Abstract

The present invention provides uses of a polypeptide having a ligand activity to a sensory epithelium neuropeptide-like receptor (SENR) which is a G protein-coupled receptor protein, and a DNA encoding the same. More specifically, the present invention provides an anti-attention-deficit-disorder or anti-narcolepsy agent, which comprises a polypeptide having a ligand activity for SENR or a salt thereof, as well as a method for screening compounds having an anti-attention-deficit-disorder or anti-narcolepsy activity or compounds having an anti-anxiety, anti-depression, anti-insomnia, anti-schizophrenia or anti-fear activity or salts thereof, which comprises using the above polypeptide or a precursor protein of the polypeptide or a salt thereof.

A method for screening compounds that change the binding property between GPR14 (SENR) protein or a salt thereof, and an isolated polypeptide or precursor protein thereof containing the amino acid sequence of SEQ ID NO: 10 or a sequence comprising a deletion, a substitution, an addition, an insertion, or a combination thereof of SEQ ID NO: 10 having substantially the same binding property as the amino acid sequence of SEQ ID NO: 10, or an amide, ester or salt thereof, having an anti-anxiety activity, wherein said method comprises:

i) contacting said polypeptide or a precursor protein thereof, with GPR14 or a salt thereof;

ii) contacting said polypeptide or a precursor protein thereof, with GPR14 or a salt thereof in the presence of a test compound;

iii) detecting the binding of said polypeptide or a precursor protein thereof, with GPR14 or a salt thereof in the instance of i) and ii):

iv) comparing the results to identify compounds that change the binding property between GPR14 or a salt thereof, and the isolated polypeptide or precursor protein thereof containing the amino acid sequence of SEQ ID NO: 10, or an amide, ester or salt thereof; and

v) subjecting the compounds identified in the above step iv) to locomotion measurement to determine whether the identified compounds have an anti-anxiety, anti-attention deficit disorder, anti-narcolepsy, anti-depression, anti-insomnia, anti-schizophrenia, or anti-phobia effect.

30 . The method of claim 7 , wherein said polypeptide consists of the amino acid sequence of SEQ ID NO: 10.

The method of claim 7 , wherein said GPR14 (SENR) protein contains the amino acid sequence of SEQ ID NO: 3 or SEQ ID NO: 11 or a sequence comprising a deletion, a substitution, an addition, an insertion, or a combination thereof of SEQ ID NO: 3 or SEQ ID NO: 11 having substantially the same binding property as the amino acid sequence of SEQ ID NO: 3 or SEQ ID NO: 11, respectively.

32 . The method of claim 7 , wherein the locomotion measurement is conducted by measuring spontaneous locomotion and rearings.

A method for screening compounds that change the binding property between GPR14 (SENR) protein or a salt thereof, and an isolated polypeptide or precursor protein thereof containing the amino acid sequence of SEQ ID NO: 10, or an amide, ester, or salt thereof, having an anti-anxiety activity, wherein said method comprises:

i) contacting said polypeptide or a precursor protein thereof, with GPR14 or a salt thereof;

ii) contacting said polypeptide or a precursor protein thereof, with GPR14 or a salt thereof in the presence of a test compound;

iii) detecting the binding of said polypeptide or a precursor protein thereof, with GPR14 or a salt thereof in the instance of i) and ii);

iv) comparing the results to identify compounds that change the binding property between GPR14 or a salt thereof, and the isolated polypeptide or precursor protein thereof containing the amino acid sequence of SEQ ID NO: 10, or an amide, ester or salt thereof; and

v) subjecting the compounds identified in the above step iv) to locomotion measurement to determine whether the identified compounds have an anti-anxiety effect.
