Endosomolytic polymers
View Patent ↗We describe pH-sensitive endosomolytic polymers, delivery particles containing pH-sensitive endosomolytic polymers. The described particles are capable of delivering polynucleotides to cells from the peripheral circulation with subsequent release from endosomes. The endosomolytic polymers are inactive outside the cell but disrupt membranes upon exposure to an acidified endosomal compartment.
1. An endosomolytic polymer made by the process comprising:
a) forming a membrane active polyamine capable of disrupting a cell membrane, and
b) reversibly modifying a plurality of amines on the polyamine by attachment of disubstituted maleic anhydrides having the structure:
to form a plurality of maleamates, thereby forming a polyanion that is not capable of disrupting red blood cells wherein cleavage of the maleamates from the polyanion regenerates the amines and restores cell membrane disruption activity of the polyamine.
2. The endosomolytic polymer of claim 1 wherein said membrane active polyamine has a molecular weight of at least 10,000 Daltons.
