Abstract

This invention provides high unit density arrays of microparticles and methods of assembling such arrays. The microparticles in the arrays may be functionalized with chemical or biological entities specific to a given target analyte. The high unit density arrays of this invention are formed on chips which may be combined to form multichip arrays according to the methods described herein. The chips and/or multichip arrays of this invention are useful for chemical and biological assays.

Claims (12)

1. A method for quality control of biochip manufacture, comprising:

exposing a patterned substrate containing a plurality of recesses, each sized for restraining an encoded bio-functionalized microbead, to a solution containing said microbeads under conditions such that microbeads are trapped in the recesses; and

optically imaging the substrate to determine what percentage of recesses are occupied with an encoded microbead.

2. The method of claim 1 wherein the microbeads are chemically-functionalized with DNA, RNA or proteins.

3. The method of claim 1 wherein the substrate is a semiconductor.

4. The method of claim 3 wherein the semiconductor is silicon.

5. The method of claim 1 wherein the optical encoding of the microbeads is with colored fluorescent dyes.

6. The method of claim 1 wherein the depth of the recesses is about 0.5 to 1.5 times the diameter of the microbeads.

7. The method of claim 6 wherein when microbeads are located in recesses, the recess depth is such that the apex of the microbeads reside at or below the planar surface of the substrate.

8. The method of claim 1 wherein the solution is 1% microbeads (10 mg/ml).

9. The method of claim 8 wherein the solution is 5% glycerol, 10 mM Tris, at pH 7.5.

10. The method of claim 1 wherein the microbeads occupy more than 95% of the recesses in the biochip.
