Compositions and methods for binding sphingosine-1-phosphate
View Patent ↗The present invention relates to anti-S1P agents, for example, humanized monoclonal antibodies, and their uses for detection of S1P or for treatment of diseases and conditions associated with S1P.
1. An isolated antibody that binds sphingosine-1-phosphate (S1P) under physiological conditions and which comprises two light chain polypeptides and two heavy chain polypeptides, wherein:
a. each light chain polypeptide comprises first, second, and third complementarity determining regions CDRL1, CDRL2, and CDRL3, respectively, wherein:
(i) CDRL1 comprises amino acid sequence ITTTDIDDDMN (SEQ ID NO: 10);
(ii) CDRL2 comprises amino acid sequence EGNILRP (SEQ ID NO: 11); and
(iii) CDRL3 comprises amino acid sequence LQSDNLPFT (SEQ ID NO: 12); and
b. each heavy chain polypeptide comprises first, second, and third complementarity determining regions CDRH1, CDRH2, and CDRH3, respectively, wherein:
(i) CDRH1 comprises amino acid sequence DHTIH (SEQ ID NO: 13);
(ii) CDRH2 comprises amino acid sequence AISPRHDITKYNEMFRG (SEQ ID NO: 31); and
(iii) CDRH3 comprises amino acid sequence GGFYGSTIWFDF (SEQ ID NO: 15).
2. A kit comprising an antibody according to claim 1 packaged in a container, and optionally further comprising instructions for use.
3. An anti-S1P antibody according to claim 1 , wherein the antibody may be a chimeric antibody, a humanized antibody, a full-length antibody, an antigen-binding antibody fragment, and an affinity matured antibody.
4. An anti-S1P antibody according to claim 1 , wherein said anti-S1P antibody is conjugated to a second moiety selected from the group consisting of a polymer, a radionuclide, a chemotherapeutic agent, and a detection agent.
5. A composition comprising an anti-S1P antibody according to claim 1 and a carrier, optionally a pharmaceutically acceptable carrier.
6. An anti-S1P antibody according to claim 1 combined with a second agent which is optionally selected from the group consisting of an antibody, an antigen-binding antibody fragment, an antibody derivative, an antibody variant, a therapeutic agent other than an anti-S1P agent, or an agent comprising a binding moiety reactive with a molecule other than S1P, wherein such combination optionally occurs via a linkage, optionally a covalent linkage, between the anti-S1P antibody and the second agent.
