Abstract

The present invention is directed to analogs of paromomycin having a variety of chemical functional groups attached at the 2″-O-position as well as their preparation and use as prophylactic or therapeutics against microbial infection.

Claims (35)

1. A compound of formula I:

wherein:

each R 1 is, independently, H or a hydroxyl protecting group;

each R 2 and R 3 is, independently, H, an amino protecting group or together R 2 and R 3 that are connected to the same nitrogen atom form a cyclic protecting group that can include additional heteroatoms selected from N, O and S;

Z has the formula:

—(CH 2 ) m -L 1 -(CH 2 ) mm -Q 1

wherein:

L 1 is S, O or NJ 1 ;

m is from 1 to about 8;

mm is 0 or from 1 to about 8;

Q 1 is H, OH, halogen, NJ 1 J 2 , C 2 -C 20 alkenyl, substituted C 2 -C 20 alkenyl, C 2 -C 20 alkynyl, substituted C 2 -C 20 alkynyl, C 5 -C 20 aryl, substituted C 5 -C 20 aryl, heteroaryl, substituted heteroaryl, a heterocyclic radical, a substituted heterocyclic radical or a substituted or unsubstituted mono or poly cyclic structure that can be unsaturated, partially saturated or fully saturated and can include one or more heteroatoms selected from O, N and S;

each of said substituted groups, is, independently, mono or poly substituted with substituent groups selected from halogen, hydroxyl, C 1 -C 20 alkyl, substituted C 1 -C 20 alkyl, C 2-20 alkenyl, substituted C 2 -C 20 alkenyl, C 2 -C 20 alkynyl, substituted C 2 -C 20 alkynyl, C 5 -C 20 aryl or substituted C 5 -C 20 aryl, heteroaryl or a substituted heteroaryl, haloalkyl, alkoxy, thioalkoxy, haloalkoxy, —NJ 1 J 2 , azido, carboxy, acyl, ═O, cyano, nitro, mercapto, sulfide, sulfonyl and sulfoxide and wherein said substituent groups are optionally protected; and

J 1 and J 2 are each, independently, H, C 1 -C 20 alkyl, substituted C 1 -C 20 alkyl, C 1 -C 20 aminoalkyl or an amino protecting group or J 1 and J 2 are joined in a heterocyclic compound including the nitrogen atom connecting them and optionally including from 1 to 2 additional heteroatoms selected from O, N, and S.

2. The compound of claim 1 wherein each R 1 is a hydroxyl protecting group.

3. The compound of claim 1 wherein each R 1 is H.

4. The compound of claim 1 wherein each R 2 is an amino protecting group and each R 3 is H, or optionally R 2 and R 3 that are connected to the same nitrogen atom form a cyclic protecting group that may include additional heteroatoms selected from N, O and S.

5. The compound of claim 1 wherein each R 1 , R 2 and R 3 is H.

6. The compound of claim 1 wherein m is from 2 to 8.

7. The compound of claim 1 wherein the sum of m and mm is from 3 to 8.

8. The compound of claim 1 wherein m is 2 and L 1 is NJ 1 .

9. The compound of claim 8 wherein mm is from 1 to 4.

10. The compound of claim 9 wherein Q 1 is NJ 1 J 2 or a substituted or unsubstituted mono or poly cyclic structure that can be unsaturated, partially saturated or fully saturated and can include one or more heteroatoms selected from O, N and S.

11. The compound of claim 10 wherein said substituted or unsubstituted mono or poly cyclic structure comprises one or more fused or linked rings wherein each ring is, independently, alicyclic, heterocyclic, aryl or heteroaryl.

The compound of claim 1 wherein Q 1 is phenyl, biphenyl, benzoyl, adamanthanyl, a steroidyl group, 1,8-naphthalenedicarboximide, pyridinyl, piperidinyl, piperazinyl, benzimidazolyl, imidazolyl, pyrrolidinyl, pyrazolyl, indolyl, 1H-indazolyl, α-carbolinyl, carbazolyl, phenothiazinyl, phenoxazinyl, quinolinyl, tetrazolyl, triazolyl, and morpholinyl.

13. The compound of claim 1 wherein m is from 1 to 5.

14. The compound of claim 1 wherein mm is 0.

15. The compound of claim 14 wherein Q 1 is a substituted or unsubstituted mono or poly cyclic structure that can be unsaturated, partially saturated or fully saturated and can include one or more heteroatoms selected from O, N and S.

16. The compound of claim 15 wherein said substituted or unsubstituted mono or poly cyclic structure comprises one or more fused or linked rings wherein each ring is, independently, alicyclic, heterocyclic, aryl or heteroaryl.

17. The compound of claim 16 wherein said mono or poly cyclic structure is a heterocycle radical, aryl or a heteroaryl group.

18. The compound of claim 1 wherein m is 2.

19. The compound of claim 8 wherein J 1 is H.

20. The compound of claim 1 wherein m is 2, L 1 is NJ 1 and mm is from 1 to 4.

21. The compound of claim 1 having the configuration:

22. The compound of claim 1 for use in therapy.

23. A method of treating a bacterial infection in a mammal comprising administering to the mammal an effective amount of a compound of claim 1 .
