Abstract

An M.times.N matrix microfluidic device for performing a matrix of reactions, the device having a plurality of reaction cells in communication with one of either a sample inlet or a reagent inlet through a via formed within an elastomeric block of the device. Methods provided include a method for forming vias in parallel in an elastomeric layer of an elastomeric block of a microfluidic device, the method comprising using patterned photoresist masks and etching reagents to etch away regions or portions of an elastomeric layer of the elastomeric block.

Claims (13)

1. A microfluidic device comprising:

a plurality of reaction cells in a first layer of said device, wherein each reaction cell comprises

(i) a sample chamber and a reagent chamber, wherein said chambers are connected to each other through an interface channel; and

(ii) an interface valve in said interface channel between the sample chamber and the reagent chamber that, when actuated, fluidically isolates said sample chamber and said reagent chamber from each other;

a plurality of first reagent fluid channels in said first layer, wherein each first reagent fluid channel (i) terminates in a reagent chamber and (ii) is in fluid communication with a via that connects the first reagent channel and a second reagent fluid channel located in a different layer of the device, wherein each second reagent fluid channel is in fluid communication with a plurality of reagent chambers;

a plurality of sample fluid channels in said first layer, wherein each sample fluid channel is in fluidic communication with a plurality of sample chambers, and

wherein each reaction cell is in fluidic communication with a different first reagent fluid channel.

2. The device of claim 1 wherein said reaction cells are formed within an elastomeric block formed from a plurality of elastomeric components bonded together and said interface valve is a deflectable membrane.

3. The device of claim 1 wherein each reaction cell of said plurality of reaction cells further comprises

a first containment valve that, when actuated, fluidically isolates the sample chamber of the reaction cell from a first other sample chamber;

a second containment valve that, when actuated, fluidically isolates the reagent chamber of the reaction cell from a second reagent fluid channel.

The device of claim 3 wherein, for each reaction cell of said plurality of reaction cells, the first containment valve comprises a membrane deflectable into a sample fluid channel upon pressurization of a control channel and the second containment valve comprises a membrane deflectable into a reagent fluid channel upon pressurization of the same control channel.

The device of claim 4 wherein some of the reaction cells comprise a third containment valve that, when actuated, fluidically isolates said sample chamber from a second other sample chamber, wherein the third containment valve comprises a membrane deflectable into a sample fluid channel upon pressurization of said same control channel.
