Abstract

Dialysis solutions comprising pyrophosphates and methods of making and using the dialysis solutions are provided. In an embodiment, the present disclosure provides a dialysis solution comprising a stable and therapeutically effective amount of pyrophosphate. The dialysis solution can be sterilized, for example, using a technique such as autoclave, steam, high pressure, ultra-violet, filtration or combination thereof. The dialysis solution can be in the form of a concentrate.

Claims (32)

1. A heat sterilized dialysis solution comprising a dialysis component selected from the group consisting of osmotic agents, electrolytes and combinations thereof, a therapeutically effective amount of pyrophosphate, a buffer comprising bicarbonate, and a pH greater than 7, wherein the bicarbonate buffer and the pH greater than 7 heat stabilize the pyrophosphate.

2. The dialysis solution of claim 1 , wherein the dialysis solution comprises a concentrate.

3. The dialysis solution of claim 1 , wherein the sterilization is performed by a technique selected from the group consisting of autoclave, steam and combinations thereof.

4. The dialysis solution of claim 1 , wherein the pyrophosphate is selected from the group consisting of pyrophosphoric acid, salt of pyrophosphate and combinations thereof.

The dialysis solution of claim 1 , wherein the osmotic agent is selected from the group consisting of glucose, glucose polymers, glucose polymer derivatives, cyclodextrins, modified starch, hydroxyethyl starch, polyols, fructose, amino acids, peptides, proteins, amino sugars, glycerol, N-acetyl glucosamine (NAG) and combinations thereof.

6. The dialysis solution of claim 1 , wherein the buffer further comprises a component selected from the group consisting of lactate, pyruvate, acetate, citrate, tris, amino acids, peptides, an intermediate of the KREBS cycle and combinations thereof.

7. The dialysis solution of claim 1 comprising at least two dialysis parts housed separately and the pyrophosphate is present with at least one of the dialysis parts and sterilized with said dialysis part.

8. The dialysis solution of claim 7 , wherein the dialysis part comprising the pyrophosphate has a pH greater than 7.

9. The dialysis solution of claim 7 , wherein the dialysis part comprising the pyrophosphate has a pH greater than 8.

10. The dialysis solution of claim 7 , wherein the dialysis part comprising the pyrophosphate has a pH greater than 9.

11. The dialysis solution of claim 7 , where the electrolytes are balanced between the first dialysis part and the second dialysis part.

A stable heat sterilized peritoneal dialysis solution comprising a therapeutically effective amount of pyrophosphate, a pH greater than 7, an osmotic agent selected from the group consisting of glucose, glucose polymers, glucose polymer derivatives, cyclodextrins, modified starch, hydroxyethyl starch, polyols, fructose, amino acids, peptides, proteins, amino sugars, glycerol, N-acetyl glucosamine (NAG) and combinations thereof, a buffer comprising bicarbonate, and electrolytes comprising sodium, potassium, magnesium, calcium and chloride, wherein the bicarbonate buffer and the pH greater than 7 heat stabilize the pyrophosphate.

13. A method of making a dialysis solution, the method comprising:

preparing a dialysis solution comprising pyrophosphate, a buffer comprising bicarbonate, a pH greater than 7, and at least one dialysis component selected from the group consisting of an osmotic agent, an electrolyte and combinations thereof, wherein the bicarbonate buffer and the pH greater than 7 heat stabilize the pyrophosphate; and

heat sterilizing the dialysis solution.

14. The method of claim 13 , wherein the dialysis solution comprises a concentrate.

15. The method of claim 13 , wherein the sterilization is performed by a technique selected from the group consisting of autoclave, steam and combinations thereof.

The method of claim 13 , wherein the dialysis solution comprises at least two separately housed dialysis parts, each dialysis part comprising at least one dialysis component selected from the group consisting of an osmotic agent, a buffer, an electrolyte and combinations thereof, at least one of the dialysis parts comprising the pyrophosphate, and wherein at least one of the dialysis parts is sterilized.

17. The method of claim 16 comprising mixing the dialysis parts to form a resultant dialysis solution.

18. The method of claim 16 , wherein the dialysis part comprising the pyrophosphate is sterilized and has a pH greater than 7.

19. The method of claim 18 , wherein the dialysis part comprising the pyrophosphate further comprises a buffer selected from the group consisting of lactate, pyruvate, acetate, citrate, tris, amino acids, peptides, an intermediate of the KREBS cycle and combinations thereof.

20. A method for providing dialysis to a patient, the method comprising:

providing at least two separately housed dialysis parts, each dialysis part comprising at least one dialysis component selected from the group consisting of an osmotic agent, an electrolyte and combinations thereof, at least one of the dialysis parts comprising pyrophosphate, a buffer comprising bicarbonate, and a pH greater than 7, wherein the bicarbonate buffer and the pH greater than 7 heat stabilize the pyrophosphate;

heat sterilizing at least the dialysis comprising the pyrophosphate;

mixing the dialysis parts to form a resultant heat sterilized dialysis solution; and

administering the heat sterilized dialysis solution to the patient.

21. The method of claim 20 , wherein the mixing is performed onsite by a mixing system.

22. A method of treating a dialysis patient, the method comprising:

providing a heat sterilized dialysis solution comprising a therapeutically effective amount of pyrophosphate, a buffer comprising bicarbonate, and a pH greater than 7 to a dialysis patient having vascular calcification, wherein the bicarbonate buffer and the pH greater than 7 heat stabilize the pyrophosphate.

23. The method of claim 22 , wherein the dialysis is peritoneal dialysis.

24. A multi-part dialysis product comprising:

at least two separately housed dialysis parts, each dialysis part comprising at least one dialysis component selected from the group consisting of an osmotic agent, an electrolyte and combinations thereof, at least one of the dialysis parts being heat sterilized and comprising pyrophosphate, a buffer comprising bicarbonate, and a pH greater than 7, wherein the bicarbonate buffer and the pH greater than 7 heat stabilize the pyrophosphate.
