Abstract

Fluid introduction is facilitated through the use of a port which extends entirely through a microfluidic substrate. Capillary forces can be used to retain the fluid within the port, and a series of samples or other fluids may be introduced through a single port by sequentially blowing the fluid out through the substrate and replacing the removed fluid with an alternate fluid, or by displacing the fluid in part with additional fluid. In another aspect, microfluidic substrates have channels which varying in cross-sectional dimension so that capillary action spreads a fluid only within a limited portion of the channel network. In yet another aspect, the introduction ports may include a multiplicity of very small channels leading from the port to a fluid channel, so as to filter out particles or other contaminants which might otherwise block the channel at the junction between the channel and the introduction port.

Claims (29)

1. A microfluidic system comprising:

a substrate having a first surface and a second surface; and

a plurality of through-hole ports extending through said substrate, each of said plurality of through-hole ports having a first open end at said first surface, a second open end at said second surface, said plurality of through-hole ports being sized to retain a fluid therein by capillary action; and

a mechanism to transport said fluid from said plurality of through-hole ports to outside said substrate, said mechanism comprises an upwardly extending member extending toward at least one of said plurality of though-hole ports and contacting said fluid to promote decanting of the fluid from said one of said plurality of through-hole ports.

2. The microfluidic system according to claim 1 wherein said upwardly extending member is a pin.

3. The microfluidic system according to claim 1 wherein said upwardly extending member comprises a hydrophilic material.

4. The microfluidic system according to claim 1 wherein each of said plurality of through-hole ports defines a total volume between said first surface and said second surface of less than 20 μL.

5. The microfluidic system according to claim 1 wherein each of said plurality of through-hole ports defines a total volume between said first surface and said second surface between about 0.5 μL and 10 μL.

6. The microfluidic system according to claim 1 wherein a first of said plurality of through-hole ports receives a first test fluid and a second of said plurality of through-hole ports receives a second test fluid, said second test fluid being different than said first test fluid.

7. A microfluidic system comprising:

a substrate having a first surface and a second surface;

a plurality of through-hole ports extending through said substrate, each of said plurality of through-hole ports having a first open end at said first surface, a second open end at said second surface, said plurality of through-hole ports being sized to retain a fluid therein;

a downwardly extending member transporting said fluid to said plurality of through-hole ports; and

a mechanism to transport said fluid from said plurality of through-hole ports to outside said substrate; said mechanism comprises an upwardly extending member extending toward at least one of said plurality of though-hole ports and contacting said fluid to promote decanting of the fluid from said one of said plurality of through-hole ports.

8. The microfluidic system according to claim 7 wherein said upwardly extending member is a pin.

9. The microfluidic system according to claim 7 wherein said upwardly extending member comprises a hydrophilic material.

10. The microfluidic system according to claim 7 wherein each of said plurality of through-hole ports defines a total volume between said first surface and said second surface of less than 20 μL.

11. The microfluidic system according to claim 7 wherein each of said plurality of through-hole ports defines a total volume between said first surface and said second surface between about 0.5 μL and 10 μL.

12. The microfluidic system according to claim 7 wherein a first of said plurality of through-hole ports receives a first test fluid and a second of said plurality of through-hole ports receives a second test fluid, said second test fluid being different than said first test fluid.

13. The microfluidic system according to claim 7 wherein said downwardly extending member is a pin.

14. A microfluidic system comprising:

a substrate having a first surface and a second surface; and

a plurality of through-hole ports extending through said substrate, each of said plurality of through-hole ports having a first open end at said first surface, a second open end at said second surface; and

a downwardly extending member transporting a fluid to said plurality of through-hole ports, said downwardly extending member being hydrophilic, wherein said downwardly extending member transports said fluid to said plurality of through-hole ports in response to capillary force when said downwardly extending member is brought into sufficient proximity to said plurality of ports.

15. The microfluidic system according to claim 14 wherein each of said plurality of through-hole ports defines a total volume between said first surface and said second surface of less than 20 μL.

16. The microfluidic system according to claim 14 wherein each of said plurality of through-hole ports defines a total volume between said first surface and said second surface between about 0.5 μL and 10 μL.

17. The microfluidic system according to claim 14 wherein a first of said plurality of through-hole ports receives a first test fluid and a second of said plurality of through-hole ports receives a second test fluid, said second test fluid being different than said first test fluid.

18. The microfluidic system according to claim 14 wherein said downwardly extending member is a pin.

19. The microfluidic system according to claim 14 wherein said downwardly extending member is an electrokinetically-actuated pin.
