Chimeric and humanized antibodies to α5β1 integrin that modulate angiogenesis
View Patent ↗The present invention provides chimeric and humanized antibodies that specifically recognize α5β1 integrin, and methods for using the antibodies for reducing or inhibiting angiogenesis in a tissue. Also provided are methods of determining therapeutically acceptable doses of the antibodies and pharmaceutical compositions including the same.
1. A chimeric or humanized anti-α5β1 integrin antibody comprising: a heavy chain variable region comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 2, 3, 4, 5, and 6; a light chain variable region comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 8, 9, 10, 11, and 12; and a constant region, wherein the source of the constant region is a human IgG.
2. The chimeric or humanized anti-α5β1 integrin antibody of claim 1 , wherein the heavy chain variable region sequence is SEQ ID NO: 2, and the light chain variable region sequence is SEQ ID NO: 8.
3. The chimeric or humanized anti-α5β1 integrin antibody of claim 1 , wherein the heavy chain variable region sequence is SEQ ID NO: 3, and the light chain variable region sequence is SEQ ID NO: 9.
4. The chimeric or humanized anti-α5β1 integrin antibody of claim 1 , wherein the heavy chain variable region sequence is SEQ ID NO: 4, and the light chain variable region sequence is SEQ ID NO: 10.
5. The chimeric or humanized anti-α5β1 integrin antibody of claim 1 , wherein the heavy chain variable region sequence is SEQ ID NO: 5, and the light chain variable region sequence is SEQ ID NO: 11.
6. The chimeric or humanized anti-α5β1 integrin antibody of claim 1 , wherein the heavy chain variable region sequence is SEQ ID NO: 6, and the light chain variable region sequence is SEQ ID NO: 12.
7. The humanized anti-α5β1 integrin antibody of claim 1 , further comprising a constant region, wherein the source of the constant region is a human IgG.
8. The humanized anti-α5β1 integrin antibody of claim 7 , wherein the source of the constant region is human IgG4 or IgG2.
