GIP analog and hybrid polypeptides with selectable properties
View Patent ↗The present invention relates generally to novel GIP analogs and GIP hybrid polypeptides with selectable properties, useful as agents for the treatment and prevention of metabolic diseases and disorders, for example those which can be alleviated by control plasma glucose levels, insulin levels, and/or insulin secretion, positive inotropic effects, reduction of catabolic effects, slowing of gastric emptying. Such conditions and disorders include, but are not limited to, hypertension, dyslipidemia, cardiovascular disease, eating disorders, critical care, insulin-resistance, obesity, and diabetes mellitus of any kind, including type 1, type 2, and gestational diabetes.
1. A gastric inhibitory peptide (GIP) compound that is a GIP hybrid polypeptide exhibiting at least two hormonal activities, said polypeptide comprising a first bio-active GIP peptide hormone modified for resistance to DPP-IV that exhibits at least one hormonal activity of the human GIP and comprises at least 28 amino acids and has at least 85% sequence identity to the human GIP 1-30 covalently linked to at least one additional bio-active peptide hormone that exhibits at least one hormonal activity of its parent peptide hormone with the proviso that serum albumin is not present, and wherein the GIP compound comprises the sequence of SEQ. ID NO: 481.
2. A gastric inhibitory peptide (GIP) compound that is a GIP hybrid polypeptide exhibiting at least two hormonal activities, said polypeptide comprising a first bio-active GIP peptide hormone modified for resistance to DPP-IV that exhibits at least one hormonal activity of the human GIP and comprises at least 28 amino acids and has at least 85% sequence identity to the human GIP (1-30) covalently linked to at least one additional bio-active peptide hormone that exhibits at least one hormonal activity of its parent peptide hormone with the proviso that serum albumin is not present, wherein the GIP compound comprises the sequence of SEQ. ID. NO: 481 or the pegylated derivative thereof.
3. A gastric inhibitory peptide (GIP) compound that is a GIP hybrid polypeptide exhibiting at least two hormonal activities, said polypeptide comprising a first bio-active GIP peptide hormone modified for resistance to DPP-IV that exhibits at least one hormonal activity of the human GIP and comprises at least 28 amino acids and has at least 85% sequence identity to the human GIP (1-30) covalently linked to at least one additional bio-active peptide hormone that exhibits at least one hormonal activity of its parent peptide hormone with the proviso that serum albumin is not present, wherein the GIP compound comprises the sequence of an analog of SEQ. ID. NO: 481 wherein the analog of sequence of SEQ. ID. NO: 481 has a first bio-active GIP peptide hormone that is at least 30 amino acids long and has at leas 85% sequence identity to the first bio-active GIP peptide hormone of SEQ. ID. NO: 481, or pegylated derivatives thereof.
