Abstract

Bone cages are disclosed including devices for biocompatible implantation. The structures of bone are useful for providing living cells and tissues as well as biologically active molecules to subjects.

Claims (63)

1. A method for delivering one or more biologically active molecules to a subject, comprising:

identifying the one or more biologically active molecules,

covering at least a portion of one or more devices with a semipermeable component prior to implantation of the one or more devices in the subject, the one or more devices constructed of organic bone and micromachined to reshape a natural internal cavity thereof and to form a plurality of holes in the one or more devices that access the natural internal cavity, wherein the semi-permeable component comprises a layer of confluent cells covering at least a portion of the plurality of holes of the one or more devices, and

implanting in the subject the one or more,

wherein the one or more biologically active molecules are included in the one or more devices and the one or more biologically active molecules are at least partially surrounded by the semi-permeable component.

2. A method for delivering one or more biologically active molecules to a subject, comprising:

identifying the one or more biologically active molecules,

covering at least a portion of one or more devices with a semipermeable component prior to implantation of the one or more devices in the subject, the one or more devices constructed of anorganic bone and micromachined to reshape a natural internal cavity thereof and to form a plurality of holes in the one or more devices that access the natural internal cavity, wherein the semi-permeable component comprises a layer of confluent cells covering at least a portion of the plurality of holes of the one or more devices, and

implanting in the subject the one or more devices,

wherein the one or more biologically active molecules are included in the one or more devices and the one or more biologically active molecules are at least partially surrounded by the semi-permeable component.

3. The method of claim 2 , wherein the anorganic bone is freeze-dried bone.

4. The method of claim 2 , further comprising:

micromachining the anorganic bone to reshape the internal cavity thereof by focused beam machining of the implantable structure.

5. The method of claim 2 , further comprising:

providing the one or more biologically active molecules to the micromachined internal cavity.

6. The method of claim 5 , wherein providing the one or more biologically active molecules to the micromachined internal cavity comprises:

providing the one or more biologically active molecules to the micromachined internal cavity following implantation of the one or more devices into the subject.

7. The method of claim 5 , wherein providing the one or more biologically active molecules to the micromachined internal cavity comprises:

providing the one or more biologically active molecules to the micromachined internal cavity before implantation of the one or more devices into the subject.

8. The method of claim 2 , further comprising:

providing the one or more biologically active molecules to the exterior of the anorganic bone.

9. The method of claim 2 , further comprising:

selecting the one or more devices constructed of anorganic bone having the semi-permeable component lining the micromachined internal cavity.

10. The method of claim 9 , further comprising:

providing the one or more biologically active molecules to the micromachined internal cavity.

11. The method of claim 2 , further comprising:

providing the one or more semi-permeable components to the micromachined internal cavity of the one or more devices.

12. The method of claim 2 , further comprising:

selecting the one or more devices constructed of anorganic bone having the semi-permeable component surrounding the anorganic bone.

13. The method of claim 2 , further comprising:

surrounding the anorganic bone with the one or more semi-permeable components.

14. The method of claim 2 , further comprising:

identifying one or more living cells or tissues to provide the one or more biologically active molecules; and

providing the one or more living cells or tissues to the internal cavity.

15. The method of claim 14 , wherein the one or more living cells or tissues are engineered to release the one or more biologically active molecules.

16. The method of claim 2 , wherein the one or more devices constructed of anorganic bone and micromachined to reshape the natural internal cavity have one or more closable openings.

17. The method of claim 16 , further comprising:

closing the one or more closable openings.

18. The method of claim 16 , further comprising:

providing the one or more biologically active molecules to the micromachined internal cavity through one or more of the one or more closable openings.

19. The method of claim 18 , providing the one or more biologically active molecules to the internal cavity through one or more of the one or more closable openings comprises:

providing the one or more biologically active molecules to the micromachined internal cavity through one or more of the one or more closable openings following implantation in the subject.

20. A method for delivering one or more biologically active molecules to a subject, comprising:

identifying the one or more biologically active molecules,

covering at least a portion of one or more devices with a semipermeable component prior to implantation of the one or more devices in the subject, the one or more devices constructed of bone allogeneic to the subject and micromachined to reshape a natural internal cavity thereof and to form a plurality of holes in the one or more devices that access the natural internal cavity, wherein the semi-permeable component comprises a layer of confluent cells covering at least a portion of the plurality of holes of the one or more devices, and

implanting in the subject the one or more devices,

wherein the one or more biologically active molecules are included in the one or more devices and the one or more biologically active molecules are at least partially surrounded by the semi-permeable component.

21. The method of claim 20 , wherein the subject is an animal.

22. The method of claim 21 , wherein the animal is selected from the group consisting of a domesticated animal, a farm animal, a marine animal, a sport animal, a wild animal, a research animal, a zoo animal, a bird, an amphibian, a reptile, a fish, and a mammal.

23. The method of claim 22 , wherein the mammal is a human.

24. A method for delivering one or more biologically active molecules to a subject, comprising:

identifying the one or more biologically active molecules,

covering at least a portion of one or more devices with a semipermeable component prior to implantation of the one or more devices in the subject, the one or more devices constructed of bone xenogeneic to the subject and micromachined to reshape a natural internal cavity thereof and to form a plurality of holes in the one or more devices that access the natural internal cavity, wherein the semi-permeable component comprises a layer of confluent cells covering at least a portion of the plurality of holes of the one or more devices, and

implanting in the subject the one or more devices,

wherein the one or more biologically active molecules are included in the one or more devices and the one or more biologically active molecules are at least partially surrounded by the semi-permeable component.

25. A method for delivering one or more biologically active molecules to a subject, comprising:

identifying the one or more biologically active molecules,

identifying one or more living cells or tissues to provide the one or more biologically active molecules;

selecting one or more implantable structures constructed of bone and micromachined to reshape a natural internal cavity thereof and to form a plurality of holes in the one or more devices constructed of bone that access the natural internal cavity, wherein the micromachined internal cavity is lined with a semi-permeable component prior to implantation of the device, the semi-permeable component comprising a layer of confluent cells covering at least a portion of the plurality of holes of one or more devices;

implanting the one or more devices into the subject; and

providing the one or more living cells or tissues engineered to release the one or more biologically active molecules to the micromachined internal cavity of the one or more devices.

26. The method of claim 2 , wherein the layer of confluent cells covering at least a portion of the plurality of holes of one or more devices includes the layer of confluent cells covering an exterior of the one or more devices with the layer of confluent cells.

27. The method of claim 2 , wherein the layer of confluent cells covering at least a portion of the plurality of holes of one or more devices includes the layer of confluent cells covering an interior of the one or more devices with the layer of confluent cells.
