Abstract

The present invention discloses a modified tumor necrosis factor-alpha converting enzyme (TACE) catalytic domain, that unlike the native TACE catalytic domain, is stable at high protein concentrations. The present invention further discloses methods for generating crystals of the modified TACE protein in protein-ligand complexes with a number of inhibitors. In addition, the present invention discloses methods of using the proteins, crystals and/or three-dimensional structures obtained to identify compounds that can modulate the enzymatic activity of TACE.

Claims (17)

1. A method for identifying an agent for use as an inhibitor of tumor necrosis factor- alpha converting enzyme (TACE) comprising:

(a) employing the atomic coordinates of Table 3 to generate a three dimensional model which defines the atomic coordinates of a TACE catalytic domain having Valine 353 substituted for Glycine in the full length enzyme (wherein said substitution is at position 139 of said SEQ ID NO: 8) complexed to a ligand;

(b) utilizing the model from step (a) to determine the points of interaction between said ligand and TACE to generate a specific three-dimensional target;

(c) employing said target from step (b) to design or select potential agents by performing rational drug design;

(d) synthesizing and/or contacting the potential agent with a proteolytic polypeptide comprising the catalytic domain of TACE or an active fragment thereof; and

(e) determining the catalytic activity of said proteolytic polypeptide in the TACE activity assay; wherein a potential agent is identified as an agent that inhibits TACE when there is a decrease in the activity of said proteolytic polypeptide in the presence of the agent relative to in its absence.

2. The method of claim 1 wherein said proteolytic polypeptide comprising the catalytic domain of TACE or an active fragment thereof comprises:

(i) a modified TACE catalytic domain, wherein said modified TACE has at least 95% identity with the amino acid sequence of SEQ ID NO: 8;

(ii) catalyzes the proteolytic cleavage of a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 17; and

(iii) is capable of binding to N-{D,L-2-(hydroxyaminocarbonyl)methyl-4-methylpentanoyl}-L-3-amino-2-dimethylbutanoyl-L- alanine, 2-(amino)ethyl amide.

3. A method for identifying a compound that is predicted to inhibit tumor necrosis factor-alpha converting enzyme (TACE) comprising:

(a) generating a three-dimensional structure of a complex between the modified catalytic domain of TACE, which modified TACE catalytic domain comprises a V353G mutation, and N-{3-(hydroxyaminocarbonyl)-l-oxo-(2R)- benzylpropyl}-Ile-Leu-OH with the atomic coordinates in Table 3;

(b) screening and selecting one or more compounds from a peptide library or chemical library; and

(c) identifying one or more of said compounds compound that are predicted to inhibit TACE; wherein said identifying is performed using the structure defined in step (a).

4. The method of claim 3 , further comprising:

(c) contacting the compound with a proteolytic polypeptide comprising the catalytic domain of tumor necrosis factor-alpha converting enzyme (TACE) or an active fragment thereof; and

(d) determining the catalytic activity of said proteolytic polypeptide in a TACE activity assay; wherein the compound is identified as an agent that inhibits TACE when there is a decrease in the activity of said proteolytic polypeptide in the presence of the agent relative to in its absence.
