Abstract

Methods for probing multiple targets in a biological sample are provided. The methods include the steps of providing a sample containing multiple targets, binding at least one probe having a binder coupled to an enzyme to one or more target present in the sample, and reacting the bound probe with an enzyme substrate coupled to a fluorescent signal generator. The methods include the steps of observing a signal from the fluorescent signal generator and applying to the sample a solution containing an oxidizing agent that substantially inactivates both the fluorescent signal generator and the enzyme. The methods further include the steps of binding at least one probe having a binder coupled to an enzyme to one or more target present in the sample of step, reacting the bound probe with an enzyme substrate coupled to a fluorescent signal generator; and observing a signal from the fluorescent signal generator. The methods disclosed herein also provide for multiple iterations of binding, observing, and oxidizing for deriving information about multiple targets in a single sample. An associated kit is also provided.

Claims (42)

1. A method of probing multiple targets in a sample comprising:

(a) providing the sample containing the multiple targets;

(b) binding at least one probe comprising a binder coupled to an enzyme to one or more target present in the sample;

(c) reacting the bound probe in step (b) with an enzyme substrate coupled to a fluorescent signal generator;

(d) observing a signal from the fluorescent signal generator of step (c);

(e) after observing the signal, inactivating both the fluorescent signal generator and the enzyme by applying a solution comprising an oxidizing agent;

(f) binding at least one probe comprising a binder coupled to an enzyme to one or more target present in the sample, after step (e);

(g) reacting the bound probe in step (f) with an enzyme substrate coupled to a fluorescent signal generator;

(h) observing a signal from the fluorescent signal generator of step (g).

2. The method of claim 1 , wherein the solution in step (e) is a basic solution.

3. The method of claim 2 , wherein the basic solution has a pH of 10.

4. The method of claim 2 , wherein the basic solution does not contain a reducing agent or a surfactant.

5. The method of claim 1 , wherein the enzyme is selected from peroxidase, alkaline phosphatase, β-D-galactosidase, and glucose oxidase.

6. The method of claim 1 , wherein the oxidizing agent is selected from hydrogen peroxide, potassium permanganate, sodium dichromate, aqueous bromine, iodine-potassium iodide, and t-butyl hydroperoxide.

7. The method of claim 1 , wherein the fluorescent signal generator comprises a cyanine dye.

8. The method of claim 1 , wherein the sample comprises whole cells or tissue sections.

9. The method of claim 1 , wherein the sample comprises proteins or nucleic acids.

10. The method of claim 1 , wherein steps (e)-(h) are repeated one or more time.

11. The method of claim 1 , wherein steps (e)-(h) are repeated at least 5, at least 10, or at least 20 times.

12. The method of claim 1 , wherein the oxidation step (e) is performed for less than 30 minutes.

13. The method of claim 1 , wherein the oxidation step (e) is performed for 30 seconds to 15 minutes.

14. The method of claim 1 , wherein the oxidation step (e) is performed at room temperature.

15. The method of claim 1 , wherein the observing step (d), step (h), or steps (d) and (h) comprises co-localizing at least two targets in the sample.

16. The method of claim 1 , wherein the observing step (d), step (h), or steps (d) and (h) comprises a quantitative measurement of at least one target in the sample.

17. The method of claim 1 , further comprising measuring one or more intensity values of the signal observed in observing step (d), step (h), or steps (d) and (h).

18. The method of claim 17 , further comprising correlating the intensity value with an amount of target present in the sample.

19. The method of claim 1 , wherein the fluorescent signal generator in step (c) is same as the fluorescent signal generator in step (g).

20. The method of claim 1 , wherein the fluorescent signal generator in step (c) is different from the fluorescent signal generator in step (g).

21. The method of claim 1 , wherein the signals observed in step (d) and step (h) are both detectable in a single detection channel.

22. The method of claim 1 , wherein the signal observed in step (d) or step (h) is independently detectable in different detection channels.

23. The method of claim 1 , wherein the enzyme is a peroxidase and the enzyme substrate is a peroxidase substrate.

24. A method of detecting multiple targets in a biological sample comprising:

(a) binding a first probe comprising a binder coupled to an enzyme to a first target present in the biological sample to form a first target-bound probe;

(b) reacting the first target-bound probe with an enzyme substrate coupled to a fluorescent signal generator to immobilize the fluorescent signal generator on or near the first target;

(c) detecting a signal from the immobilized fluorescent signal generator of step (b) to detect the first target;

(d) applying an oxidizing agent to the biological sample, after step (c), to modify the detected signal and to inactivate the enzyme;

(e) binding a second probe comprising a binder coupled to an enzyme to a second target present in the biological sample, after step (d), to form a second target-bound probe;

(f) reacting the second target-bound second probe with an enzyme substrate coupled to a fluorescent signal generator to immobilize the fluorescent signal generator on or near the second target; and

(g) detecting a signal from the immobilized fluorescent signal generator of step (f) to detect the second target.

25. The method of claim 24 , wherein the enzyme is a peroxidase and the enzyme substrate is a peroxidase substrate.

26. The method of claim 25 , wherein the peroxidase substrate coupled to the fluorescent signal generator is a tyramide substrate coupled to a cyanine dye.

27. The method of claim 26 , wherein the oxidizing agent is hydrogen peroxide.
