Storage stable power compositions of interleukin-4 receptor
View Patent ↗The present invention provides storage stable dry powder compositions of IL-4R. The powder compositions demonstrate superior chemical and physical stability over their solution counterparts, particularly upon storage under varying conditions of temperature and humidity. Moreover, the powders, as prepared, possess good aerosol properties, which are maintained upon storage.
1. A method for preparing a dry powder composition of soluble Interleukin-4 receptor (IL-4R), said method comprising:
(a) preparing a mixture or a solution of soluble IL-4R and one or more excipients selected from the group consisting of carbohydrates, lipids, divalent metal cations, buffers, amino acids, oligopeptides, peptides, and proteins, in solvent, and
(b) spray-drying the mixture or solution to obtain a spray-dried powder comprising IL-4R; wherein the powder composition comprises particles having a mass median aerodynamic diameter (MMAD) of less than about 10 microns.
2. The method of claim 1 , wherein the excipient is a carbohydrate.
3. The method of claim 2 , wherein the carbohydrate is a sugar or sugar alcohol.
4. The method of claim 1 , wherein the excipient is an amino acid.
5. The method of claim 4 , wherein the amino acid is a hydrophobic amino acid.
6. The method of claim 1 , wherein the excipient is selected from the group consisting of citrate salts, leucine, raffinose, zinc salts, and combinations thereof.
7. The method of claim 1 , wherein the excipient is a buffer.
8. The method of claim 1 , wherein the excipient is a divalent metal cation.
9. The method of claim 1 , wherein the spray-dried powder comprises particles having a mass median aerodynamic diameter (MMAD) of less than about 5 microns.
10. The method of claim 1 , wherein the spray-dried powder comprises particles having a mass median aerodynamic diameter (MMAD) of less than about 3.5 microns.
11. The method of claim 1 , wherein the spray-dried powder comprises particles having a mass median diameter (MMAD) of between about 0.1 to 3 microns.
12. The method of claim 1 , wherein the spray-dried powder has a residual moisture content of less than about 10% by weight.
13. The method of claim 1 , wherein the spray-dried powder has a residual moisture content of less than about 5% by weight.
14. The method of claim 1 , wherein the spray-dried powder has a bulk density ranging from about 0.1-10 g/cc.
15. The method of claim 1 , wherein the mixture or solution of IL-4R contains IL-4R dissolved in a concentration from 0.01% (weight/volume) to 20% (weight/volume).
16. The method of claim 1 , wherein the solvent comprises a water-miscible solvent.
17. The method of claim 16 , wherein the water-miscible solvent is selected from the group consisting of acetone and alcohols.
