Abstract

Methods are described for preventing or reducing ischemia and/or systemic inflammatory response in a patient such as perioperative blood loss and/or systemic inflammatory response in a patient subjected to cardiothoracic surgery, e.g. coronary artery bypass grafting and other surgical procedures, especially when such procedures involve extra-corporeal circulation, such as cardiopulmonary bypass.

Claims (16)

1. A pharmaceutical composition comprising a polypeptide comprising the amino acid sequence: Met His Ser Phe Cys Ala Phe Lys Ala Asp Asp Gly Pro Cys Arg Ala Ala His Pro Arg Trp Phe Phe Asn Ile Phe Thr Arg Gln Cys Glu Glu Phe Ile Tyr Gly Gly Cys Glu Gly Asn Gln Asn Arg Phe Glu Ser Leu Glu Glu Cys Lys Lys Met Cys Thr Arg Asp (amino acids 3-60 of SEQ ID NO:2), wherein the polypeptide inhibits plasma kallikrein,

and one or more pharmaceutically acceptable buffers, carriers and excipients.

The pharmaceutical composition of claim 1 , wherein the polypeptide comprises the amino acid sequence: Glu Ala Met His Ser Phe Cys Ala Phe Lys Ala Asp Asp Gly Pro Cys Arg Ala Ala His Pro Arg Trp Phe Phe Asn Ile Phe Thr Arg Gln Cys Glu Glu Phe Ile Tyr Gly Gly Cys Glu Gly Asn Gln Asn Arg Phe Glu Ser Leu Glu Glu Cys Lys Lys Met Cys Thr Arg Asp (SEQ ID NO:2).

The pharmaceutical composition of claim 2 , wherein the polypeptide consists of the amino acid sequence: Glu Ala Met His Ser Phe Cys Ala Phe Lys Ala Asp Asp Gly Pro Cys Arg Ala Ala His Pro Arg Trp Phe Phe Asn Ile Phe Thr Arg Gln Cys Glu Glu Phe Ile Tyr Gly Gly Cys Glu Gly Asn Gln Asn Arg Phe Glu Ser Leu Glu Glu Cys Lys Lys Met Cys Thr Arg Asp (SEQ ID NO:2).

The pharmaceutical composition of claim 1 , wherein the polypeptide consists of the amino acid sequence: Met His Ser Phe Cys Ala Phe Lys Ala Asp Asp Gly Pro Cys Arg Ala Ala His Pro Arg Trp Phe Phe Asn Ile Phe Thr Arg Gln Cys Glu Glu Phe Ile Tyr Gly Gly Cys Glu Gly Asn Gln Asn Arg Phe Glu Ser Leu Glu Glu Cys Lys Lys Met Cys Thr Arg Asp (amino acids 3-60 of SEQ ID NO:2).

5. The pharmaceutical composition of claim 1 , wherein the composition is a dry lyophilized powder.

6. A container comprising the pharmaceutical composition of claim 1 .

7. The container of claim 6 , wherein the container is an infusion bottle.

8. The container of claim 7 , wherein the infusion bottle comprises sterile pharmaceutical grade “water for injection”.

9. The container of claim 7 , wherein the infusion bottle comprises sterile saline.

10. The container of claim 6 , wherein the container is an ampoule.

11. The container of claim 10 , wherein the ampoule comprises sterile pharmaceutical grade “water for injection”.

12. The container of claim 10 , wherein the ampoule comprises sterile saline.

13. The container of claim 6 , wherein the container is a sachette.

14. The pharmaceutical composition of claim 1 , wherein the composition comprises buffered saline.

15. The pharmaceutical composition of claim 14 , wherein the composition comprises phosphate buffered saline.
