Abstract

A cassette for preparing a sample is disclosed herein. The cassette includes a housing, which encloses the structures and the processes used to prepare the sample.

Claims (60)

1. An apparatus, comprising:

a first housing defining a first chamber, a second chamber and an opening in fluid communication with the first chamber, the first chamber configured to contain a sample, a binding solution and a magnetic particle, the binding solution formulated to bind a portion of the sample to the magnetic particle;

a second housing including a side wall and a puncturable member, the side wall and the puncturable member collectively defining a first reagent chamber containing the binding solution and a second reagent chamber containing a plurality of magnetic particles, the magnetic particle being one of the plurality of magnetic particles, a portion of the second housing configured to be disposed within the first chamber of the first housing via the opening such that the first reagent chamber is in fluid communication with the first chamber when a first portion of the puncturable member is punctured and the second reagent chamber is in fluid communication with the first chamber when a second portion of the puncturable member is punctured;

a valve configured to transfer the magnetic particle and the portion of the sample from the first chamber to the second chamber while fluidically isolating the first chamber from the second chamber; and

a pump coupled to the first housing, the pump configured to mix a reagent with the portion of the sample, the pump including an actuator disposed outside of the first housing and a nozzle disposed within the second chamber.

The apparatus of claim 1 , wherein the valve includes a valve housing and a valve member, the valve housing defining a first opening in fluid communication with the first chamber and a second opening in fluid communication with the second chamber, the valve member defining a cavity in fluid communication with the first opening when the valve member is in a first position, the cavity in fluid communication with the second opening when the valve member is in a second position.

3. The apparatus of claim 1 , wherein the valve includes a valve housing and a valve member, the valve member movably disposed within the valve housing, the valve member defining an opening configured to receive a magnetic shaft.

4. The apparatus of claim 1 , wherein the housing defines a third chamber, and the valve is a first valve, the apparatus further comprising:

a second valve configured to transfer the magnetic particle and the portion of the sample from the second chamber to the third chamber while fluidically isolating the second chamber from the third chamber.

5. The apparatus of claim 1 , wherein the valve includes a valve housing and a valve member, the valve member movably disposed within the valve housing, a longitudinal axis of the valve member being substantially parallel to a longitudinal axis of the nozzle of the pump.

6. The apparatus of claim 1 , wherein a first surface of the housing defines a valve opening through which a portion of the valve is disposed and a second surface of the housing defines a pump opening through which a portion of the pump is disposed, the first surface opposite the second surface.

The apparatus of claim 1 , wherein a first surface of the housing defines a valve opening through which a portion of the valve is disposed and a second surface of the housing defines a pump opening through which a portion of the pump is disposed, the first surface substantially parallel to the second surface.

8. The apparatus of claim 1 , wherein the actuator of the pump includes a bellows.

9. The apparatus of claim 1 , wherein the actuator of the pump is constructed from a flexible material.

10. The apparatus of claim 1 , wherein the pump is a first pump, the apparatus further comprising:

a second pump coupled to the housing, the second pump configured to mix at least the binding solution and the magnetic particle within the first chamber, the second pump including an actuator disposed outside of the housing and a nozzle disposed within the first chamber.

11. The apparatus of claim 1 , wherein the second housing includes an attachment portion configured to be matingly coupled to an attachment portion of the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

12. The apparatus of claim 1 , wherein the second housing includes a seal portion configured to produce a seal between the second housing and the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

13. The apparatus of claim 1 , further comprising: an actuator disposed within the first reagent chamber, the actuator configured to puncture the first portion of the puncturable member.

The apparatus of claim 1 , further comprising: an actuator disposed within the first reagent chamber, the actuator including a plunger portion configured to move the binding solution from the first reagent chamber to the first chamber when the actuator is moved, the actuator including a protrusion configured to puncture the first portion of the puncturable member when the actuator is moved.

15. An apparatus, comprising:

a first housing defining a first chamber, a second chamber and an opening, the first chamber configured to contain a sample;

a second housing including a side wall and a puncturable member, the side wall and the puncturable member collectively defining a reagent chamber, a portion of the second housing configured to be disposed within the first chamber of the first housing via the opening such that the reagent chamber is in fluid communication with the first chamber when a portion of the puncturable member is punctured;

an actuator configured to puncture the portion of the puncturable member;

a valve configured to transfer a portion of the sample from the first chamber of the first housing to the second chamber of the first housing while fluidically isolating the first chamber from the second chamber; and

a pump configured to mix a reagent with the portion of the sample when the reagent and the portion of the sample are in the first chamber of the first housing, the pump including an actuator disposed outside of the first housing and a nozzle disposed within the first chamber.

16. The apparatus of claim 15 , wherein the second housing includes an attachment portion configured to be matingly coupled to an attachment portion of the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

17. The apparatus of claim 15 , wherein:

the first housing defines an attachment opening; and

the second housing includes an attachment protrusion configured to be matingly disposed within the attachment opening to limit movement of the second housing relative to the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

18. The apparatus of claim 15 , further comprising:

a seal member disposed about the portion of the second housing, the seal member configured to produce a seal between the second housing and the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

19. The apparatus of claim 15 , wherein the actuator is disposed within the reagent chamber.

20. The apparatus of claim 15 , wherein the actuator is disposed within the reagent chamber, the actuator including a plunger and a protrusion, the plunger configured to move a reagent from the reagent chamber into the first chamber when the actuator is moved, the protrusion configured to puncture the portion of the puncturable member when the actuator is moved.

21. The apparatus of claim 15 , wherein the actuator is disposed within the reagent chamber, the actuator including a plunger configured to move a reagent from the reagent chamber into the first chamber when the actuator is moved by an actuator shaft, the plunger defining a recess configured to receive the actuator shaft.

The apparatus of claim 15 , wherein the valve includes a valve housing and a valve member, the valve housing defining a first opening in fluid communication with the first chamber and a second opening in fluid communication with the second chamber, the valve member defining a cavity in fluid communication with the first opening when the valve member is in a first position, the cavity in fluid communication with the second opening when the valve member is in a second position.

23. The apparatus of claim 15 , wherein the valve includes a valve housing and a valve member, the valve member movably disposed within the valve housing, the valve member defining an opening configured to receive a magnetic shaft.

24. An apparatus, comprising:

a first housing defining a first chamber, a second chamber and an opening, the first chamber configured to contain a sample;

a second housing including a side wall and a puncturable member, the side wall and the puncturable member collectively defining a reagent chamber configured to contain a reagent, a portion of the second housing configured to be disposed within the first chamber of the first housing via the opening such that the reagent chamber is in fluid communication with the first chamber when a portion of the puncturable member is punctured;

a mixing mechanism coupled to the second housing, the mixing mechanism configured to mix a reagent with the sample when the reagent and the portion of the sample are in the first chamber of the first housing, the mixing mechanism including a mixing actuator disposed outside of the second housing and an elongated portion disposed within the first chamber; and

a valve configured to transfer a portion of the sample from the first chamber of the first housing to the second chamber of the first housing while fluidically isolating the first chamber from the second chamber.

25. The apparatus of claim 24 , wherein:

the first housing defines an attachment opening; and

the second housing includes an attachment protrusion configured to be matingly disposed within the attachment opening to limit movement of the second housing relative to the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

26. The apparatus of claim 24 , further comprising:

a seal member disposed about the portion of the second housing, the seal member configured to produce a seal between the second housing and the first housing when the portion of the second housing is disposed within the first chamber of the first housing.

The apparatus of claim 24 , wherein the valve includes a valve housing and a valve member, the valve housing defining a first opening in fluid communication with the first chamber and a second opening in fluid communication with the second chamber, the valve member defining a cavity in fluid communication with the first opening when the valve member is in a first position, the cavity in fluid communication with the second opening when the valve member is in a second position.

28. The apparatus of claim 24 , wherein the valve includes a valve housing and a valve member, the valve member movably disposed within the valve housing, the valve member defining an opening configured to receive a magnetic shaft.

29. The apparatus of claim 24 , wherein the mixing mechanism is a pump, the mixing actuator being a bellows, the elongated portion being a nozzle.

30. The apparatus of claim 24 , further comprising:

a reagent actuator configured to puncture the portion of the puncturable member.

31. The apparatus of claim 24 , further comprising:

a reagent actuator disposed within the reagent chamber, the reagent actuator configured to move a reagent from the reagent chamber into the first chamber when the reagent actuator is moved.

32. The apparatus of claim 24 , further comprising:

a reagent actuator disposed within the reagent chamber, the reagent actuator including a plunger and a protrusion, the plunger configured to move a reagent from the reagent chamber into the first chamber when the reagent actuator is moved, the protrusion configured to puncture the portion of the puncturable member when the reagent actuator is moved.

The apparatus of claim 24 , further comprising: a reagent actuator disposed within the reagent chamber, the reagent actuator including a plunger configured to move a reagent from the reagent chamber into the first chamber when the reagent actuator is moved by a reagent actuator shaft, the plunger defining a recess configured to receive the reagent actuator shaft.

34. The apparatus of claim 24 , wherein the sample contains a nucleic acid, the reagent is an enzyme configured to isolate the nucleic acid from a remainder of the sample, the apparatus further comprising:

an actuator configured to puncture a portion of the puncturable member and move the enzyme from the reagent chamber into the first chamber,

the valve configured to transfer the nucleic acid from the first chamber of the first housing to the second chamber of the first housing.
