Nitrogen containing heterocyclyl substituted imidazoquinolines and imidazonaphthyridines
View Patent ↗Imidazoquinoline and imidazonaphthyridine compounds having a nitrogen-containing heterocyclyl substituent at the 5-, 6-, 7-, or 8-position, pharmaceutical compositions containing the compounds, intermediates, and methods of making and methods of use of these compounds as immunomodulators, for modulating cytokine biosynthesis in animals and in the treatment of diseases including viral and neoplastic diseases are disclosed.

wherein the heterocyclic ring system is unsubstituted or substituted by one or more substituents selected from the group consisting of:
alkoxy,
hydroxy,
nitro,
oxo,
hydroxyalkyl,
alkyl,
phenyl optionally substituted by one or more halogens,
benzyl optionally substituted by one or more halogens,
—N(R 8 )-Q-R 4 ,
—X—N(R 8 )-Q-R 4 ,
—X—O-phenyl, wherein phenyl is optionally substituted by one or more substituents selected from the group consisting of alkoxy and halogen,
—X-pyrrolidinyl, —X-piperidinyl, —X-morpholinyl, and —X-piperazinyl, wherein pyrrolidinyl, piperidinyl, morpholinyl, and piperazinyl are unsubstituted or substituted by one or more substituents selected from the group consisting of alkyl, hydroxy, hydroxyalkyl, acetyl, and acetamido;
R 1 is selected from the group consisting of alkyl, alkoxyalkylenyl, and hydroxyalkylenyl;
R 2 is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, and hydroxyalkylenyl;
X is alkylene,
R 4 is alkyl;
Q is selected from the group consisting of a bond and —C(R 6 )—;
R 6 is selected from the group consisting of ═O and ═S;
R 8 is selected from the group consisting of hydrogen and alkyl; and
R is selected from the group consisting of hydrogen, alkyl, alkoxy, trifluoromethyl, chloro, fluoro, and hydroxy; or a pharmaceutically acceptable salt thereof.
2. The compound or salt of claim 1 wherein
is attached at the 7 position.
3. The compound or salt of claim 1 wherein R 1 is selected from the group consisting of propyl, 2-methylpropyl, 2-hydroxy-2-methylpropyl, 2,3-dihydroxypropyl, and 3-isopropoxypropyl.
4. The compound or salt of claim 1 wherein

5. A pharmaceutical composition comprising a therapeutically effective amount of a compound or salt of claim 1 and a pharmaceutically acceptable carrier.
6. The compound or salt of claim 1 , wherein R 2 is selected from the group consisting of hydrogen, C 1-4 alkyl, C 1-4 alkyl-O—C 1-4 alkylenyl, and HO—C 1-4 alkyleneyl.
7. The compound or salt of claim 6 , wherein R 2 is selected from the group consisting of hydrogen, methyl, ethyl, n-propyl, n-butyl, ethoxymethyl, methoxymethyl, 2-methoxyethyl, hydroxymethyl, and 2-hydroxyethyl.
8. The compound or salt of claim 1 , wherein R is hydrogen.
9. The compound or salt of claim 4 wherein
is attached at the 7 position.
10. The compound or salt of claim 4 wherein R 1 is selected from the group consisting of propyl, 2-methylpropyl, 2-hydroxy-2-methylpropyl, 2,3-dihydroxypropyl, and 3-isopropoxypropyl.
11. A pharmaceutical composition comprising a therapeutically effective amount of a compound or salt of claim 4 and a pharmaceutically acceptable carrier.
12. The compound or salt of claim 4 , wherein R 2 is selected from the group consisting of hydrogen, C 1-4 alkyl, C 1-4 alkyl-O—C 1-4 alkylenyl, and HO—C 1-4 alkyleneyl.
13. The compound or salt of claim 12 , wherein R 2 is selected from the group consisting of hydrogen, methyl, ethyl, n-propyl, n-butyl, ethoxymethyl, methoxymethyl, 2-methoxyethyl, hydroxymethyl, and 2-hydroxyethyl.
14. The compound or salt of claim 4 , wherein R is hydrogen.
