Abstract

Pyrazolopyridine-1,4-diamines and analogs thereof, e.g., pyrazolo[3,4-c]pyridine-1,4-diamines, pyrazolo[3,4-c]quinoline-1,4-diamines, 6,7,8,9-tetrahydro pyrazolo[3,4-c]quinoline-1,4-diamines, and pyrazolo[3,4-c]naphthyridine-1,4-di-amines, pharmaceutical compositions containing the compounds, intermediates, methods of making these compounds, and methods of use of these compounds as immunomodulators, for inducing cytokine biosynthesis in animals and in the treatment of diseases including viral and neoplastic diseases are disclosed.

Claims (206)

1. A compound of the Formula I:

wherein:

R 1 is selected from the group consisting of:

—R 4 ,

—Y—R 4 ,

—X—N(R 8 )—Y—R 4 ,

—X—C(R 6 )—N(R 8 )—R 4 ,

—X—O—C(R 6 )—N(R 8 )—R 4 ,

—X—S(O) 2 —N(R 8 )—R 4 ,

—X—O—R 4 , and

—X—R 5 ;

R 1 ′ is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, and hydroxyalkylenyl wherein the alkylenyl group contains at least two carbon atoms between the hydroxy or alkoxy substituent and the nitrogen atom to which R 1 ′ is bonded;

or R 1 and R 1 ′ together with the nitrogen atom to which they are bonded can join to form a group selected from the group consisting of:

A is selected from the group consisting of —CH(R 8 )—, —O—, —N(R 8 )—, —N(Y—R 4 )—, and —N(X—N(R 8 )—Y—R 4 )—;

X is C 2-20 alkylene;

Y is selected from the group consisting of —C(R 6 )—, —C(R 6 )—O—, —S(O) 2 —, —S(O) 2 —N(R 8 )—, and —C(R 6 )—N(R 11 )—; wherein R 11 is selected from the group consisting of hydrogen, alkyl, and arylalkylenyl; or R 11 and R 4 together with the nitrogen atom to which R 11 is bonded can join to form the group

a and b are independently integers from 1 to 4 with the proviso that when A is —O—, —N(R 8 )—, —N(Y—R 4 )—, or —N(X—N(R 8 )—Y—R 4 )— then a and b are independently integers from 2 to 4;

R A and R B are taken together to form a fused benzo ring unsubstituted or substituted by one or more R′ groups;

R 4 is selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl wherein the alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl groups can be unsubstituted or substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkylenyl, amino, alkylamino, (arylalkylenyl)amino, dialkylamino, and in the case of alkyl, alkenyl, alkynyl, and heterocyclyl, oxo, with the proviso that when R 1 is R 4 , R 4 is a substituted alkyl group, and the substituent contains a hetero atom which bonds directly to the alkyl group, then the alkyl group contains at least two carbons between the substituent and the nitrogen atom to which R 1 is bonded;

R 5 is selected from the group consisting of:

R 6 is selected from the group consisting of ═O and ═S;

R 7 is C 2-7 alkylene;

R 8 is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, hydroxyalkylenyl, arylalkylenyl, and heteroarylalkylenyl;

R 9 is selected from the group consisting of hydrogen and alkyl;

R′ is a non-interfering substituent; and

R″ is hydrogen or a non-interfering substituent;

or a pharmaceutically acceptable salt thereof.

2. A compound of the Formula II:

wherein:

R 1 is selected from the group consisting of:

—R 4 ,

—Y—R 4 ,

—X—N(R 8 )—Y—R 4 ,

—X—C(R 6 )—N(R 8 )—R 4 ,

—X—O—C(R 6 )—N(R 8 )—R 4 ,

—X—S(O) 2 —N(R 8 )—R 4 ,

—X—O—R 4 , and

—X—R 5 ;

R 1 ′ is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, and hydroxyalkylenyl wherein the alkylenyl group contains at least two carbon atoms between the hydroxy or alkoxy substituent and the nitrogen atom to which R 1 ′ is bonded;

or R 1 and R 1 ′ together with the nitrogen atom to which they are bonded can join to form a group selected from the group consisting of:

A is selected from the group consisting of —CH(R 8 )—, —O—, —N(R 8 )—, —N(Y—R 4 )—, and —N(X—N(R 8 )—Y—R 4 )—;

X is C 2-20 alkylene;

Y is selected from the group consisting of —C(R 6 )—, —C(R 6 )—O—, —S(O) 2 —, —S(O) 2 —N(R 8 )—, and —C(R 6 )—N(R 11 )—; wherein R 11 is selected from the group consisting of hydrogen, alkyl, and arylalkylenyl; or R 11 and R 4 together with the nitrogen atom to which R 11 is bonded can join to form the group

a and b are independently integers from 1 to 4 with the proviso that when A is —O—, —N(R 8 )—, —N(Y—R 4 )—, or —N(X—N(R 8 )—Y—R 4 )— then a and b are independently integers from 2 to 4;

R A1 and R B1 are taken together to form a fused benzo ring unsubstituted or substituted by one or more R groups, or substituted by one R 3 group, or substituted by one R 3 group and one R group;

R is selected from the group consisting of:

halogen,

hydroxy,

alkyl,

alkenyl,

haloalkyl,

alkoxy,

alkylthio, and

—N(R 9 ) 2 ;

R 2 is selected from the group consisting of:

—R 4 ′,

—X′—R 4 ′

—X′—Y′—R 4 ′, and

—X′—R 5 ′;

R 3 is selected from the group consisting of:

—Z—R 4 ′,

—Z—X′—R 4 ′,

—Z—X′—Y′—R 4 ′,

—Z—X′—Y′—X′—Y′—R 4 ′, and

—Z—X′—R 5 ′;

R 4 is selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl wherein the alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl groups can be unsubstituted or substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkylenyl, amino, alkylamino, (arylalkylenyl)amino, dialkylamino, and in the case of alkyl, alkenyl, alkynyl, and heterocyclyl, oxo, with the proviso that when R 1 is R 4 , R 4 is a substituted alkyl group, and the substituent contains a hetero atom which bonds directly to the alkyl group, then the alkyl group contains at least two carbons between the substituent and the nitrogen atom to which R 1 is bonded;

Q is selected from the group consisting of a bond, —C(R 6 )—, —C(R 6 )—C(R 6 )—, —S(O) 2 —, —C(R 6 )—N(R 8 )—W—, —S(O) 2 —N(R 8 )—, —C(R 6 )—O—, —C(R 6 )—S—, and —C(R 6 )—N(OR 9 )—;

V is selected from the group consisting of —C(R 6 )—, —O—C(R 6 )—, N(R 8 )—C(R 6 )—, and —S(O) 2 —;

W is selected from the group consisting of a bond, —C(O)—, and —S(O) 2 —;

X′ is selected from the group consisting of alkylene, alkenylene, alkynylene, arylene, heteroarylene, and heterocyclylene wherein the alkylene, alkenylene, and alkynylene groups can be optionally interrupted or terminated by arylene, heteroarylene or heterocyclylene and optionally interrupted by one or more —O—groups;

Y′ is selected from the group consisting of:

Z is a bond or —O—; and

c and d are independently integers from 1 to 6 with the proviso that c+d is ≦7; or a pharmaceutically acceptable salt thereof.

3. A compound of the Formula IV:

R 1 is selected from the group consisting of:

R 1 ′ is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, and hydroxyalkylenyl wherein the alkylenyl group contains at least two carbon atoms between the hydroxy or alkoxy substituent and the nitrogen atom to which R 1 ′ is bonded;

or R 1 and R 1 ′ together with the nitrogen atom to which they are bonded can join to form a group selected from the group consisting of:

A is selected from the group consisting of —CH(R 8 )—, —O—, —N(R 8 )—, —N(Y—R 4 )—, and —N(X—N(R 8 )—Y—R 4 )—;

X is C 2-20 alkylene;

4. A compound of the Formula XIV:

wherein:

X 1 is a bond or C 1-4 alkylene;

A 1 is selected from the group consisting of —N(R 8 )—and —N(—Y—R 4 )—;

Y is selected from the group consisting of —C(R 6 )—, —C(R 6 )—O—, —S(O) 2 —, —S(O) 2 —N(R 8 )—, and —C(R 6 )—N(R 11 )—; wherein R 11 is selected from the group consisting of hydrogen, alkyl, and arylalkylenyl; or R 11 and R 4 together with the nitrogen atom to which R 11 , is bonded can join to form the group

A is selected from the group consisting of —CH(R 8 )—, —O—, —N(R 8 )—, —N(Y—R 4 )—, and —N(X—N(R 8 )—Y—R 4 )—;

a and b are independently integers from 1 to 4 with the proviso that when A is —O—, —N(R 8 )—, —N(Y—R 4 )—, or —N(X—N(R 8 )—Y—R 4 )—then a and b are independently integers from 2 to 4;

X is C 2-20 alkylene;

R 1 ′ is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, and hydroxyalkylenyl wherein the alkylenyl group contains at least two carbon atoms between the hydroxy or alkoxy substituent and the nitrogen atom to which R 1 ′ is bonded;

R 4 is selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl wherein the alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl groups can be unsubstituted or substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkylenyl, amino, alkylamino, (arylalkylenyl)amino, dialkylamino, and in the case of alkyl, alkenyl, alkynyl, and heterocyclyl, oxo, with the proviso that when R 1 is R 4 , R 4 is a substituted alkyl group, and the substituent contains a hetero atom which bonds directly to the alkyl group, then the alkyl group contains at least two carbons between the substituent and the nitrogen atom to which R 1 is bonded;

R 5 ′ is selected from the group consisting of:

R 6 is selected from the group consisting of ═O and ═S;

R 7 is C 2-7 alkylene;

R 8 is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, hydroxyalkylenyl, arylalkylenyl, and heteroarylalkylenyl;

R 9 is selected from the group consisting of hydrogen and alkyl;

R 10 is C 3-8 alkylene;

A′ is selected from the group consisting of —O—, —C(O)—, —S(O) 0-2 —, and —N(R 4 )—;

A″ is selected from the group consisting of —O—, —S(O) 0-2 —, —N(—Q—R4)—, and —CH 2 —;

Q is selected from the group consisting of a bond, —C(R 6 )—, —C(R 6 )—C(R 6 )—, —S(O) 2 —, —C(R 6 )—N(R 8 )—W—, —S(O) 2 —N(R 8 )—, —C(R 6 )—O—, —C(R 6 )—S—, and —C(R 6 )—N(OR 9 )—;

V is selected from the group consisting of —C(R 6 )—, —O—C(R 6 )—, —N(R 8 )—C(R 6 )—, and —S(O) 2 —;

W is selected from the group consisting of a bond, —C(O)—, and —S(O) 2 —;

X′ is selected from the group consisting of alkylene, alkenylene, alkynylene, arylene, heteroarylene, and heterocyclylene wherein the alkylene, alkenylene, and alkynylene groups can be optionally interrupted or terminated by arylene, heteroarylene or heterocyclylene and optionally interrupted by one or more —O—groups;.

Y′ is selected from the group consisting of:

Z is a bond or —O—; and

c and d are independently integers from 1 to 6 with the proviso that c+d is ≦7; or a pharmaceutically acceptable salt thereof.

5. The compound or salt of claim 3 , wherein n is 0.

6. The compound or salt of claim 1 , 2 , or 3 , wherein R 1 is R 4 .

7. The compound or salt of claim 6 wherein R 4 is alkyl or arylalkylenyl.

8. The compound or salt of claim 7 wherein R 4 is methyl, ethyl, n-propyl, 1-methylethyl, n-butyl, 1-ethylpropyl, 2-methylpropyl, 3-methylbutyl, benzyl, 2-phenylethyl, or 3-phenylpropyl.

9. The compound or salt of claim 1 , 2 , or 3 , wherein R 1 is —X—N(R 8 )—Y—R 4 .

10. The compound or salt of claim 9 wherein X is C 2-4 alkylene, R 8 is hydrogen, R 4 is C 1-6 alkyl, Y is —C(O)—, —S(O) 2 —, or —C(O)—N(R 11 )— wherein R 11 is hydrogen or R 11 and R 4 join to form a morpholine ring.

11. The compound or salt of claim 1 , 2 , 3 , or 4 , wherein R 1 ′ is hydrogen.

12. The compound or salt of claim 2 , 3 or 4 , wherein R 2 is hydrogen, alkyl, alkoxyalkylenyl, or hydroxyalkylenyl.

13. The compound or salt of claim 2 , 3 , or 4 , wherein R is hydroxy.

14. The compound or salt of claim 2 , 3 , or 4 , wherein R 3 is selected from the group consisting of phenyl, p-tolyl, benzyloxy, (4-chlorobenzyl)oxy, (4-methylbenzyl)oxy, 3-furyl, pyridin-3-yl, 5-(hydroxymethyl)pyridin-3-yl, 6-chloropyridin-3-yl, 6-fluoropyridin-3-yl, 6-methylpyridin-3-yl, 3-quinolin-3-yl, and thiazol-4-ylmethoxy.

15. The compound or salt of claim 3 or 5 , wherein m is 0.

16. The compound or salt of claim 3 or 5 , wherein m is 1.

17. The compound or salt of claim 4 , wherein X 1 is C 1-4 alkylene; and A 1 is —N(R 8 )—.

18. The compound or salt of claim 4 , wherein X 1 is C 1-4 alkylene; and A 1 is —N(—Y—R 4 )—.

19. The compound or salt of claim 4 or 18 , wherein X 1 is C 1-4 alkylene; and Y is —C(O)—, —S(O) 2 —, or —C(O)—N(R 11 )—.

20. The compound or salt of claim 19 , wherein R 4 is C 1-6 alkyl; and R 11 is hydrogen or methyl.

21. The compound or salt of claim 19 , wherein Y is —C(O)—N(R 11 )—; and R 4 and R 11 form the group:

wherein A is —CH 2 —or —O—and a and b are each independently 1 or 2, with the proviso that when A is —O—, then a and b are each 2.

22. The compound or salt of claim 12 , wherein R 2 is C 1-4 alkyl, C 1-4 alkyl-O—C 2-4 alkylenyl, or hydroxyC 2-4 alkylenyl.

23. The compound or salt of claim 22 , wherein R 2 is methyl, ethyl, n-propyl, n-butyl, 2-methoxyethyl, or 2-hydroxyethyl.

24. A pharmaceutical composition comprising a therapeutically effective amount of a compound or salt of claim 2 in combination with a pharmaceutically acceptable carrier.
