Immediate release and sustained release ibuprofen dosing regimen
View Patent ↗A method of administering non-steroidal-anti-inflammatory drugs, in particular propionic acid derivatives such as ibuprofen, is provided. The inventive dosage forms provide an initial release of the NSAID and a second sustained release of NSAID, preferably in a single dosing step.
1. A dosage form capable of providing relief of pain over a 12 hour period, comprising an immediate release portion comprising about 200 mg to about 400 mg of an immediate release dose of ibuprofen, wherein said immediate release dose is released at the beginning of said 12 hour period, and a sustained release portion comprising about 100 mg to about 200 mg of a sustained release dose of said ibuprofen, wherein said sustained release dose is released about 2 hours to about 10 hours after release of said immediate release dose, and wherein said immediate release portion contains a dose that is at least about twice the dose in the sustained release portion.
2. The dosage form of claim 1 , wherein said dosage form is a solid dosage form.
3. The dosage form of claim 1 , wherein the sustained release dose is zero-order.
4. The dosage form of claim 1 , wherein the sustained release dose is first-order.
5. A method of treating pain, comprising administering to a mammal in need thereof the dosage form of claim 1 , wherein said ibuprofen has a duration of therapeutic effect which lasts about 6 hours after administration of said sustained release dose.
6. The method of claim 5 , wherein the sustained release dose is zero-order.
7. The method of claim 5 , wherein the sustained release dose is first-order.
8. The method of claim 5 , wherein said immediate release dose is ibuprofen and provides about 5.7 mg/kg to about 12 mg/kg, and said sustained release dose of ibuprofen is about 2.5 mg/kg to about 4 mg/kg.
9. The method of claim 5 , wherein said immediate release dose is ibuprofen and provides about 5.7 mg/kg, and the sustained release dose of ibuprofen is about 2.86 mg/kg.
