Abstract

Identified herein is the location of the MN protein binding site, and MN proteins/polypeptides that compete for attachment to vertebrate cells with immobilized MN protein. Such MN proteins/polypeptides prevent cell-cell adhesion and the formation of intercellular contacts. The MN protein binding site is a therapeutic target that can be blocked by organic or inorganic molecules, preferably organic molecules, more preferably proteins/polypeptides that specifically bind to that site. Therapeutic methods for inhibiting the growth of preneoplastic/neoplastic vertebrate cells that abnormally express MN protein are disclosed. Vectors are provided that encode the variable domains of MN-specific antibodies and a flexible linker polypeptide separating those domains. Further vectors are disclosed that encode a cytotoxic protein/polypeptide operatively linked to the MN gene promoter or a MN gene promoter fragment comprising the HIF-1 consensus binding sequence, and which vectors preferably further encode a cytokine. The MN gene promoter is characterized, and the binding site for a repressor of MN transcription is disclosed. Further, the hypoxia inducibility of the MN gene and the uses of such inducibility are disclosed.

Claims (27)

1. A method for determining the percentage of the cells in a vertebrate tumor that are hypoxic, comprising:

a) administering to said vertebrate tumor an expression vector comprising a nucleic acid encoding a reporter gene operatively linked to a hypoxia-inducible MN/CA9 promoter fragment; and

b) detecting and quantifying reporter gene expression product expressed by said vector in cells of said tumor or in a tissue sample taken from said tumor; and

c) determining the percentage of hypoxic cells in said tumor based on the percentage of cells expressing said reporter gene expression product;

wherein said hypoxia-inducible MN/CA9 promoter fragment comprises a nucleotide sequence selected from the group consisting of:

(1) SEQ ID NO: 146 and its complement; and

(2) nucleotide sequences that are 90% homologous to the nucleotide sequences of (1); and

wherein said hypoxia-inducible MN/CA9 promoter fragment has hypoxia-inducible activity.

2. A method for determining the relative degree of hypoxia in a tissue in a vertebrate, comprising:

a) administering to said vertebrate tissue an expression vector comprising a nucleic acid encoding a reporter gene operatively linked to a hypoxia-inducible MN/CA9 promoter fragment;

b) detecting MN/CA9 reporter gene expression product in said tissue; and

c) quantifying said reporter gene expression product, wherein the relative level of reporter gene expression product indicates the relative degree of hypoxia;

wherein said hypoxia-inducible MN/CA9 promoter fragment comprises a nucleotide sequence selected from the group consisting of:

(1) SEQ ID NO: 146 and its complement; and

(2) nucleotide sequences that are 90% homologous to the nucleotide sequences of (1); and

wherein said hypoxia-inducible MN/CA9 promoter fragment has hypoxia-inducible activity.

3. The method of claim 2 wherein said hypoxia is chronic hypoxia.

4. The method of claim 2 wherein a higher degree of hypoxia in said tissue is considered indicative of a poorer prognosis of a preneoplastic/neoplastic disease.

5. The method of claim 4 , wherein a poorer prognosis is measured in terms of shortened survival or increased risk of recurrence of said preneoplastic/neoplastic disease.

6. The method of claim 5 wherein said method is used as an aid in making a therapeutic decision in the treatment of said preneoplastic/neoplastic disease afflicting said vertebrate.

7. The method of claim 6 wherein said method comprises predicting patient response to a specific therapy or specific therapies.

8. The method of claim 6 wherein said therapeutic decision comprises a decision to treat said disease with a hypoxia-modifying therapy if hypoxia is present in high levels.

9. The method of claim 6 wherein said therapeutic decision comprises a decision to treat said disease with bio-reductive drugs if hypoxia is present in high levels.

10. The method of claim 6 wherein said therapeutic decision comprises a decision to treat said disease with radiation and/or chemotherapy if hypoxia is absent or present in low levels.

11. The method of claim 6 , wherein said therapeutic decision comprises a decision to treat said disease with a combination of radiotherapy and hypoxia-directed therapy if hypoxia is present in high levels.

12. The method of claim 6 wherein said therapeutic decision comprises a decision to treat said disease more aggressively if hypoxia is present in high levels in said tissue, or less aggressively if hypoxia is absent or present in low levels in said tissue.

The method of claim 4 , wherein said disease is selected from the group consisting of preneoplastic/neoplastic diseases related to head and neck cancers, head and neck squamous cell carcinoma, uterine carcinoma, uterine cervical carcinoma, renal cancer, breast cancer, lung cancer, pancreatic cancer, soft tissue sarcoma, bladder cancer, colorectal cancer, esophageal cancer, prostate cancer, ovarian cancer, endometrial cancer, squamous cell and adenosquamous carcinomas, mesodermal tumors, neuroblastomas, retinoblastomas, sarcomas, osteosarcomas, Ewing's sarcoma, melanomas, vaginal cancer, vulval cancer, gastrointestinal cancer, urinary tract cancer, kidney cancer, non-small cell lung cancer, skin cancer, liver cancer, cervical squamous cell carcinoma, cervical adenosquamous carcinoma, cervical adenocarcinoma, cervical metaplasia, and condyloma.
