Abstract

Amphoteric liposomes are proposed, which comprise positive and negative membrane-based or membrane-forming charge carriers as well as the use of these liposomes.

Claims (35)

1. Amphoteric liposomes loaded with nucleic acids, wherein the liposomes comprise at least one amphipathic cationic lipid, at least one amphipathic anionic lipid, and at least one neutral lipid, and wherein the liposomes are stable at pH 4.2 and pH 7.5 and are prepared by a process comprising the steps of:

a. providing a mixture of at least one amphipathic cationic lipid, at least one amphipathic anionic lipid, at least one neutral lipid, and nucleic acids and at least one solvent, wherein said mixture has a pH between about 3-6

b. forming liposomes in said mixture; and

c. changing the pH of said liposomes to pH 7-9.

Amphoteric liposomes loaded with nucleic acids, wherein the liposomes comprise at least one amphipathic lipid with both a positive and a negative charge, and at least one neutral lipid, and wherein the amphoteric liposomes are stable at pH 4.2 and pH 7.5 and are prepared by a process comprising the steps of:

a. providing a mixture of at least one amphipatic lipid with both a positive and a negative charge, at least one neutral lipid, and nucleic acids and at least one solvent, wherein said mixture has a pH between about 3-6;

b. forming liposomes in said mixture; and

c. changing the pH of said liposomes to pH 7-9.

3. The liposomes of claim 2 , wherein said liposomes further comprise at least one amphipatic lipid with a positive charge or at least one amphipatic lipid with a negative charge.

4. The liposomes of any one of claims 1 , 2 , or 3 , wherein said process further comprises the step:

d. removing non-encapsulated nucleic acids from said mixture.

5. The liposomes of any one of claims 1 , 2 , or 3 , wherein said neutral lipid is selected from the group consisting of phosphatidyl choline, phosphatidyl ethanolamine, cholesterol, tetraether lipid, ceramide, sphingolipid, and diacyl glycerol.

6. The liposomes of claim 1 , wherein said anionic lipid is a weak anion and said cationic lipid is a strong cation and said anionic lipid is present in excess over said cationic lipid.

7. The liposomes of claim 6 , wherein said anionic lipid is selected from the group consisting of cholesterol hemisuccinate (CHEMS), diacyl glycerol hemisuccinate, fatty acids and phosphatidylserine.

8. The liposomes of claim 6 , wherein cationic lipid is selected from the group consisting of DOTAP, DC-Chol, DORIE, DDAB, TC-Chol, DOTMA, DOGS, (C18) 2 Gly + N,N-dioctadecylamido-glycin, CTAB, CPyC and DOEPC.

9. The liposomes of claim 6 , wherein said anionic lipid is CHEMS or diacylglycerol hemisuccinate, said cationic lipid is DOTAP or DC-Chol, and said neutral lipid is phosphatidylcholine, cholesterol, phosphatidylethanolamine, or mixtures thereof.

10. The liposomes of claim 9 , wherein the lipids comprise about 30-60 mol. % POPC, about 10-30 mol. % DOTAP and about 25-40 mol. % CHEMS.

11. The liposomes of claim 1 , wherein said anionic lipid is a weak anion and the cationic lipid is a weak cation.

12. The liposomes of claim 11 , wherein said anionic lipid is selected from the group consisting of cholesteryl hemisuccinate (CHEMS), diacyl glycerol hemisuccinate, fatty acids and phosphatidyl serine.

13. The liposomes of claim 11 , wherein said cationic lipid is selected from the group consisting of HisChol and MoChol.

14. The liposomes of claim 11 , wherein said anionic lipid is CHEMS or diacylglycerol hemisuccinate, said cationic lipid is HisChol or MoChol and said neutral lipid is phosphatidylcholine, cholesterol, phosphatidylethanolamine, or mixtures thereof.

15. The liposomes of claim 14 , wherein said lipids comprise about 55-60 mol. % POPC, about 20-40 mol. % HisChol and about 5-20 mol. % CHEMS.

16. The liposomes of claim 1 , wherein said anionic lipid is a strong anion and said cationic lipid is a weak cation and said cationic lipid is present in excess over said anionic lipid.

17. The liposomes of claim 16 , wherein said anionic lipid is selected from the group consisting of cholesterol sulphate, cholesterol phosphate, phosphatidyl glycerol, phosphatidic acid, phosphatidyl inositol, and cetyl phosphate.

18. The liposomes of claim 16 , wherein said cationic lipid is selected from the group consisting of HisChol and MoChol.

19. The liposomes of claim 16 , wherein said anionic lipid is phosphatidylglycerol, phosphatidic acid or cetyl phosphate and said cationic lipid is HisChol or MoChol and said neutral lipid is phosphatidylcholine, cholesterol, phosphatidylethanolamine, or mixtures thereof.

20. The liposomes of claim 19 , wherein said lipids comprise about 47.5 mol. % POPC, about 40 mol. % HisChol and about 12.5 mol. % DPPG.

21. The liposomes of any one of claims 1 , 2 , or 3 , wherein said liposomes have an average size of between 50 and 1000 nm.

22. The liposomes of claim 21 , wherein said liposomes have an average size of between 70 and 250 nm.

23. The liposomes of claim 21 , wherein said liposomes have an average size of between 60 and 130 nm.

24. The liposomes of any one of claims 1 , 2 , or 3 , wherein the nucleic acids are selected from the group consisting of DNA, RNA, antisense nucleotides, and decoy nucleotides.

25. The liposomes of claim 4 , wherein, following step d., at least 80% of said nucleic acids are encapsulated within said liposomes.

The liposomes of any one of claims 1 , 2 , or 3 , wherein said forming step b. comprises a method selected from the group consisting of the injection of an ethanolic lipid solution into an aqueous nucleic acid solution, drying an organic solution of said lipids and hydrating the dry lipid films with aqueous nucleic acid solution, or detergent dialysis of said lipids and nucleic acids.

27. The liposomes of any one of claims 1 , 2 , or 3 , wherein said liposomes are sized using a method selected from the group consisting of high pressure homogenization and extrusion.

28. The liposomes of any one of claims 1 , 2 , or 3 , wherein said liposomes have an isoelectric point of between 5 and 7.
