Abstract

Disclosed herein is a method for measuring the contractility of intestinal tissue upon treatment with GLP-2 or a GLP-2 ligand. Also disclosed is an assay which directly measures the activity of GLP-2 or GLP-2 ligands ex vivo and permits the screening of putative GLP-2 ligands in native tissue.

Claims (23)

1. A method for determining whether a compound potentially modulates Glucagon-like peptide 2 (“GLP-2”) receptor activity comprising the step of measuring contractility of intestinal tissue segments ex vivo in the presence and absence of said compound, wherein a difference in contractility in the presence of said compound indicates potential GLP-2 receptor agonist or antagonist activity of said compound.

2. A method according to claim 1 wherein the intestinal tissue segments are of mammalian origin.

3. A method according to claim 2 wherein the intestinal tissue segments are of human origin.

4. A method according to claim 2 wherein the intestinal tissue segments are of rat origin.

5. A method according to claim 1 wherein the intestinal tissue segments are small intestine tissue segments.

6. A method according to claim 1 wherein the intestinal tissue segments are colon tissue segments.

7. A method for determining whether a compound is a potential GLP-2 receptor agonist comprising the step of measuring contractility of intestinal tissue segments ex vivo in the presence and absence of said compound, wherein inhibition of contractility in the presence of said compound indicates potential GLP-2 receptor agonist activity of said compound.

8. A method according to claim 7 wherein the intestinal tissue segments are rat colon tissue segments.

9. A method according to claim 7 wherein the intestinal tissue segments are human colon tissue segments.

A method for determining whether a compound is a potential GLP-2 receptor antagonist comprising the step of measuring the effect of a GLP-2 receptor agonist on contractility of intestinal tissue segments ex vivo in the presence and absence of said compound, wherein a decrease of agonist inhibition of contractility in the presence of said compound indicates potential GLP-2 receptor antagonist activity of said compound.

11. A method according to claim 10 wherein the GLP-2 receptor agonist is GLP-2.

12. A method according to claim 10 wherein the GLP-2 receptor agonist is a GLP-2 analogue.

13. method according to claim 12 wherein the GLP-2 analogue is teduglutide.

14. A method according to claim 10 wherein the intestinal tissue segments are colon tissue segments.

15. A method according to claim 14 wherein the intestinal tissue segments are rat colon tissue segments.

16. A method for determining whether a compound potentially modulates Glucagon-like peptide 2 (“GLP-2”) receptor activity comprising the steps of:

i. Obtaining a segment of intestinal tissue,

ii. Suspending the segment in an organ bath,

iii. Placing the segment under tension,

iv. Incubating the segment with said compound and

v. Measuring the contractility of the tissue; wherein an effect on contractility of the segment indicates potential modulation of GLP-2 receptor activity by said compound.

17. A method according to claim 16 wherein the intestinal tissue segments are human colon tissue segments.

18. A method according to claim 16 wherein the intestinal tissue segments are rat colon tissue segments.
