Abstract

The present invention relates to IL-17 receptor A (IL-17RA or IL-17R) antigen binding proteins, such as antibodies, and the polynucleotide sequences encoding them, as well as host cells, expression vectors, and methods of making IL-17 receptor A antigen binding proteins.

Claims (113)

1. An isolated polynucleotide, comprising a polynucleotide that encodes a polypeptide comprising a light chain variable domain comprising SEQ ID NO:40 and a heavy chain variable domain comprising SEQ ID NO:14.

2. An isolated polynucleotide, comprising a polynucleotide that encodes a polypeptide comprising a light chain CDR1 comprising SEQ ID NO:224, a light chain CDR2 comprising SEQ ID NO:225, a light chain CDR3 comprising SEQ ID NO:226, a heavy chain CDR1 comprising SEQ ID NO:146, a heavy chain CDR2 comprising SEQ ID NO:147, and a heavy chain CDR3 comprising SEQ ID NO:148.

3. The polynucleotide of claim 1 , wherein said polynucleotide comprises a light chain variable domain-encoding polynucleotide comprising SEQ ID NO:93 and a heavy chain variable domain-encoding polynucleotide comprising SEQ ID NO:67.

The polynucleotide of claim 2 , wherein said polynucleotide comprises a light chain CDR1-encoding polynucleotide comprising SEQ ID NO:384, a light chain CDR2-encoding polynucleotide comprising SEQ ID NO:385, a light chain CDR3-encoding polynucleotide comprising SEQ ID NO:386, a heavy chain CDR1-encoding polynucleotide comprising SEQ ID NO:305, a heavy chain CDR2-encoding polynucleotide comprising SEQ ID NO:306, and a heavy chain CDR3-encoding polynucleotide comprising SEQ ID NO:307.

5. An isolated plasmid, comprising said polynucleotide according to any of claim 1 , 2 , 3 or 4 .

6. The isolated plasmid of claim 5 , wherein said plasmid is an expression vector.

7. An isolated cell, comprising said plasmid of claim 5 .

8. The isolated cell of claim 7 , wherein a chromosome of said cell comprises said polynucleotide.

9. The isolated cell of claim 7 , wherein said cell is a hybridoma.

10. An isolated cell comprising the expression vector of claim 6 .

11. The isolated cell of claim 10 , wherein said cell is selected from the group consisting of:

a. a prokaryotic cell;

b. a eukaryotic cell;

c. a mammalian cell;

d. an insect cell; and

e. a CHO cell.

12. A method of making a polypeptide that specifically binds IL-17 receptor A, comprising incubating said isolated cell of claim 10 under conditions that allow it to express said polypeptide.

13. The polynucleotide according to any of claim 1 , 2 , 3 , or 4 , wherein said polynucleotide encodes said polypeptide and wherein said polypeptide comprises an antibody that specifically binds IL-17 receptor A, wherein said antibody is selected from the group consisting of:

a. a human antibody;

b. a humanized antibody;

c. a chimeric antibody;

d. a recombinant antibody;

e. a single chain antibody;

f. a diabody;

g. a triabody;

h. a tetrabody;

i. a Fab fragment;

j. a F(ab′)2 fragment;

k. an IgD antibody;

l. an IgE antibody;

m. an IgM antibody;

n. an IgG1 antibody;

o. an IgG2 antibody;

p. an IgG3 antibody; and

q. an IgG4 antibody.

14. The polynucleotide of claim 13 , wherein said polynucleotide encodes said antibody and wherein said antibody is selected from the group consisting of:

a) an antibody consisting of a heavy chain sequence of SEQ ID NO:427 and a light chain sequence of SEQ ID NO:429;

b) an antibody comprising a heavy chain sequence comprising SEQ ID NO: 427 and a light chain sequence comprising SEQ ID NO:429;

d) an antibody comprising a light chain variable region sequence comprising SEQ ID NO:40 and a heavy chain variable region sequence comprising SEQ ID NO:14; and

e) an antibody comprising a heavy chain CDR1 comprising SEQ ID NO:146, a heavy chain CDR2 comprising SEQ ID NO:147, a heavy chain CDR3 comprising SEQ ID NO:148, a light chain CDR1 comprising SEQ ID NO:224, a light chain CDR2 comprising SEQ ID NO:225, and a light chain CDR3 comprising SEQ ID NO:226.

15. The polynucleotide of claim 14 , wherein said polynucleotide comprises a polynucleotide selected from the group consisting of:

a) a heavy chain-encoding polynucleotide sequence consisting of SEQ ID NO:426 and a light chain-encoding polynucleotide sequence consisting of SEQ ID NO:428;

b) a heavy chain-encoding polynucleotide sequence comprising SEQ ID NO: 426 and a light chain-encoding polynucleotide sequence comprising SEQ ID NO:428;

c) a heavy chain variable region-encoding polynucleotide sequence comprising SEQ ID NO:67 and a light chain variable region-encoding polynucleotide sequence comprising SEQ ID NO:93; and

d) a light chain CDR1-encoding polynucleotide comprising SEQ ID NO:384, CDR2-encoding polynucleotide comprising SEQ ID NO:385, CDR3-encoding polynucleotide comprising SEQ ID NO:386, a heavy chain CDR1-encoding polynucleotide comprising SEQ ID NO:305, CDR2-encoding polynucleotide comprising SEQ ID NO:306, and a CDR3-encoding polynucleotide comprising SEQ ID NO:307.

16. An isolated plasmid, comprising the polynucleotide of claim 14 .

17. An isolated cell, comprising a plasmid comprising the polynucleotide of claim 14 .

18. An isolated cell, comprising an expression vector comprising the polynucleotide of claim 14 .

19. An isolated cell, wherein the cell is a CHO cell and wherein the CHO cell comprises an expression vector comprising the polynucleotide of claim 14 .

20. A method of making a polypeptide that specifically binds IL-17 receptor A, comprising incubating the isolated cell of claim 19 under conditions that allow it to express said polypeptide.

21. An isolated plasmid, comprising the polynucleotide of claim 15 .

22. An isolated cell, comprising a plasmid comprising the polynucleotide of claim 15 .

23. An isolated cell, comprising an expression vector comprising the polynucleotide of claim 15 .

24. An isolated cell, wherein the cell is a CHO cell and wherein the CHO cell comprises an expression vector comprising the polynucleotide of claim 15 .

25. A method of making a polypeptide that specifically binds IL-17 receptor A, comprising incubating the isolated cell of claim 24 under conditions that allow it to express said polypeptide.

26. An isolated polynucleotide, wherein said polynucleotide encodes an antibody and wherein said antibody is selected from the group consisting of:

a) an antibody consisting of a heavy chain sequence of SEQ ID NO:427 and a light chain sequence of SEQ ID NO:429;

b) an antibody comprising a heavy chain sequence comprising SEQ ID NO: 427 and a light chain sequence comprising SEQ ID NO:429;

d) an antibody comprising a light chain variable region sequence comprising SEQ ID NO:40 and a heavy chain variable region sequence comprising SEQ ID NO:14; and

e) an antibody comprising a heavy chain CDR1 comprising SEQ ID NO:146, a heavy chain CDR2 comprising SEQ ID NO:147, a heavy chain CDR3 comprising SEQ ID NO:148, a light chain CDR1 comprising SEQ ID NO:224, a light chain CDR2 comprising SEQ ID NO:225, and a light chain CDR3 comprising SEQ ID NO:226.

27. The polynucleotide of claim 26 , wherein said polynucleotide comprises a polynucleotide selected from the group consisting of:

a) a heavy chain-encoding polynucleotide sequence consisting of SEQ ID NO:426 and a light chain-encoding polynucleotide sequence consisting of SEQ ID NO:428;

b) a heavy chain-encoding polynucleotide sequence comprising SEQ ID NO: 426 and a light chain-encoding polynucleotide sequence comprising SEQ ID NO:428;

c) a heavy chain variable region-encoding polynucleotide sequence comprising SEQ ID NO:67 and a light chain variable region-encoding polynucleotide sequence comprising SEQ ID NO:93; and

d) a light chain CDR1-encoding polynucleotide comprising SEQ ID NO:384, CDR2-encoding polynucleotide comprising SEQ ID NO:385, CDR3-encoding polynucleotide comprising SEQ ID NO:386, a heavy chain CDR1-encoding polynucleotide comprising SEQ ID NO:305, CDR2-encoding polynucleotide comprising SEQ ID NO:306, and a CDR3-encoding polynucleotide comprising SEQ ID NO:307.

28. An isolated plasmid, comprising said polynucleotide of claim 26 .

29. The isolated plasmid of claim 28 , wherein said plasmid is an expression vector.

30. An isolated cell, comprising said plasmid of claim 29 .

31. The isolated cell of claim 30 , wherein a chromosome of said cell comprises said polynucleotide.

32. The isolated cell of claim 31 , wherein said cell is a hybridoma.

33. The isolated cell of claim 30 , wherein said cell is selected from the group consisting of:

a. a prokaryotic cell;

b. a eukaryotic cell;

c. a mammalian cell;

d. an insect cell; and

e. a CHO cell.

34. A method of making an antibody that specifically binds IL-17 receptor A, comprising incubating said isolated cell of claim 30 under conditions that allow it to express said antibody.

35. An isolated polynucleotide, wherein said polynucleotide encodes an IgG human monoclonal antibody and wherein said antibody is selected from the group consisting of:

a) an antibody consisting of a heavy chain sequence of SEQ ID NO:427 and a light chain sequence of SEQ ID NO:429;

b) an antibody comprising a heavy chain sequence comprising SEQ ID NO: 427 and a light chain sequence comprising SEQ ID NO:429;

c) an antibody comprising a light chain variable region sequence comprising SEQ ID NO:40 and a heavy chain variable region sequence comprising SEQ ID NO:14; and

d) an antibody comprising a heavy chain CDR1 comprising SEQ ID NO:146, a heavy chain CDR2 comprising SEQ ID NO:147, a heavy chain CDR3 comprising SEQ ID NO:148, a light chain CDR1 comprising SEQ ID NO:224, a light chain CDR2 comprising SEQ ID NO:225, and a light chain CDR3 comprising SEQ ID NO:226.

36. The polynucleotide of claim 35 , wherein said polynucleotide comprises a polynucleotide selected from the group consisting of:

a) a heavy chain-encoding polynucleotide sequence consisting of SEQ ID NO:426 and a light chain-encoding polynucleotide sequence consisting of SEQ ID NO:428;

b) a heavy chain-encoding polynucleotide sequence comprising SEQ ID NO: 426 and a light chain-encoding polynucleotide sequence comprising SEQ ID NO:428;

c) a heavy chain variable region-encoding polynucleotide sequence comprising SEQ ID NO:67 and a light chain variable region-encoding polynucleotide sequence comprising SEQ ID NO:93; and

d) a light chain CDR1-encoding polynucleotide comprising SEQ ID NO:384, CDR2-encoding polynucleotide comprising SEQ ID NO:385, CDR3-encoding polynucleotide comprising SEQ ID NO:386, a heavy chain CDR1-encoding polynucleotide comprising SEQ ID NO:305, CDR2-encoding polynucleotide comprising SEQ ID NO:306, and a CDR3-encoding polynucleotide comprising SEQ ID NO:307.

37. An isolated plasmid, comprising said polynucleotide of claim 35 .

38. The isolated plasmid of claim 37 , wherein said plasmid is an expression vector.

39. An isolated cell, comprising said plasmid of claim 38 .

40. The isolated cell of claim 39 , wherein a chromosome of said cell comprises said polynucleotide.

41. The isolated cell of claim 40 , wherein said cell is a hybridoma.

42. The isolated cell of claim 39 , wherein said cell is selected from the group consisting of:

a. a prokaryotic cell;

b. a eukaryotic cell;

c. a mammalian cell;

d. an insect cell; and

e. a CHO cell.

43. A method of making an antibody that specifically binds IL-17 receptor A, comprising incubating said isolated cell of claim 39 under conditions that allow it to express said antibody.

44. An isolated polynucleotide, wherein said polynucleotide encodes an IgG2 human monoclonal antibody comprising SEQ ID NO:427 and SEQ ID NO:429.

45. The polynucleotide of claim 44 , wherein said polynucleotide comprises SEQ ID NO:426 and SEQ ID NO:428.

46. An isolated plasmid, comprising said polynucleotide of claim 44 .

47. The isolated plasmid of claim 46 , wherein said plasmid is an expression vector.

48. An isolated cell, comprising said plasmid of claim 47 .

49. The isolated cell of claim 48 , wherein a chromosome of said cell comprises said polynucleotide.

50. The isolated cell of claim 49 , wherein said cell is a hybridoma.

51. The isolated cell of claim 48 , wherein said cell is selected from the group consisting of:

a. a prokaryotic cell;

b. a eukaryotic cell;

c. a mammalian cell;

d. an insect cell; and

e. a CHO cell.

52. A method of making an antibody that specifically binds IL-17 receptor A, comprising incubating said isolated cell of claim 48 under conditions that allow it to express said antibody.
