Inhibitors of Akt activity
View Patent ↗The instant invention provides for compounds that inhibit Akt activity. In particular, the compounds disclosed selectively inhibit one or two of the Akt isoforms. The invention also provides for compositions comprising such inhibitory compounds and methods of inhibiting Akt activity by administering the compound to a patient in need of treatment of cancer.
1. A compound of the Formula E:
wherein:
a is 0 or 1; b is 0 or 1; n is 0, 1 or 2: p is 0, 1 or 2;
Ring K is selected from:
Q is selected from: 2-azepinone, benzimidazolyl, benzimidazolonyl, 2-diazapinone, imidazolyl, 2-imidazolidinone, indolyl, isoquinolinyl, morpholinyl, piperidyl, piperazinyl, pyridyl, pyrrolidinyl, 2-piperidinone, 2-pyrimidinone, 2-pyrollidinone, quinolinyl, tetrazolyl, tetrahydrofuryl, tetrahydroisoquinolinyl, and thienyl, optionally substituted with one to three substituents selected from R 6b ;
R 1 is selected from: oxo, (C═O) a O b (C 1 -C 10 )alkyl, CO 2 H, halo, OH, CN, (C 1 -C 6 )alkoxy, O(C═O)(C 1 -C 6 )alkyl and N(R b ) 2 ;
R 2 is selected from: oxo, (C═O) a O b (C 1 -C 10 )alkyl, CO 2 H, halo, OH, CN, (C 1 -C 6 )alkoxy, O(C═O)(C 1 -C 6 )alkyl,(C 2 -C 10 )alkenyl and N(R b ) 2 , said alkenyl optionally substituted with oxo;
R 6b is selected from:
optionally substituted with 1 to 3 substituents selected from oxo, OH, N(R b ) 2 , halogen and O a (C 1 -C 6 )alkyl; and
R b is independently selected from H and (C 1 -C 6 )alkyl;
or a pharmaceutically acceptable salt or a stereoisomer thereof.
2. A compound of the Formula F:
wherein:
a is 0 or 1; b is 0 or 1; n is 0, 1 or 2; p is 0, 1 or 2;
Ring K is selected from:
R 1 is selected from: oxo, (C═O) a O b (C 1 -C 10 )alkyl, CO 2 H, halo, OH, CN, (C 1 -C 6 )alkoxy, O(C═O)(C 1 -C 6 )alkyl and N(R b ) 2 ;
R 2 is selected from: oxo, (C═O) a O b (C 1 -C 10 )alkyl, CO 2 H, halo, OH, CN, (C 1 -C 6 )alkoxy, O(C═O)(C 1 -C 6 )alkyl, (C 2 -C 10 )alkenyl and N(R b ) 2 , said alkenyl optionally substituted with oxo;
R 3 and R 4 are independently: H and (C 1 -C 6 )alkyl; and
R b is independently selected from H and (C 1 -C 6 )alkyl;
or a pharmaceutically acceptable salt or a stereoisomer thereof.
3. A compound of the Formula G:
wherein:
a is 0 or 1; b is 0 or 1; p is 0, 1 or 2;
Ring K is selected from:
R 2 is selected from: oxo, (C═O) a O b (C 1 -C 10 )alkyl, CO 2 H, halo, OH, CN, (C 1 -C 6 )alkoxy, O(C═O)(C 1 -C 6 )alkyl, (C 2 -C 10 )alkenyl and N(R b ) 2 , said alkenyl optionally substituted with oxo; and
R 3 and R 4 are independently: H and (C 1 -C 6 )alkyl, wherein said alkyl is optionally substituted with up to three substituents selected from: OH and halo; and wherein R 3 and 4 may be joined to form a (C 3 -C 7 )cycloalkyl;
or a pharmaceutically acceptable salt or a stereoisomer thereof.
4. A compound which is selected from:
5-Methoxy-2(4-{[4-5-pyridin-2-yl-4H-1,2,4-triazol)piperidin- 1-yl]methyl}phenyl)-3-(2-thienyl)-1,6-naphthyridine;
2-(4-{[4-(5-Pyridin-2-yl-4H-1,2,4-4triazol-3-yl)piperidin-1-yl]methyl}phenyl)-3-(2-thienyl)-1,6-naphthyridin-5(6H)-one;
1-{4-[4-(3-pyrimidin-5-ylquinoxalin-2-yl)benzyl]cyclohexyl}-1,3-dihydro-2H-benimidazol-2-one;
3-[3-(4-{[4-2-oxo-2,3-dihydro-1H-benimidazol-1-yl)cyclohexyl]methyl}phenyl)quinoxalin-2-yl]thiophene-2-carbaldehyde;
1-(4- {4-[3(H-pyrazol-5-yl)quinoxalin-2-yl]benzyl}cyclohexyl)-1,3-dihydro-2H-benzimidazol-2-one; and
2-[4-(1-amino-I-methylethyl)phenyl]-3-(2-thienyl)-1,6-naphthyridin-5(6H)-one;
or a pharmaceutically acceptable salt or stereoisomer thereof.
5. A compound according to claim 1 of the Formula E, wherein;

which are optionally substituted with one substituent selected from R 6b ;
or a pharmaceutically acceptable salt or a storeoisomer thereof.
