Abstract

The invention provides an ocular iontophoresis device for delivering medication, the device comprising a medication reservoir suitable for being positioned on the eye, at least one medication in solution in the reservoir, an active electrode disposed in the reservoir, and a passive electrode, the device including at least one medication dissolved in non-saline water, the said solution having a pH lying in the range 6.5 to 8.5, the medication having a pKa lying in the range about 5.5 to about 9.5 and includes an active principle which is associated with an additive, such as a dendrimer, a polymer, a nanoparticle, a microsphere, a liposome, or an emulsion, and having an ionic form of valency greater than or equal to 1.

Claims (38)

1. An ocular iontophoresis device for transporting medication across a biological membrane of the eye, the device comprising:

a medication reservoir configured to store at least one medication in solution, the reservoir suitable for being positioned on the eye,

an active electrode disposed in the reservoir, the active electrode arranged so as to receive an electric field suitable for polarizing it to electrolyze when stored therein the at least one medication in a solution containing few or no ions competing with the at least one medication, and

a passive electrode also polarizable by the external electrical source,

the at least one medication being transported efficiently across the biological membrane of the eye through iontophoresis induced by the active and passive electrode.

An ocular iontophoresis device according to claim 1 , wherein the device includes at least one medication dissolved in non-saline water, the medication having a pH within the range of about 4.5 to about 11.5 and including an active principle, which is associated with an additive, and having an ionic form of valency greater than or equal to 1.

3. An ocular iontophoresis device according to claim 2 , wherein the additive is selected from the group consisting of: a dendrimer; a polymer; a nanoparticle; a microsphere; a liposome; and an emulsion.

4. An ocular iontophoresis device according to claim 2 , wherein the additive is a liposome having an aqueous core, the medication being contained in the aqueous core of the liposome.

5. An ocular iontophoresis device according to claim 2 , wherein the solution of at least one medication dissolved in non-saline water has a pH lying within the range of about 6.5 to about 8.5.

6. An ocular iontophoresis device according to claim 2 , wherein said medication(s) is/are of molecular weight less than about 150,000 Da.

7. An ocular iontophoresis device according to claim 1 , wherein the device includes at least one medication in an acid or basic form for administration to bond the active molecule to an additive presenting terminating ions.

8. An ocular iontophoresis device according to claim 7 , wherein the additive comprises nanoparticles.

9. An ocular iontophoresis device according to claim 7 , wherein the additive comprises microspheres.

10. An ocular iontophoresis device according to claim 1 , wherein the active electrode comprises:

an electrically conductive layer arranged so as to receive an electric field suitable for polarizing it sufficiently to electrolyze the medication(s) in solution, and

wherein the active electrode further comprises a protective layer situated between the conductive layer and the solution, said protective layer reacting significantly less than the conductive layer during electrolysis in a solution comprising at least one medication dissolved in non-saline water, the medication having a pH within the range of about 4.5 to about 11.5 and including an active principle, which is associated with an additive, and having an ionic form of valency greater than or equal to 1.

11. An ocular iontophoresis device according to claim 10 , wherein the active electrode is arranged in such a manner as to cover in projection at least a portion of the cornea.

12. An ocular iontophoresis device according to claim 10 , wherein the active electrode is arranged in such a manner as to cover in projection at least a portion of the cornea and at least a portion of the sclera.

13. An ocular iontophoresis device according to claim 10 , wherein the active electrode is arranged in such a manner as to cover in projection at least a portion of the sclera.

14. An ocular iontophoresis device according to claim 10 , wherein the active electrode is arranged, in operation, to present current density of less than 10 mA/cm 2 and to be polarized for 10 minutes.

15. An ocular iontophoresis device according to claim 10 , wherein the active electrode comprises one of the following:

electroplate forming the conductive layer and projected particles forming the protective layer;

successively deposited ink layers filled with an electrically conductive material to form the conductive layer, and an ink layer filled with a material that is less electrically conductive forming the protective layer;

a successively deposited solid film filled with an electrically conductive material forming the conductive layer, and a solid film filled with a less electrically conductive material forming the protective layer;

successively overmolded polymers respectively filled with an electrically conductive material forming the conductive layers, and polymers filled with a material that is electrically less conductive forming the protective layers; and

at least one wire of electrically conductive material (constituting said conductive layer) covered in a protective material that reacts substantially less than the conductive material during electrolysis in the solution, said covering of protective material then forming said protective layer.

16. An ocular iontophoresis device according to claim 10 , wherein the conductive layer is made of metal.

17. An ocular iontophoresis device according to claim 10 , wherein the protective layer covers the entire conductive layer.

18. An ocular iontophoresis device according to claim 10 , wherein the protective layer contains carbon or a semiconductor material.

An ocular iontophoresis device according to claim 18 , wherein the protective layer is: selected from the group consisting of: intrinsically-conductive polymers, such as polyacetylene; a polyaniline; a polypyrrole; a polyphenylene; a polythiophene a polymer filled with carbon black; a carbon fiber; a graphite; a diamond-like carbon (DLC), and combinations thereof.

20. An ocular iontophoresis device according to claim 2 , further comprising means for filling the reservoir with active substances.

21. An ocular iontophoresis device according to claim 2 , further comprising means for circulating the medication(s) in the reservoir.

22. A method of transporting medication across a biological membrane of the eye comprising:

positioning an iontophoresis device on the biological membrane of the eye, the device comprising a reservoir, an active electrode placed in the reservoir, and a passive electrode, the medication in solution having a pH within the range of about 4.5 to about 11.5 and including an active principle, which is associated with an additive and having an ionic form of valency greater than or equal to 1; and

polarizing the active electrode with respect to the passive electrode,

wherein polarization of the electrodes with respect to each other transports medication across the biological membrane of the eye through ocular iontophoresis.

23. The method of claim 22 , wherein the additive comprises nanoparticles.

24. The method of claim 22 , wherein the additive comprises microspheres.
