Abstract

A human Kunitz-type inhibitor polypeptide with enhanced antifibrinolytic activity, methods of making, and methods of use. The novel polypeptide is structurally similar to the KD1 domain of human tissue factor pathway inhibitor-2 (TFPI-2).

Claims (45)

1. An isolated polypeptide comprising the primary structure of the wild-type KD 1 domain of human tissue factor pathway inhibitor-2 (TFPI-2) of SEQ ID NO: 2 or a biologically active subunit thereof, wherein the biological activity is serine protease inhibition, with the proviso that said polypeptide or a biologically active subunit thereof includes a lysine instead of arginine at position 24 and at least one of the following additional changes with respect to the wild-type KD 1 primary structure selected from the group consisting of:

an arginine instead of a leucine at position 26;

a threonine instead of a tyrosine at position 20;

a glutamate or a tyrosine instead of an aspartate at position 19; and

a glutamate or a lysine instead of a tyro sine at position 55;

wherein the amino acid numbering is as defined for the wild-type KD 1 amino acid sequence.

2. The isolated polypeptide of claim 1 , as set forth in SEQ ID NO:3 modified to include at least one of the following additional changes with respect to the wild-type KD 1 primary structure selected from the group consisting of:

an arginine instead of a leucine at position 26;

a threonine instead of a tyrosine at position 20;

a glutamate or a tyrosine instead of an aspartate at position 19; and

a glutamate or a lysine instead of a tyrosine at position 55;

wherein the amino acid numbering is as defined for the wild-type KD1 amino acid sequence.

3. The isolated polypeptide of claim 1 wherein the biologically active subunit of KD 1 is characterized by deletions of 1, 2, 3, 4, 5, 6, 7, 8, 9, or 10 amino acid residues at either or both of the amino-terminal or carboxy-terminal end relative to the wild-type KD 1 sequence.

The isolated polypeptide of claim 1 comprising a primary structure that is similar to the primary structure of the wild-type KD1 domain of TFPI-2 of SEQ ID NO: 2, or a biologically active subunit thereof, wherein the biological activity is serine protease inhibition, with the proviso that said polypeptide includes at least one conservative amino acid substitution relative to the primary structure of the wild-type KD1 domain of TFPI-2, and with the further proviso that said polypeptide includes a lysine instead of arginine at position 24 and at least one of the following additional changes with respect to the wild-type KD 1 primary structure selected from the group consisting of:

an arginine instead of a leucine at position 26;

a threonine instead of a tyrosine at position 20;

a glutamate or a tyrosine instead of an aspartate at position 19; and

a glutamate or a lysine instead of a tyrosine at position 55;

wherein the amino acid numbering is as defined for the wild-type KD1 amino acid sequence.

5. The isolated polypeptide of claim 1 wherein the polypeptide exhibits inhibitory activity against a serine protease that degrades fibrinogen.

6. An isolated polypeptide comprising the polypeptide of claim 1 ; and

a multiple positively charged amino acid sequence disposed at either or both of the N-terminal or C-terminal end of the polypeptide.

7. The isolated polypeptide of claim 6 wherein the multiple positively charged amino acid sequence comprises an amino acid sequence from the C-terminus of wild-type TFPI-2.

8. The isolated polypeptide of claim 7 wherein the multiple positively charged amino acid sequence comprises amino acids 192 through 211 of SEQ ID NO: 1.

9. A pharmaceutical composition comprising:

the isolated polypeptide of claim 1 ; and

a pharmaceutically acceptable carrier.

10. A method for treating a subject for a condition treatable by aprotinin, said method

comprising administering to the subject an effective amount of the polypeptide of claim 1 .

11. The method of claim 10 wherein the condition treatable by aprotinin is associated

with surgery or cardiovascular disease.

12. A method for treating a subject afflicted with allergies or asthma, said method

comprising administering to a subject an effective amount of the polypeptide of claim 1 .

13. A method for treating a subject afflicted with cancer or a precancerous condition,

said method comprising administering to a subject an effective amount of the polypeptide of claim 1 .

14. A method for treating a subject afflicted with an influenza infection, said method comprising administering to a subject an effective amount of the polypeptide of claim 1 .

15. A method for treating a subject in need of surgery comprising administering an effective amount of the polypeptide of claim 1 to the subject before, during and/or after surgery.

16. The method of claim 15 wherein the surgery is performed on a component of the cardiovascular system.

17. The method of claim 15 wherein the surgery is orthopedic surgery.

18. A method for treating a subject in need of inhibition of plasmin activity, said method comprising administering to a subject an effective amount of the polypeptide of claim 1 .

19. The method of claim 18 wherein the subject is undergoing bone remodeling.

20. The method of claim 18 wherein the subject is afflicted with hemophilia.

21. The method of claim 18 wherein the subject is afflicted with systemic inflammatory response syndrome (SIRS).

22. A method for inhibiting at least one activity of plasmin comprising contacting plasmin with an effective amount of the polypeptide of claim 1 .

23. The isolated polypeptide of claim 1 wherein a ligand is attached to the polypeptide.
