Abstract

A novel class or family of TGF-β binding proteins is disclosed. Also disclosed are assays for selecting molecules for increasing bone mineralization and methods for utilizing such molecules.

Claims (37)

1. An isolated polypeptide that retains the biological activity of SEQ ID NO: 2 in regulating bone mineral content, wherein said polypeptide comprises an amino acid sequence selected from the group consisting of:

(a) amino acid sequences having at least 85% sequence identity to SEQ ID NO: 2;

(b) amino acid sequences encoded by a polynucleotide sequence having at least 85% sequence identity to SEQ ID NO: 1;

(c) amino acids 25-213 of SEQ ID NO: 2;

(d) amino acid sequences having at least 85% sequence identity to amino acids 25-213 of SEQ ID NO: 2;

(e) amino acid sequences encoded by a polynucleotide sequence having at least 85% sequence identity to nucleotides 119-686 of SEQ ID NO: 1; and

(f) amino acid sequences encoded by a polynucleotide sequence that specifically hybridizes to the full complement of SEQ ID NO: 1, under conditions of high stringency comprising hybridization in 5×SSPE, 5×Denhardt's solution and 0.5% SDS overnight at 55 to 60° C.; followed by a post hybridization wash at 0.5×SSC at 55-60° C.

2. The isolated polypeptide according to claim 1 , which comprises an amino acid sequence having at least 85% sequence identity to SEQ ID NO: 2.

3. The isolated polypeptide according to claim 1 , which comprises an amino acid sequence having at least 85% sequence identity to amino acids 25-213 of SEQ ID NO: 2.

4. The isolated polypeptide according to claim 1 , which comprises amino acids 25-213 of SEQ ID NO: 2.

5. A method of detecting binding of a polypeptide according to claim 4 to an antibody, comprising the step of incubating an antibody with said polypeptide under conditions and for a time sufficient to permit said antibody to bind to said polypeptide, and detecting said binding.

The isolated polypeptide according to claim 1 , which comprises an amino acid sequence encoded by a polynucleotide sequence that specifically hybridizes to the full complement of SEQ ID NO: 1, under conditions of high stringency comprising hybridization in 5×SSPE, 5×Denhardt's solution and 0.5% SDS overnight at 55 to 60° C.; followed by a post hybridization wash at 0.5×SSC at 55-60° C.

7. A polypeptide according to claim 1 , further comprising a detectable label.

8. A method of detecting binding of a polypeptide according to claim 1 to an antibody, comprising the step of incubating an antibody with said polypeptide under conditions and for a time sufficient to permit said antibody to bind to said polypeptide, and detecting said binding.

9. A composition comprising a polypeptide according to claim 1 and a pharmaceutically acceptable carrier.

10. A fragment of SEQ ID NO: 2 consisting of at least 100 consecutive amino acid residues of SEQ ID NO: 2 and having the biological activity of SEQ ID NO: 2 in regulating bone mineral content.

11. The fragment according to claim 10 , further comprising a detectable label.

12. A method of detecting binding of a fragment of SEQ ID NO: 2 according to claim 10 to an antibody, comprising the step of incubating an antibody with said fragment under conditions and for a time sufficient to permit said antibody to bind to said polypeptide, and detecting said binding.

A polypeptide that retains the biological activity of SEQ ID NO: 2 in regulating bone mineral content produced by (a) culturing host cells under conditions and for a time sufficient to produce said polypeptide, and (b) isolating said polypeptide, wherein said host cells comprise an isolated nucleic acid molecule operably linked to a regulatory sequence that controls transcriptional expression, wherein said nucleic acid molecule (i) encodes said polypeptide and (ii) comprises a polynucleotide sequence selected from the group consisting of:

(1) SEQ ID NO: 1;

(2) polynucleotide sequences that specifically hybridize to the full complement of SEQ ID NO: 1, under conditions of high stringency comprising hybridization in 5×SSPE, 5×Denhardt's solution and 0.5% SDS overnight at 55 to 60° C.; followed by a post hybridization wash at 0.5×SSC at 55-60° C.;

(3) polynucleotide sequences having at least 85% sequence identity to the full complement of SEQ ID NO: 1;

(4) polynucleotide sequences that encode SEQ ID NO: 2;

(5) polynucleotide sequences that encode a polypeptide having at least 85% sequence identity to SEQ ID NO: 2;

(6) nucleotides 119-686 of SEQ ID NO: 1;

(7) polynucleotide sequences having at least 85% sequence identity to nucleotides 119-686 of SEQ ID NO: 1;

(8) polynucleotide sequences that encode amino acids 25-213 of SEQ ID NO: 2; and

(9) polynucleotide sequences that encode a polypeptide having at least 85% sequence identity to amino acids 25-213 of SEQ ID NO: 2.

14. The polypeptide of claim 13 , wherein said polypeptide is encoded by a polynucleotide comprising SEQ ID NO: 1.

15. The polypeptide of claim 13 , wherein said polypeptide is encoded by a polynucleotide having at least 85% sequence identity to SEQ ID NO: 1.

The polypeptide of claim 13 , wherein the polypeptide is encoded by a polynucleotide sequence that specifically hybridize to the full complement of SEQ ID NO: 1, under conditions of high stringency comprising hybridization in 5×SSPE, 5×Denhardt's solution and 0.5% SDS overnight at 55 to 60° C.; followed by a post hybridization wash at 0.5×SSC at 55-60° C.

17. The polypeptide of claim 13 , wherein the polypeptide is encoded by a polynucleotide sequence having at least 85% sequence identity to nucleotides 119-686 of SEQ ID NO: 1.

18. The polypeptide of claim 13 , wherein the polypeptide is encoded by a polynucleotide sequence comprising nucleotides 119-686 of SEQ ID NO: 1.

19. The polypeptide of claim 13 , wherein the polypeptide is encoded by a polynucleotide sequence that encodes a polypeptide having at least 85% sequence identity to SEQ ID NO: 2.

20. The polypeptide of claim 13 , wherein the polypeptide is encoded by a polynucleotide sequence that encodes polypeptide having at least 85% sequence identity to amino acids 25-213 of SEQ ID NO: 2.

A polypeptide that retains the biological activity of SEQ ID NO: 2 in regulating bone mineral content produced by (a) culturing host cells under conditions and for a time sufficient to produce said polypeptide, and (b) isolating said polypeptide, wherein said host cells comprise an isolated nucleic acid molecule operably linked to a regulatory sequence that controls transcriptional expression, and wherein said nucleic acid molecule comprises a polynucleotide sequence that encodes amino acids 25-213 of SEQ ID NO: 2.

A polypeptide that retains the biological activity of SEQ ID NO: 2 in regulating bone mineral content produced by (a) culturing host cells under conditions and for a time sufficient to produce said polypeptide, and (b) isolating said polypeptide, wherein said host cells comprise a nucleic acid molecule operably linked to a heterologous regulatory sequence that controls transcriptional expression, and wherein said nucleic acid molecule comprises a polynucleotide sequence that encodes amino acids 25-213 of SEQ ID NO: 2.
