KINASE AND PHOSPHATASE ACTIVITY MEASUREMENT USING FLUORESCENT POLARIZATION
A method for quantitating enzyme activity that causes phosphorylation or dephosphorylation of a peptide or protein substrate is provided. The method utilizes the differences in fluorescence polarization of a phosphorylated amino acid-containing fluorescence-emitting reporter molecule free in solution as compared to being bound to a binding molecule.
1 . A kit comprising:
(a) a first peptide containing a first phosphorylated amino acid and a fluorescent moiety,
(b) a second peptide containing an unphosphorylated amino acid, wherein a kinase is capable of phosphorylating the unphorphorylated amino acid to form a third peptide containing a second phosphorylated amino acid, and
(c) a binding molecule capable of binding to the first peptide and third peptide, wherein the binding molecule exhibits discriminate recognition between the second peptide versus the first and third peptides.
2 . The kit of claim 1 , wherein the binding molecule is a protein.
3 . The kit of claim 1 , wherein the binding molecule is an antibody.
4 . The kit of claim 1 , wherein the binding molecule is an antibody capable of binding to an epitope containing a phosphoserine residue, a phosphothreonine residue, or a phosphotyrosine residue.
5 . The kit of claim 1 , wherein the fluorescent moiety is a fluorescein moiety.
6 . The kit of claim 1 , wherein the fluorescent moiety is a rhodamine moiety.
7 . The kit of claim 1 , wherein the fluorescent moiety is selected from the group consisting of a pyrene, a lucifer yellow, a BODIPY®, a malachite green, a coumarin, a dansyl derivative, a mansyl derivative, a dabsyl derivative, a NBD flouride, a stillbene, an anthrocene, an acridine, a rosamine, a TNS chloride, an ATTO-TAG™, a Lissaminem™ derivative, an eosin, a naphthalene derivative, an ethidium bromide derivative, a thiazole orange derivative, an ethenoadenosine, a CyDyes™, an aconitine, an Oregon Green, and a Cascade Blue.
8 . The kit of claim 1 , wherein the first peptide has the sequence F-RRRVTpSARRS (SEQ ID NO:1) or F-pSAARRS (SEQ ID NO:2), wherein F represents a fluorescein moiety, and wherein pS represents a phosphoserine residue.
9 . The kit of claim 1 , further comprising a kinase.
10 . The kit of claim 1 , further comprising a solution comprising EDTA.
11 . The kit of claim 1 , further comprising a solution comprising ATP.
12 . The kit of claim 1 , wherein (a) and (c) form a complex.
13 . A kit comprising:
(a) a first peptide containing a first phosphorylated amino acid and a fluorescent moiety,
(b) a second peptide containing an unphosphorylated amino acid, wherein a kinase is capable of phosphorylating the unphorphorylated amino acid to form a third peptide containing a second phosphorylated amino acid,
(c) an antibody capable of binding to the first peptide and third peptide, wherein the antibody exhibits discriminate recognition between the first and third peptides versus the second peptide, and
(d) a solution comprising EDTA.
14 . The kit of claim 13 , wherein the antibody is capable of binding to an epitope containing a phosphoserine residue, a phosphothreonine residue, or a phosphotyrosine residue.
15 . The kit of claim 13 , wherein the fluorescent moiety is a fluorescein moiety.
16 . The kit of claim 13 , wherein the fluorescent moiety is selected from the group consisting of a rhodamine, a pyrene, a lucifer yellow, a BODIPY®, a malachite green, a coumarin, a dansyl derivative, a mansyl derivative, a dabsyl derivative, a NBD flouride, a stillbene, an anthrocene, an acridine, a rosamine, a TNS chloride, an ATTO-TAG™, a Lissaminem™ derivative, an eosin, a naphthalene derivative, an ethidium bromide derivative, a thiazole orange derivative, an ethenoadenosine, a CyDyes™, an aconitine, an Oregon Green, and a Cascade Blue.
17 . The kit of claim 13 , wherein the first peptide has the sequence F-RRRVTpSARRS (SEQ ID NO:1) or F-pSAARRS (SEQ ID NO:2), wherein F represents a fluorescein moiety, and wherein pS represents a phosphoserine residue.
18 . The kit of claim 13 , further comprising a kinase.
19 . The kit of claim 13 , wherein (a) and (c) form a complex.
20 . The kit of claim 13 , further comprising a solution comprising ATP.
