —SH, amino, a C1-C6 alkoxy, alkylsulfanyl, alkylamino, dialkylamino or a cycloalkyl; or R a and R b taken together with the nitrogen to which they are attached form a substituted or unsubstituted nonaromatic, nitrogen-containing heterocyclyl.

10. The compound of claim 9 , wherein R 6 is a C1-C6 alkyl, a C1-C6 haloalkyl, a C1-C6 alkoxy, a C1-C6 haloalkoxy, a C1-C6 alkyl sulfanyl or a C3-C6 cycloalkyl.

11. The compound of claim 10 , wherein R 1 is —OH, —SH or —NHR 7 .

12. The compound of claim 11 , wherein R 30 is a —OH, —SH, halogen, cyano, a C1-C6 alkyl, C1-C6 haloalkyl, C1-C6 alkoxy, C1-C6 haloalkoxy or C1-C6 alkyl sulfanyl.

13. The compound of claim 12 , wherein R 1 , R 3 and R 25 for each occurance, is independently —SH or —OH, and R 6 is cyclopropyl or isopropyl.

14. The compound of claim 13 , wherein R 30 is a methyl, ethyl, propyl, isopropyl, methoxy or ethoxy.

15. The compound of claim 2 , wherein R 1 , R 3 and R 25 are each independently —SH or —OH; and R 6 is cyclopropyl or isopropyl.

16. The compound of claim 14 , wherein R a and R b are each independently a hydrogen, methyl, ethyl, propyl, isopropyl, or taken together with the nitrogen to which they are attached, are:

18. A method of treating a cancer in a mammal, wherein the cancer is selected from the group consisting of breast carcinoma, non-small cell lung cancer, leukemia and mammary carcinoma, comprising administering to the mammal an effective amount of a compound represented by the following structure:

or a tautomer or a pharmaceutically acceptable salt thereof, wherein:

ring A is an aryl, wherein the aryl is optionally further substituted with one or more substituents in addition to R 3 ;

R 26 is a C1-C6 alkyl;

19. A pharmaceutical composition, comprising a pharmaceutically acceptable carrier and one or more compounds represented by the following structural formula:

or a tautomer, pharmaceutically acceptable salt, wherein:

ring A is an aryl, wherein the aryl is optionally further substituted with one or more substituents in addition to R 3 ;

30. The method of claim 29 , wherein R 30 is a —OH, —SH, halogen, cyano, a C1-C6 alkyl, C1-C6 haloalkyl, C1-C6 alkoxy, C1-C6 haloalkoxy or C1-C6 alkyl sulfanyl.

31. The method of claim 30 , wherein R 1 , R 3 and R 25 for each occurance, is independently —SH or —OH, and R 6 is cyclopropyl or isopropyl.
