Abstract

It is intended to provide an adenosine A 2A receptor antagonist containing as an active ingredient a thiazole derivative represented by the general formula (I) [in the formula, R 1 represents a substituted or unsubstituted five-membered aromatic heterocyclic group containing at least one oxygen atom, R 2 represents a substituted or unsubstituted heterocyclic group, —COR 6 (in the formula, R 6 represents a substituted or unsubstituted heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group or the like) or the like, R 3 represents the general formula (II), —NHCOR (in the formula, R 7 represents —NR 8 R 9 or the like) or the like], or a pharmacologically acceptable salt thereof, etc. (I) (II)

Claims (40)

1 . A method for antagonizing adenosine A 2A receptor comprising the step of administering an effective amount of a thiazole derivative represented by the general formula (I):

{wherein R 1 represents a substituted or unsubstituted 5-membered aromatic heterocyclic group containing at least one oxygen atom;

R 2 represents halogen, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, substituted or unsubstituted aromatic heterocyclic-alkyl, —NR 4 R 5 (wherein R 4 and R 5 may be the same or different, and each represents a hydrogen atom, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted lower alkanoyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl), or —COR 6 (wherein R 6 represents substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl); and

R 3 represents —NHCOR 7 [wherein R 7 represents —NR 8 R 9 (wherein R 8 and R 9 may be the same or different, and each represents a hydrogen atom, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted lower alkanoyl, substituted or unsubstituted lower alkoxy, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, substituted or unsubstituted aroyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl, or R 8 and R 9 are combined together with the adjacent nitrogen atom thereto to form a substituted or unsubstituted alicyclic heterocyclic group) or —OR 10 (wherein R 10 represents substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl)], —CONHR 11 (wherein R 11 represents a hydrogen atom, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted lower alkanoyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, substituted or unsubstituted aroyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl), or the general formula (II):

 (wherein ═X— represents ═CH— or ═N—, R 12 and R 13 may be the same or different, and each represents a hydrogen atom, halogen, hydroxy, nitro, azido, amino, cyano, carboxy, formyl, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted lower alkanoyl, substituted or unsubstituted lower alkoxy, substituted or unsubstituted cycloalkyl, substituted or unsubstituted lower alkylamino, substituted or unsubstituted di-lower alkylamino, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, substituted or unsubstituted aroyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl, or R 12 and R 13 are combined with the two carbon atoms adjacent thereto, respectively, to form a substituted or unsubstituted carbon ring, or a substituted or unsubstituted heterocyclic ring)}

or a pharmaceutically acceptable salt thereof.

2 . The method for antagonizing adenosine A 2A receptor according to claim 1 , wherein R 1 is substituted or unsubstituted furyl.

4 . A thiazole derivative represented by the general formula (IA):

(wherein R 1A represents a substituted or unsubstituted 5-membered aromatic heterocyclic group containing at least one oxygen atom,

R 6A represents substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl)

or a pharmaceutically acceptable salt thereof.

5 . The thiazole derivative or the pharmaceutically acceptable salt thereof according to claim 4 , wherein R 1A is substituted or unsubstituted furyl.

6 . The thiazole derivative or the pharmaceutically acceptable salt thereof according to claim 4 or 5 , wherein R 6A is a substituted or unsubstituted alicyclic heterocyclic group, or a substituted or unsubstituted aromatic heterocyclic group, and —CO— in —COR 6A binds to a carbon atom of R 6A .

7 . The thiazole derivative or the

pharmaceutically acceptable salt thereof according to claim 4 , wherein R 6A is a substituted or unsubstituted alicyclic heterocyclic group.

8 . The thiazole derivative or the pharmaceutically acceptable salt thereof according to claim 4 , wherein R 6A is substituted or unsubstituted tetrahydropyranyl, substituted or unsubstituted pyridyl or substituted or unsubstituted dioxepanyl.

9 . The thiazole derivative or the

pharmaceutically acceptable salt thereof according to claim 4 , wherein R 3A is —CONHR 11 (wherein R 11 has the same meaning as defined above).

10 . The thiazole derivative or the

pharmaceutically acceptable salt thereof according to claim 4 , wherein R 3A is the formula (II):

(wherein X represents ═CH— or ═N—, and

R 12 and R 13 may be the same or different, and each represents a hydrogen atom, halogen, hydroxy, nitro, azido, amino, cyano, carboxy, formyl, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted lower alkanoyl, substituted or unsubstituted lower alkoxy, substituted or unsubstituted cycloalkyl, substituted or unsubstituted lower alkylamino, substituted or unsubstituted di-lower alkylamino, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, substituted or unsubstituted aroyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl, or R 12 and R 13 are combined with the two carbon atoms adjacent thereto, respectively, to form a substituted or unsubstituted carbon ring, or a substituted or unsubstituted heterocyclic ring).

11 . The thiazole derivative or the pharmaceutically acceptable salt thereof according to claim 10 , wherein R 12 and R 13 are combined with the two carbon atoms adjacent thereto, respectively, to form a substituted or unsubstituted carbon ring, or a substituted or unsubstituted heterocyclic ring.

12 . The thiazole derivative or the

pharmaceutically acceptable salt thereof according to claim 4 , wherein R 3A is —NHCOR 7 .

13 . A pharmaceutical composition comprising, as an active ingredient, the thiazole derivative or the pharmaceutically acceptable salt thereof described in claim 4 .

17 . A method for preventing and/or treating a disease associated with adenosine A 2A receptor comprising the step of administering an effective amount of a thiazole derivative represented by the general formula (I):

{wherein R 1 represents a substituted or unsubstituted 5-membered aromatic heterocyclic group containing at least one oxygen atom;

R 2 represents halogen, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, substituted or unsubstituted aromatic heterocyclic-alkyl, —NR 4 R 5 (wherein R 4 and R 5 may be the same or different, and each represents a hydrogen atom, substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted lower alkanoyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl), or —COR 6 (wherein R 6 represents substituted or unsubstituted lower alkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted aralkyl, a substituted or unsubstituted alicyclic heterocyclic group, a substituted or unsubstituted aromatic heterocyclic group, substituted or unsubstituted alicyclic heterocyclic-alkyl, or substituted or unsubstituted aromatic heterocyclic-alkyl);

or a pharmaceutically acceptable salt thereof.

18 . A method for antagonizing adenosine A 2A receptor comprising the step of administering an effective amount of the thiazole derivative or the pharmaceutically acceptable salt thereof described in claim 6 .

19 . A method for preventing and/or treating a disease associated with adenosine A 2A receptor comprising the step of administering an effective amount of the thiazole derivative or the pharmaceutically acceptable salt thereof described in claim 6 .

24 . The method for preventing and/or treating a disease associated with adenosine A 2A receptor according to claim 17 , wherein R 1 is substituted or unsubstituted furyl.
