Hybrid cells
A rapid, simple-to-use method for preparing hybrid cells, applicable to fully differentiated, non-dividing cells, entails bringing at least two different cells into contact under conditions that promote cell fusion and then purifying the resultant hybrid without antibiotic or metabolic selection. This approach yields hybrid cells useful in a variety of applications, including clinical treatment regimens, as cellular modulators of the immune system.

A hybrid cell preparation, comprising a hybrid cell prepared from a tumor cell from a first site and a first antigen presenting cell, and a hybrid cell prepared from a tumor cell from a second site and a second antigen presenting cell, wherein the resulting hybrid cell preparation is substantially free of non-hybrid cells.
2 . The hybrid cell preparation of claim 1 , further comprising hybrid cells prepared from tumor cells from at least one additional site.
3 . The preparation according of claim 1 , wherein the first and second antigen presenting cells are dendritic cells.
4 . A method of making a hybrid cell preparation, comprising:
(a) obtaining a tumor cell from a first site,
(b) obtaining a tumor cell from at least one additional site in the same patient;
(c) bringing a population of antigen presenting cells in contact with the tumor cells in (a) and (b) under conditions that promote cell fusion; and
(d) purifying the resulting hybrid cell without antibiotic or metabolic selection.
5 . The method of claim 4 , wherein the population of antigen presenting cells is a population of dendritic cells.
6 . A method of making the hybrid cell preparation of claim 1 , comprising:
(a) obtaining a tumor cell from a first site,
(b) bringing a first antigen presenting cell in contact with the tumor cell under conditions that promote cell fusion;
(c) purifying the hybrid cell resulting from (b) without antibiotic or metabolic selection;
(d) obtaining a tumor cell from at least one additional site in the same patient;
(e) bringing a second antigen presenting cell in contact with the tumor cell in (d) under conditions that promote cell fusion;
(f) purifying the hybrid cell resulting from (e) without antibiotic or metabolic selection;
(g) pooling the hybrid cells obtained in (c) and (f).
7 . The method of claim 6 , wherein the first and second antigen presenting cells are dendritic cells.
8 . A method of treating cancer in a subject, comprising administering to a patient a hybrid cell preparation that comprises (i) a hybrid cell prepared from a tumor cell from a first site and a first antigen presenting cell, and (ii) a hybrid cell prepared from a tumor cell from a second site and a second antigen presenting cell, wherein the hybrid cell preparation is substantially free of non-hybrid cells.
9 . The method of claim 8 , wherein the first and second antigen presenting cells are dendritic cells.
