Abstract

Nucleic acid molecules which encode a branching enzyme from a bacterium of the genus Neisseria , vectors, host cell, plant cells and plants containing said nucleic acid molecules as well as starch obtainable from the plants described are described. Furthermore, an in-vitro method for producing α-1,6-branched α-1,4-glucans on the basis of sucrose and a combination of enzymes of an amylosucrase and a branching enzyme as well as the α-1,6-branched α-1,4-glucans obtainable by said method are described.

Claims (21)

1. A method for preparing starch, comprising the step of extracting starch from

(1) a transgenic plant comprising plant cells containing a nucleic acid molecule linked to regulatory sequences guaranteeing the transcription in plant cells said nucleic acid molecule encoding a branching enzyme selected from the group consisting of

(a) a nucleic acid molecule encoding a protein which comprises the amino acid sequence depicted in SEQ ID NO. 2;

(b) a nucleic acid molecule comprising the coding region depicted in SEQ ID NO. 1;

(c) a nucleic acid molecule encoding a protein which comprises the amino acid sequence encoded by the insert in the plasmid deposited with the Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) with the accession number of DSM 12425; and

(d) a nucleic acid molecule comprising the coding region for a branching enzyme, which is contained in the insert of the plasmid deposited with the Deutsche Sammlung von Mikrooranismen und Zelkelturen (DSMZ) with the accession number of DSM 12425; and

(e) a nucleic acid molecule encoding a sequence having more than 95% identity with SEQ ID NO:2; and

(f) a nucleic acid molecule the sequence of which deviates from the sequence of a nucleic acid molecule of (e) due to the degeneracy of the genetic code;

or

(2) from transgenic plants obtained by a method comprising

(a) genetically modifying a plant cell by introducing the nucleic acid molecule according to subsection 1 (a)-(f) or a vector comprising said nucleic acid or a vector comprising said nucleic acid linked in the sense orientation to regulatory sequences guaranteeing transcription in prokaryotic or eukaryotic cells wherein the nucleic acid molecule is expressed;

(b) regenerating a plant from the cell produced according to step (a); and

(c) optionally propagating further plants produced from the plant produced according to step (b),

or

(3) from starch-storing parts of a plant according to (1) or (2) or from plant cells containing a nucleic acid molecule linked either to regulatory sequences guaranteeing the transcription in plant cells or to a sequence encoding a signal sequence which guarantees the localisation of the encoded protein in the plastids of the cells, said nucleic acid molecule encoding a branching enzyme selected from the group consisting of

(a) a nucleic acid molecule encoding a protein which comprises the amino acid sequence depicted in SEQ ID NO. 2;

(b) a nucleic acid molecule comprising the coding region depicted in SEQ ID NO. 1;

(c) a nucleic acid molecule encoding a protein which comprises the amino acid sequence encoded by the insert in the plasmid deposited with the Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) with the accession number of DSM 12425; and

(d) a nucleic acid molecule comprising the coding region for a branching enzyme, which is contained in the insert of the plasmid deposited with the Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) with the accession number of DSM 12425; and

(e) a nucleic acid encoding a sequence having more than 95% identity with SEQ ID NO:2; and

(f) a nucleic acid molecule the sequence of which deviates from the sequence of a nucleic acid molecule of (e) due to the degeneracy of the genetic code wherein the nucleic acid molecule is expressed.
