Abstract

Efficient sequence specific gene silencing is possible through the use of siRNA technology. By selecting particular siRNAs by rationale design, one can maximize the generation of an effective gene silencing reagent, as well as methods for silencing genes.

Claims (24)

1. An siRNA molecule, wherein said siRNA molecule consists of: (a) a sense region and an antisense region, wherein said sense region and said antisense region together form a duplex region comprising 19-30 base pairs and said antisense region comprises a sequence that is the complement of SEQ. ID NO. 453; and (b) either no overhang regions or at least one overhang region, wherein each overhang region is six or fewer bases in length.

2. The siRNA molecule of claim 1 , wherein said antisense region and said sense region are each 19-25 nucleotides in length.

3. The siRNA molecule of claim 2 , wherein said antisense region and said sense region are each 19 nucleotides in length.

4. The siRNA molecule of claim 1 , wherein said antisense region is 100% complementary to a region of a target mRNA.

5. The siRNA molecule of claim 1 , wherein said siRNA molecule has at least one overhang region.

6. The siRNA molecule of claim 1 , wherein said siRNA molecule has no overhang regions.

7. The siRNA molecule of claim 2 , wherein said siRNA molecule has at least one overhang region.

8. The siRNA molecule of claim 2 , wherein said siRNA molecule has no overhang regions.

9. A chemically synthesized double stranded siRNA molecule, wherein:

a. each strand of said double stranded siRNA molecule is between 19 and 30 nucleotides in length; and

b. one strand of said double stranded siRNA molecule comprises a sequence that is the complement of SEQ. ID. NO. 453.

10. The chemically synthesized double stranded siRNA molecule of claim 9 , wherein each strand of said siRNA molecule is 19-25 nucleotides in length.

11. The chemically synthesized double stranded siRNA molecule of claim 10 , wherein each strand of said double stranded siRNA molecule is 19 nucleotides in length.

12. A pool of chemically synthesized double stranded siRNA molecules comprising a first chemically synthesized double stranded siRNA molecule and a second chemically synthesized double stranded siRNA molecule, wherein said first chemically synthesized double stranded siRNA molecule is the chemically synthesized double stranded siRNA molecule of claim 9 , and wherein in the second chemically synthesized double stranded siRNA molecule each strand is between 19 and 30 nucleotides in length and one strand of said second chemically synthesized double stranded siRNA molecule comprises a sequence that is the complement of SEQ. ID. NO. 440.

13. The pool of chemically synthesized double stranded siRNA molecules of claim 12 , wherein each strand of said first chemically synthesized double stranded siRNA molecule and said second chemically synthesized double stranded siRNA molecule is 19-25 nucleotides in length.

14. A pool of at least two siRNAs, wherein said pool comprises a first siRNA and a second siRNA, wherein

(a) said first siRNA consists of: (i) duplex region of 19-30 base pairs that has first antisense region that comprises a sequence that is the complement of SEQ. ID. NO. 453; and (ii) either no overhang regions or at least one overhang region, wherein each overhang region is six or fewer bases in length; and

(b) said second siRNA consists of: (i) a duplex region of 19-30 base pairs that has antisense region that comprises a sequence that is the complement of SEQ. ID. NO. 440; and (ii) either no overhang regions or at least one overhang region, wherein each overhang region is six or fewer bases in length.

15. The pool of claim 14 , wherein at least one of said first siRNA and said second siRNA has no overhang regions.

16. The pool of claim 14 , wherein said first siRNA and said second siRNA each have no overhang regions.

17. The pool of claim 15 , wherein said duplex region of said first siRNA and said duplex region of said second siRNA are each 19-25 base pairs in length.

18. The pool of claim 16 , wherein said duplex region of said first siRNA and said duplex region of said second siRNA are each 19-25 base pairs in length.

19. The pool of claim 17 , wherein said duplex region of said first siRNA and said duplex region of said second siRNA are each 19 base pairs in length.

20. The pool of claim 18 , wherein said duplex region of said first siRNA and said duplex region of said second siRNA are each 19 base pairs in length.
