Fluid manipulation assembly
Microfluidic assemblies, systems, and methods are provided for manipulating fluid samples. Assemblies include an elastically deformable cover layer and a less elastically deformable substrate. The methods include deforming the substrate through the cover layer so that when the cover layer rebounds a new communication results in the assembly between the cover layer and the substrate and/or so that a new barrier wall is formed. Systems for carrying out the methods are also provided.
1 . A fluid manipulation assembly comprising:
a substrate;
a first recess formed in the substrate;
a second recess formed in the substrate;
an intermediate wall interposed between the first recess and the second recess, wherein the intermediate wall is formed from a deformable material having a first elasticity; and
an elastically deformable cover layer covering the first recess and having a second elasticity that is greater than the first of elasticity, wherein at least one of the first recess and the second recess comprises polymerase chain reaction reagants preloaded therein and the elastically deformable cover layer is in contact with the intermediate wall.
2 . The assembly of claim 1 , wherein the elastically deformable cover layer comprises a PCR tape material.
3 . The assembly of claim 1 , wherein the elastically deformable cover layer comprises an aluminum-containing PCR tape.
4 . The assembly of claim 1 , wherein the elastically deformable cover layer comprises a polyolefinic film.
5 . The assembly of claim 1 , wherein the elastically deformable cover layer comprises a silicone pressure sensitive adhesive layer.
6 . The assembly of claim 1 , wherein the polymerase chain reaction reagents comprise polymerase chain reaction primers.
7 . The assembly of claim 1 , wherein the assembly is disk-shaped.
8 . The assembly of claim 1 , wherein the substrate comprises a cyclo-olefin polymer.
9 . The assembly of claim 1 , wherein the substrate comprises polycarbonate.
10 . The assembly of claim 1 , wherein at least one of the first recess and the second recess has filtration media preloaded therein.
11 . A fluid manipulation assembly comprising:
a substrate;
a first recess formed in the substrate;
a second recess formed in the substrate;
an intermediate wall interposed between the first recess and the second recess, wherein the intermediate wall is formed from a deformable material having a first elasticity; and
an elastically deformable cover layer covering the first recess and having a second elasticity that is greater than the first of elasticity, wherein at least one of the first recess and the second recess comprises sequencing reaction reactants that enable at least one of a forward and a reverse sequencing reaction, preloaded therein, and the elastically deformable cover layer is in contact with the intermediate wall.
12 . The assembly of claim 11 , wherein the elastically deformable cover layer comprises a PCR tape material.
13 . The assembly of claim 11 , wherein the elastically deformable cover layer comprises an aluminum-containing PCR tape.
14 . The assembly of claim 11 , wherein the elastically deformable cover layer comprises a polyolefinic film.
15 . The assembly of claim 11 , wherein the elastically deformable cover layer comprises a silicone pressure sensitive adhesive layer.
16 . The assembly of claims 11 , wherein the sequencing reaction reactants comprise sequencing reaction primers.
17 . The assembly of claim 11 , wherein the assembly is disk-shaped.
18 . The assembly of claim 11 , wherein the substrate comprises a cyclo-olefin polymer.
19 . The assembly of claim 11 , wherein the substrate comprises polycarbonate.
20 . The assembly of claim 11 , wherein at least one of the first recess and the second recess has filtration media preloaded therein.
