Abstract

Specific oxygen uptake (OUR) is used as a process control parameter in fermentation processes. OUR is determined during at least the production phase of a fermentation process, and process parameters are adjusted to maintain the OUR within desired ranges. The invention is particularly applicable when the fermentation is conducted using a microorganism having a natural PDC pathway that has been disrupted so that it no longer functions. Microorganisms of this sort often produce poorly under strictly anaerobic conditions. Microaeration controlled by monitoring OUR allows the performance of the microorganism to be optimized.

Claims (7)

1. A method of conducting a fermentation process in a fermentation medium comprising a genetically engineered yeast having a disruption of a native PDC pathway, a substrate that is fermentable by the genetically engineered yeast cell, wherein the fermentation broth has a quantity of dissolved oxygen (DO) and the fermentation exhibits a specific oxygen uptake (OUR), comprising

a) measuring the OUR during a production phase of a fermentation;

b) adjusting aeration conditions such that the OUR is maintained in a range of from about 0.8 to about 3.0 mmol O 2 /gdw/h while maintaining the DO below 10 μmol O 2 /L during the production phase of the fermentation.

2. The process of claim 1 , wherein the genetically engineered yeast cell exhibits a Crabtree negative phenotype.

3. The process of claim 2 , wherein the yeast cell is of the genera Kluyveromyces or Candida.

4. The process of claim 3 , wherein the yeast cell has at least one functional exogenous gene that enables the cell to produce a desired fermentation product.

5. The process of claim 4 , wherein the exogenous gene is a lactate dehydrogenase gene.
