Optimized Fc Variants and methods for their generation
The present invention relates to Fc variants having increased affinity for FcγRIIc, methods for their generation, Fc polypeptides comprising optimized Fc variants, and methods for using optimized Fc variants.
1 . An antibody or immunoadhesin comprising an Fc region of an IgG polypeptide, wherein said wherein said Fc region comprises:
a) an amino acid modification in the Fc region of said IgG polypeptide at position 298;
b) an amino acid modification in the Fc region of said IgG polypeptide at position 333;
c) an amino acid modification in the Fc region of said IgG polypeptide at position 334;
wherein said Fc variant has greater affinity for an Fc gamma receptor relative to said parent antibody or immunoadhesin, and wherein numbering is according to the EU index.
2 . An antibody or immunoadhesin according to claim 1 wherein said amino modification at position 298 is 298A, said amino acid modification at position 333 is 333A, and wherein said amino acid modification at position 334 is 334A.

An antibody or immunoadhesin according to claim 1 further comprising at least one modification at a position selected from the group consisting of: 221, 222, 223, 224, 225, 227, 228, 230, 231, 232, 233, 234, 235, 236, 237, 238, 239, 240, 241, 243, 244, 245, 246, 247, 249, 260, 262, 263, 264, 265, 266, 268, 269, 270, 271, 273, 274, 275, 278, 281, 282, 284, 288, 291, 292, 293, 294, 295, 296, 297, 299, 300, 302, 303, 304, 313, 317, 318, 323, 324, 325, 327, 328, 329, 330, 332, 335 and 336.
