Abstract

Methods of evaluating molecular isomers of branched-chain amino acids are featured. The methods can include: derivatizing one or more molecular isomers of branched-chain amino acids in a sample comprising a branched-chain amino acid labeled with one or more heavy atoms as a first standard; adding, to the sample, after derivatization, a nonderivatized or derivatized branched chain amino acid that is labeled with one or more heavy atoms, as a second standard; evaluating the sample using tandem mass spectrometry; and detecting peaks indicative of derivatized and nonderivatized forms of one or more branched-chain amino acids in the sample.

Claims (34)

1. A method of detecting one or more molecular isomers of one or more branched-chain amino acids in a sample, the method comprising:

derivatizing one or more molecular isomers of one or more branched-chain amino acids in a sample, wherein the sample comprises one or more molecular isomers of one or more branched-chain amino acids and a first standard, the first standard being a branched-chain amino acid labeled with one or more heavy atoms;

adding to the sample, after derivatization, a nonderivatized or derivatized branched-chain amino acid that is labeled with one or more heavy atoms as a second standard;

evaluating the sample using tandem mass spectrometry; and

detecting peaks indicative of derivatized and nonderivatized forms of one or more branched-chain amino acids in the sample, thereby distinguishing between branched-chain amino acids that are molecular isomers in the sample.

The method of claim 1 , wherein the second standard is a derivatized branched chain amino acid that is labeled with one or more heavy atoms, and wherein after the derivatization step, a derivatized form of the first standard and second standard have different molecular weights as a result of different heavy atom labeling.

The method of claim 1 , wherein the second standard is a nonderivatized branched chain amino acid that is labeled with one or more heavy atoms, and wherein the first standard prior to the derivatization step and the second standard have different molecular weights as a result of different heavy atom labeling.

4. The method of claim 1 , further comprising determining a derivatization efficiency of a branched-chain amino acid by reference to the first standard and the second standard.

5. The method of claim 4 , further comprising determining the concentration of a branched chain amino acid in the sample using the derivatization efficiency.

6. The method of claim 1 , wherein the one or more isomers of the one or more branched-chain amino acids include one or more of leucine, isoleucine, and allo-isoleucine.

7. The method of claim 6 , wherein the one or more molecular isomers of the one or more branched-chain amino acids include isoleucine and/or allo-isoleucine.

8. The method of claim 1 , wherein the derivatizing comprises esterification.

9. The method of claim 8 , wherein the esterification is carried out with an alcohol.

10. The method of claim 9 , wherein the alcohol is selected from the group consisting of butanol, isopropyl alcohol, ethanol, and methanol.

11. The method of claim 1 , wherein less than 100% of branched-chain amino acids are derivatized.

12. The method of claim 1 , wherein the first standard is selected from the group consisting of labeled leucine, isoleucine, and allo-isoleucine.

13. The method of claim 1 , wherein the second standard is selected from the group consisting of labeled leucine, isoleucine, and allo-isoleucine.

14. The method of claim 1 , wherein the detecting comprises acquiring data signals representing butylated and unbutylated forms of the one or more branched-chain amino acids.

15. A method of diagnosing a metabolic disorder, the method comprising:

treating a sample from a subject under conditions in which one or more branched-chain amino acids in the sample are derivatized, wherein the sample comprises one or more molecular isomers of one or more branched-chain amino acids and a first standard, the first standard being a branched-chain amino acid labeled with one or more heavy atoms;

adding to the sample, after derivatization, a nonderivatized or derivatized branched-chain amino acid that is labeled with one or more heavy atoms as a second standard;

evaluating the sample using tandem mass spectrometry; and

detecting signals indicative of derivatized and nonderivatized forms of one or more branched-chain amino acids in the sample, thereby distinguishing between branched-chain amino acids that are molecular isomers in the sample,

wherein an altered level of the one or more branched-chain amino acids in the sample relative to the level in one or more reference samples is an indication that the subject has, or is at risk of developing, a metabolic disorder characterized by altered levels of one or both of branched-chain amino acids and branched-chain ketoacids.

16. The method of claim 15 , wherein the metabolic disorder is Maple Syrup Urine Disorder (MSUD).

17. The method of claim 15 , wherein the subject is a human.

18. The method of claim 15 , wherein the subject is a newborn human.

19. The method of claim 15 , further comprising determining a derivatization efficiency of a branched-chain amino acid by reference to the first standard and the second standard.

20. The method of claim 19 , further comprising determining the concentration of one or more branched chain amino acids in the sample using the derivatization efficiency.

21. The method of claim 1 , wherein the one or more isomers of the one or more branched-chain amino acids include one or more of leucine, isoleucine, and allo-isoleucine.

22. The method of claim 21 , wherein the molecular isomers of the one or more branched-chain amino acids include isoleucine and/or allo-isoleucine.

23. The method of claim 15 , wherein the conditions in which one or more branched-chain amino acids in the sample are derivatized comprises esterification.

The method of claim 15 , wherein the second standard is a derivatized branched chain amino acid that is labeled with one or more heavy atoms, and wherein after the derivatization step, a derivatized form of the first standard and the second standard have different molecular weights as a result of different heavy atom labeling.

25. The method of claim 15 , wherein the second standard is a nonderivatized branched chain amino acid that is labeled with one or more heavy atoms, and wherein the first standard prior to the derivatization step and the second standard have different molecular weights as a result of different heavy atom labeling.
