Abstract

Novel conjugates of taxol and novel taxol immunogens derived from the 9 and 7 positions of taxol and monoclonal antibodies generated by these taxol linked immunogens are useful in immunoassays for the quantification and monitoring of taxol in biological fluids.

Claims (77)

1 . A compound of the formula:

Y is an organic spacing group;

X is a functional terminal group capable of binding to a polyamine polypeptide; and

p is an integer from 0 to 1.

2 . The compound of claim 1 , wherein p is 0.

3 . The compound of claim 2 , wherein X is

wherein R 3 is hydrogen or taken together with its attached oxygen atom forms a reactive ester and R 4 is oxygen or sulfur.

4 . The compound of claim 3 , wherein X is

and R 3 is hydrogen.

5 . The compound of claim 3 , wherein X is

and R 3 forms a reactive ester.

6 . The compound of claim 5 , wherein the ester formed is a lower alkyl ester, imidoester or amidoester.

7 . The compound of claim 1 , wherein p is 1.

8 . The compound of claim 7 , wherein Y is alkylene containing from 1 to 10 carbon atoms,

wherein n and o are integers from 0 to 6, and m is an integer from 1 to 6.

9 . A compound of the formula:

wherein Y is an organic spacing group;

X is a functional terminal group capable of binding to a polyamine

polypeptide; and B is —CH 2 — or

or mixtures thereof.

10 . The compound of claim 9 , wherein p is 0.

11 . The compound of claim 10 , wherein X is

wherein R 3 is hydrogen or taken together with its attached oxygen atom forms a reactive ester and R 4 is oxygen or sulfur.

12 . The compound of claim 11 , wherein X is

and R 3 is hydrogen.

13 . The compound of claim 12 , wherein X is

and R 3 forms a reactive ester.

14 . The compound of claim 13 , wherein the ester formed is a lower alkyl ester, imidoester or amidoester.

15 . The compound of claim 14 , wherein p is 1.

16 . The compound of claim 15 , wherein X is

wherein R 3 is hydrogen or taken together with its attached oxygen atom forms a reactive ester and R 4 is oxygen or sulfur.

17 . The compound of claim 16 , wherein Y is alkylene containing from 1 to 10 carbon atoms,

wherein n and o are integers from 0 to 6, and m is an integer from 1 to 6.

18 . A conjugate of a carrier with a compound of the formula:

Y is an organic spacing group;

X is a functional terminal group capable of binding to a polyamine polypeptide; and

p is an integer from 0 to 1.

19 . The conjugate of claim 18 , wherein p is 0.

20 . The conjugate of claim 19 , wherein X is

wherein R 3 is hydrogen or taken together with its attached oxygen atom forms a reactive ester and R 4 is oxygen or sulfur.

21 . The conjugate of claim 19 , wherein p is 1 and Y is alkylene containing from 1 to 10 carbon atoms,

wherein n and o are integers from 0 to 6, and m is an integer from 1 to 6.

22 . The conjugate of claim 21 , wherein the carrier contains one or more amino groups linked by

wherein R 4 is oxygen or sulfur.

23 . A compound of the formula:

wherein X, Y and p are as above and B is —CH 2 — or

or mixtures thereof.

24 . The compound of claim 23 , wherein p is 0.

25 . The compound of claim 24 , wherein X is

wherein R 3 is hydrogen or taken together with its attached oxygen atom forms a reactive ester and R 4 is oxygen or sulfur.

26 . The compound of claim 23 , wherein p is 1.

27 . The compound of claim 26 , wherein Y is alkylene containing from 1 to 10 carbon atoms,

wherein n and o are integers from 0 to 6, and m is an integer from 1 to 6.

28 . The compound of claim 27 , wherein X is

wherein R 3 is hydrogen or taken together with its attached oxygen atom forms a reactive ester and R 4 is oxygen or sulfur.

29 . An immunogen containing an immunogenic polyamine polymer linked to a compound of the formula:

Y is an organic spacing group;

X is a functional terminal group capable of binding to a polyamine polypeptide; and

p is an integer from 0 to 1.

30 . The compound of claim 29 , wherein p is 0.

31 . The compound of claim 29 , wherein p is 1.

32 . The compound of claim 31 , wherein Y is alkylene containing from 1 to 10 carbon atoms,

wherein n and o are integers from 0 to 6, and m is an integer from 1 to 6.

33 . The compound of claim 32 , wherein the immunogenic polymer contains one or more amino groups linked by

wherein R 4 is oxygen or sulfur.

34 . An immunogen comprising an immunogenic polyamine polymer linked to a compound of the formula:

Y is an organic spacing group;

X is a functional terminal group capable of binding to a polyamine polypeptide; and

p is an integer from 0 to 1.

35 . The compound of claim 34 , wherein p is 0.

36 . The compound of claim 35 , wherein the immunogenic polymer contains one or more amino groups linked by

wherein R 4 is oxygen or sulfur.

37 . The compound of claim 34 , wherein p is 1.

38 . The compound of claim 37 , wherein Y is alkylene containing from 1 to 10 carbon atoms,

wherein n and o are integers from 0 to 6, and m is an integer from 1 to 6.

39 . The compound of claim 38 , wherein the immunogenic polymer contains one or more amino groups linked by

wherein R 4 is oxygen or sulfur.
