Abstract

Compositions suitable for oral administration to an animal comprising at least one immune stress-reducing enzyme in an amount effective to decrease the level of positive acute phase protein in an animal, increase the level of negative acute phase protein in an animal, and/or improve animal growth performance is provided, as are methods using such compositions. The compositions include animal feed compositions, liquid compositions other than animal feed, and solid compositions other than animal feed.

Claims (58)

1. A composition suitable for oral administration to an animal comprising an immune stress reducing enzyme in an orally acceptable carrier, wherein

said composition is selected from the group consisting of:

(i) an animal feed comprising at least 20 IU 1,3-β-glucanase/kg feed;

(ii) a liquid composition other than an animal feed comprising at least 155,000 IU 1,3-β-glucanase/L and

(iii) a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase/kg.

2. The composition of claim 1 , wherein the composition is a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase/kg.

3. The composition of claim 1 , wherein the composition is a solid composition other than an animal feed comprising at least 450,0000 IU 1,3-β-glucanase/kg.

4. The composition of claim 1 , wherein the composition is an animal feed that further comprises an ingredient that induces an immune response in the animal and wherein the ingredient is degraded by 1,3-β-glucanase.

5. The composition of claim 4 , wherein said ingredient is an antigen displayed by a pathogenic microorganism.

The composition of claim 1 , wherein the composition is selected from the group consisting of (i) an animal feed comprising at least 30 IU 1,3-β-glucanase/kg feed; (ii) a liquid composition other than an animal feed comprising at least 230,000 IU 11,3-β-glucanase/L and (iii) a solid composition other than an animal feed comprising at least 450,000 IU 1,3-β-glucanase/kg.

7. The composition of claim 1 , wherein the composition further comprises at least one additional immune stress reducing enzyme.

8. The composition of claim 7 , wherein the composition is a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase and at least one additional immune stress-reducing enzyme in an amount of at least 80,000 IU enzyme/kg.

9. The composition of claim 7 , wherein the composition is a solid composition other than an animal feed comprising at least 450,000 IU 1,3-β-glucanase and the at least one additional immune stress-reducing enzyme is present in an amount of at least 160,000 IU enzyme/kg.

10. The composition according to claim 7 , wherein the at least one additional immune stress reducing enzyme is selected from the group consisting of: (i) chitanase; (ii) xyloglucanase; (iii)-arabinanase; and (iv) 1,4-β-mannanase and arabinanase.

A composition suitable for oral administration to an animal comprising 1,4-β-mannanase and 1,3-β-glucanase, wherein said composition is selected from the group consisting of (i) an animal feed comprising 1,4-β-mannanase and at least 20 IU 1,3-β-glucanase/kg feed, (ii) a liquid composition other than an animal feed comprising 1,4-β-mannanase and at least 155,000 IU 1,3-β-glucanase/L and (iii) a solid composition other than an animal feed comprising 1,4-β-mannanase and at least 300,000 IU 1,3-β-glucanase/kg.

The composition of claim 11 , wherein the composition is selected from the group consisting of (i) an animal feed comprising 1,4-β-mannanase and at least 30 IU 1,3-β-glucanase/kg feed; (ii) a liquid composition other than an animal feed comprising 1,4-β-mannanase and at least 230,000 IU 1,3-β-glucanase/L and (iii) a solid composition other than an animal feed comprising 1,4-β-mannanase and at least 450,000 IU 1,3-β-glucanase/kg.

13. The composition of claim 11 , further comprising one or more additional immune stress reducing enzymes.

The composition of claim 1 , wherein the animal feed comprises the 1,3-β-glucanase in an amount selected from the group consisting of: at least 50,000 IU/ton of feed, at least 60,000 IU/ton of feed, at least 70,000 IU/ton of feed, at least 80,000 IU/ton of feed, at least 90,000 IU/ton of feed, at least 100,000 IU/ton of feed, at least 200,000 IU/ton of feed, and at least 500,000 IU/ton of feed.

15. The composition of claim 14 , wherein the amount of the 1,3-β-glucanase is at least 500,000 IU/ton of feed.

16. The composition of claim 1 , wherein the animal feed comprises the 1,3-β-glucanase in an amount selected from the group consisting of: at least 25 IU/kg feed, at least 30 IU/kg feed, at least 35 IU/kg feed, at least 40 IU/kg feed, at least 45 IU/kg feed, and at least 50 IU/kg feed.

The composition of claim 7 , wherein the animal feed comprises the at least one additional immune stress reducing enzyme in an amount selected from the group consisting of: at least 50,000 IU/ton of feed, at least 60,000 IU/ton of feed, at least 70,000 IU/ton of feed, at least 80,000 IU/ton of feed, at least 90,000 IU/ton of feed, at least 100,000 IU/ton of feed, at least 200,000 IU/ton of feed, and at least 500,000 IU/ton of feed.

18. The composition of claim 17 , wherein the amount of the at least one additional immune stress reducing enzyme is at least 500,000 IU/ton of feed.

The composition of claim 7 , wherein the animal feed comprises the at least one additional immune stress reducing enzyme in an amount selected from the group consisting of: at least 25 IU/kg feed, at least 30 IU/kg feed, at least 35 IU/kg feed, at least 40 IU/kg feed, at least 45 IU/kg feed, and at least 50 IU/kg feed.

The composition of claim 1 , wherein said composition further comprises as least one additional immune stress reducing enzyme selected from the group consisting of xyloglucanase, RNAse L, dsRNA specific adenosine deaminase, CG specific restriction endonuclease, α-1,6-mannosidase, α-1,2-mannosidase, α-1,3-mannosidase, endo-α-1,6-D-mannanase, α-mannanase, α-mannosidase, chitinase, phosphatidylserine decarboxylase, arabinanase, hyaluronidase and alkaline phosphatase.

The method of claim 7 , wherein the at least one additional immune stress reducing enzymes is selected from the group consisting of xyloglucanase, RNAse L, dsRNA specific adenosine deaminase, CG specific restriction endonuclease, α-1,6-mannosidase, α-1,2-mannosidase, α-1,3-mannosidase, endo-α-1,6-D-mannanase, α-mannanase, α-mannosidase, chitinase, phosphatidylserine decarboxylase, arabinanase, hyaluronidase and alkaline phosphatase.

22. A method of improving animal growth performance and/or reducing immune stress in an animal, comprising orally administering to said animal a composition comprising an immune stress reducing enzyme in an orally acceptable carrier, wherein

said composition is selected from the group consisting of:

(i) an animal feed comprising at least 20 IU 1,3-β-glucanase/kg feed;

(ii) a liquid composition other than an animal feed comprising at least 155,000 IU 1,3-β-glucanase/L and

(iii) a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase/kg.

23. The method of claim 22 , wherein said method further comprises administering an ingredient that induces an immune response in the animal, wherein said ingredient is degraded by 1,3-β-glucanase.

24. The method of claim 23 , wherein said ingredient and said 1,3-β-glucanase are administered in the same composition.

25. The method of claim 22 , wherein said composition is an animal feed.

26. The method of claim 23 , wherein said ingredient is an antigen displayed by a pathogenic microorganism.

27. The method of claim 22 , wherein said composition further comprises 1,4-β-mannanase.

28. The method of claim 22 , wherein the composition is a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase/kg.

29. The method of claim 22 , wherein the composition is a solid composition other than an animal feed comprising at least 450,000 IU 1,3-β-glucanase/kg.

30. The method of claim 22 , wherein the composition is selected from the group consisting of (i) an animal feed comprising at least 30 IU 1,3-β-glucanase/kg feed; (ii) a liquid composition other than an animal feed comprising at least 230,000 IU 1,3-β-glucanase/L and (iii) a solid composition other than an animal feed comprising at least 450,000 IU 1,3-β-glucanase/kg.

31. The method of claim 22 , wherein the composition further comprises at least one additional immune stress reducing enzyme.

32. The method of claim 31 , wherein the composition is a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase and the at least one additional immune stress-reducing enzyme is present in an amount of at least 80,000 IU enzyme/kg.

33. The method of claim 31 , wherein the composition is a solid composition other than an animal feed comprising at least 450,000 IU 1,3-β-glucanase and the at least one additional immune stress-reducing enzyme is present in an amount of at least 160,000 IU enzyme/kg.

34. The method of claim 31 , wherein the at least one additional immune stress reducing enzyme is selected from the group consisting of: (i) chitanase; (ii) xyloglucanase; (iii) arabinanase; and (iv) 1,4-β-mannanase and arabinanase.

A method of improving animal growth performance and/or reducing immune stress in an animal, comprising orally administering to said animal a composition comprising 1,4-β-mannanase and 1,3-β-glucanase in an orally acceptable carrier, wherein said composition is selected from the group consisting of: (i) an animal feed comprising 1,4-β-mannanase and at least 20 IU 1,3-β-glucanase/kg feed, (ii) a liquid composition other than an animal feed comprising 1,4-β-mannanase and at least 155,000 IU 1,3-β-glucanase/L and (iii) a solid composition other than an animal feed comprising 1,4-β-mannanase and at least 300,000 IU 1,3-β-glucanase/kg.

The method of claim 35 , wherein the composition is selected from the group consisting of (i) an animal feed comprising 1,4-β-mannanase and at least 30 IU 1,3-β-glucanase/kg feed; (ii) a liquid composition other than an animal feed comprising 1,4-β-mannanase and at least 230,000 IU 1,3-β-glucanase/L and (iii) a solid composition other than an animal feed comprising 1,4-β-mannanase and at least 450,000 IU 1,3-β-glucanase/kg.

37. The method of claim 35 , wherein the composition further comprises one or more additional immune stress reducing enzymes.

The method of claim 22 , wherein the animal feed comprises the 1,3-β-glucanase in an amount selected from the group consisting of: at least 50,000 IU/ton of feed, at least 60,000 IU/ton of feed, at least 70,000 IU/ton of feed, at least 80,000 IU/ton of feed, at least 90,000 IU/ton of feed, at least 100,000 IU/ton of feed, at least 200,000 IU/ton of feed, and at least 500,000 IU/ton of feed.

39. The method of claim 38 , wherein the amount of the 1,3-β-glucanase is at least 500,000 IU/ton of feed.

40. The method of claim 22 , wherein the animal feed comprises the 1,3-β-glucanase in an amount selected from the group consisting of: at least 25 IU/kg feed, at least 30 IU/kg feed, at least 35 IU/kg feed, at least 40 IU/kg feed, at least 45 IU/kg feed, and at least 50 IU/kg feed.

The method of claim 31 , wherein the animal feed comprises the at least one additional immune stress reducing enzyme in an amount selected from the group consisting of: at least 50,000 IU/ton of feed, at least 60,000 IU/ton of feed, at least 70,000 IU/ton of feed, at least 80,000 IU/ton of feed, at least 90,000 IU/ton of feed, at least 100,000 IU/ton of feed, at least 200,000 IU/ton of feed, and at least 500,000 IU/ton of feed.

42. The method of claim 41 , wherein the amount of the at least one additional immune stress reducing enzyme is at least 500,000 IU/ton of feed.

The method of claim 31 , wherein the animal feed comprises the at least one additional immune stress reducing enzyme in an amount selected from the group consisting of: at least 25 IU/kg feed, at least 30 IU/kg feed, at least 35 IU/kg feed, at least 40 IU/kg feed, at least 45 IU/kg feed, and at least 50 IU/kg feed.

44. A method of preventing or treating infection associated with a pathogenic microorganism that displays an antigen that is degraded by 1,3-β-glucanase, comprising orally administering to an animal in need thereof a composition comprising an immune stress reducing enzyme in an orally acceptable carrier, wherein said composition is selected from the group consisting of:

(i) an animal feed comprising at least 20 IU 1,3-β-glucanase/kg feed;

(ii) a liquid composition other than an animal feed comprising at least 155,000 IU 1,3-β-glucanase/L and

(iii) a solid composition other than an animal feed comprising at least 300,000 IU 1,3-β-glucanase/kg; and

wherein said antigen is degraded by 1-3-β-glucanase.

The method of claim 22 , wherein said composition further comprises an immune stress-reducing enzyme selected from the group consisting of xyloglucanase, RNAse L, dsRNA specific adenosine deaminase, CG specific restriction endonuclease, α-1,6-mannosidase, α-1,2-mannosidase, α-1,3-mannosidase, endo-α-1,6-D-mannanase, α-mannanase, α-mannosidase, chitinase, phosphatidylserine decarboxylase, arabinanase, hyaluronidase and alkaline phosphatase.

The method of claim 44 , wherein said composition further comprises an immune stress-reducing enzyme selected from the group consisting of xyloglucanase, RNAse L, dsRNA specific adenosine deaminase, CG specific restriction endonuclease, α-1,6-mannosidase, α-1,2-mannosidase, α-1,3-mannosidase, endo-α-1,6-D-mannanase, α-mannanase, α-mannosidase, chitinase, phosphatidylserine decarboxylase, arabinanase, hyaluronidase and alkaline phosphatase.
