Abstract

In a wide variety of human solid tumors, an aggressive, metastatic phenotype and poor clinical prognosis are associated with expression of the receptor tyrosine kinase Met. Disclosed herein are (a) a monoclonal antibody named Met4, which antibody is specific for Met, and (b) a hybridoma cell line that produces Met4. The Met4 antibody is particularly useful for detecting Met in formalin-fixed tissue. Methods of using the Met4 antibody for detection, diagnosis, prognosis, and evaluating therapeutic efficacy are provided.

Claims (31)

1. A hybridoma cell line deposited in the American Type Culture Collection under Accession Number PTA-7680.

2. A monoclonal antibody that binds to cMet hepatocyte growth factor receptor (Met-specific mAb) produced by the hybridoma cell line deposited in the American Type Culture Collection under Accession Number PTA-7680, or an antigen-binding fragment of said Met-specific mAb.

3. An isolated Met-specific monoclonal antibody (mAb) that binds to cMet hepatocyte growth factor receptor (Met) at amino acids 236-239 of SEQ ID NO:1, or an antigen-binding fragment of said Met-specific mAb.

4. A composition comprising

(a) the Met-specific mAb or fragment of claim 2 or 3 and

(b) a diagnostically acceptable carrier or excipient.

5. The Met-specific mAb or fragment of claim 2 or 3 , wherein the Met-specific mAb or fragment is linked to a detectable moiety.

6. The composition of claim 4 , wherein the Met-specific mAb or fragment is linked to a detectable moiety.

7. A kit, comprising:

(a) a first container comprising the Met-specific mAb or fragment of claim 2 or 3 ;

(b) a second container comprising a carrier for said Met-specific mAb or fragment; and

(c) instructions for using the Met-specific mAb or fragment to detect or measure Met in a sample or to diagnose a Met-related disease.

8. The kit of claim 7 wherein the Met-specific mAb or fragment is linked to a detectable moiety.

9. The kit of claim 7 further comprising

(d) a second antibody, or antigen-binding fragment thereof, which is specific for and binds to the Met-specific mAb or fragment.

10. The kit of claim 9 wherein the second antibody is labeled with a detectable moiety.

11. A method for detecting a presence of Met in a tissue sample or another biological sample from a subject, comprising

(a) contacting the sample with the composition of claim 4 , and

(b) detecting the binding of the Met-specific mAb or fragment to said sample, wherein binding of the Met-specific mAb or fragment indicates the presence of Met in the sample.

12. The method of claim 11 , wherein the Met-specific mAb or fragment is linked to a detectable moiety.

13. The method of claim 11 , wherein the sample has been fixed by treatment with formaldehyde before the contacting step (a).

14. The method of claim 12 wherein the sample has been fixed by treatment with formaldehyde before the contacting step (a).

15. A method of diagnosing a cancer having increased Met expression in a subject, comprising:

(a) contacting a tissue sample or other biological sample suspected of expressing Met from the subject with the composition of claim 4 ;

(b) detecting or measuring the binding of the Met-specific mAb or fragment to the sample, thereby determining a presence or a level of Met in the sample; and

(c) comparing the level of Met determined in (b) to a level of Met in a corresponding control sample from healthy subjects or subjects who do not have a disease with altered Met expression,

wherein

(i) the presence of Met determined in step (b) compared to an absence of Met in the control sample, or

(ii) the increased level of Met determined in step (b) compared to the level of Met in the control sample

is diagnostic of a cancer having increased Met expression.

16. The method of claim 15 wherein the sample has been fixed by treatment with formaldehyde before the contacting step (a).
