Abstract

A method and apparatus for the manipulation of colloidal particulates and biomolecules at the interface between an insulating electrode such as silicon oxide and an electrolyte solution. Light-controlled electrokinetic assembly of particles near surfaces relics on the combination of three functional elements: the AC electric field-induced assembly of planar aggregates; the patterning of the electrolyte/silicon oxide/silicon interface to exert spatial control over the assembly process; and the real-time control of the assembly process via external illumination. The present invention provides a set of fundamental operations enabling interactive control over the creation and placement of planar arrays of several types of particles and biomolecules and the manipulation of array shape and size. The present invention enables sample preparation and handling for diagnostic assays and biochemical analysis in an array format, and the functional integration of these operations. In addition, the present invention provides a procedure for the creation of material surfaces with desired properties and for the fabrication of surface-mounted optical components.

Claims (21)

1. A method of identifying targets capable of binding with bioactive agents, where the targets are contained in fluid samples, and wherein different types of bioactive agent are attached to differently-encoded microspheres, comprising:

a) providing a substrate having said microspheres randomly distributed thereon, wherein the substrate surface comprises a plurality of assay locations, each assay location comprising an array location, said array locations comprising a plurality of discrete sites such that the microspheres form planar crystalline arrays;

b) contacting the microspheres with the fluid samples; and

c) contacting said microspheres with a plurality of different decoder binding ligands, whereby the location of said bioactive agents is identified and the targets in the fluid samples which bound to said bioactive agents are identified.

2. A method according to claim 1 , wherein each of said assay locations comprises the same set of bioactive agents.

3. A method according to claim 1 , wherein said substrate is a microtiter plate and each assay location is a microtiter well.

4. A method according to claim 1 , further comprising binding an identifier binding ligand to a decoder binding ligand for identification of said bioactive agent.

5. The method according to claim 4 , wherein said identifier binding ligands are nucleic acids.

6. A method according to claim 5 , wherein said decoder binding ligands are nucleic acid and hybridize with said identifier binding ligands.

7. A method according to claim 1 , wherein at least one of said bioactive agents comprises a nucleic acid.

8. A method according to claim 7 , wherein all of said bioactive agents comprise nucleic acids.

9. A method according to claim 8 , wherein at least one of said nucleic acids is a nucleic acid analog.

10. The method according to claim 9 , whereby said decoder binding ligands are nucleic acids and hybridize with said bioactive agents.

11. A method according to claim 1 , wherein at least one of said bioactive agents comprises a protein.

12. A method according to claim 1 , wherein each of said assay locations comprises at least 1000 discrete sites per cm 2 .

13. A method according to claim 1 , wherein each of said assay locations comprises at least 1,000,000 sites/cm 2 .

14. A method according to claim 1 , wherein each of said assay locations comprises at least 100 discrete sites.

15. A method according to claim 14 , wherein each of said assay locations comprises at least 500 discrete sites.

16. A method according to claim 15 , wherein each of said assay locations comprises at least 10,000 discrete sites.

17. The method according to claim 1 , wherein contacting in step (c) comprises contacting said microspheres with a pool comprising said plurality of different decoder binding ligands.

18. The method according to claim 17 , wherein said decoder binding ligands are nucleic acids.
