Abstract

The present invention concerns methods and compositions for isolating, enriching, and/or labeling miRNA molecules and for preparing and using arrays or other detection techniques for miRNA analysis. Moreover, the present invention concerns methods and compositions for generating miRNA profiles and employing such profiles for therapeutic, diagnostic, and prognostic applications.

Claims (28)

148 . An miRNA array comprising at least five miRNA probes immobilized on a solid support, wherein the probes comprise:

a) at least 10 contiguous nucleic acids of an miRNA coding sequence or its compliment; and

b) a 5′ or 3′ amine group.

149 . The miRNA array of claim 148 , comprising between 5 and 5000 different miRNA probes.

150 . The miRNA array of claim 149 , comprising between 50 and 3000 different miRNA probes.

151 . The miRNA array of claim 150 , comprising between 100 and 2000 different miRNA probes.

152 . The miRNA array of claim 151 , comprising between 500 and 1000 different miRNA probes.

153 . The miRNA array of claim 149 , wherein the miRNA array comprises miRNA probes having human miRNA coding sequences.

154 . The miRNA array of claim 148 , wherein the miRNA array comprises miRNA probes having a 3′ amine.

155 . The miRNA array of claim 154 , wherein the miRNA array comprises miRNA probes having miRNA coding sequence that is between 19-34 nucleotides in length.

156 . The miRNA array of claim 148 , wherein the miRNA array comprises miRNA probes having miRNA coding sequence that includes the entire processed miRNA sequence.

157 . The miRNA array of claim 156 , wherein the miRNA coding sequence also includes at least 2-5 nucleotides of coding sequence upstream and/or downstream of the processed miRNA sequence.

158 . The miRNA array of claim 157 , wherein the miRNA coding sequence includes 4 nucleotides of coding sequence upstream and/or downstream of the processed miRNA sequence.

159 . The miRNA array of claim 158 , wherein the miRNA coding sequence includes 4 nucleotides of coding sequence upstream and downstream of the processed miRNA sequence.

160 . The miRNA array of claim 148 , wherein the miRNA array comprises miRNA probes further comprising:

c) at least a first linker sequence flanking the miRNA coding sequence.

161 . The miRNA array of claim 160 , wherein the first linker sequence is between 3 to 25 nucleotides in length.

162 . The miRNA array of claim 161 , wherein the first linker sequence is at the 3′ end of the miRNA coding sequence.

163 . The miRNA array of claim 148 , wherein the solid support is glass, plastic, or metal.

164 . An miRNA array comprising at least five miRNA probes immobilized on a solid support, wherein the probes comprise:

a) at least 10 contiguous nucleic acids of an miRNA coding sequence or its compliment; and

b) a linker sequence positioned at the 5′ and/or 3′ end of the probe.

165 . The miRNA array of claim 164 , wherein the linker is between 3 to 25 nucleotides in length.

166 . The miRNA array of claim 165 , wherein the linker is between 15 to 21 nucleotides in length.

167 . The miRNA array or claim 166 , wherein the linker is 21 nucleotides in length.

168 . The miRNA array of claim 164 , wherein the linker is positioned at the 3′ end of the probe.

169 . The miRNA array of claim 148 , further comprising an amine group at the 3′ and/or 5′ end of the probe.
